Humane Endpoint Implementation in Animal Research
Humane endpoints are a critical component of the Refinement principle (3Rs). Proper implementation requires:
Prospective criteria: All endpoint criteria must be defined BEFORE the study begins and documented in the IACUC protocol. Ad hoc endpoints are not acceptable.
Objective scoring: Use numerical scoring systems (0–3 per criterion) rather than subjective assessments. This enables consistent scoring across personnel and shifts. Body Condition Score (Ullman-Cullere & Foltz 1999) is the gold standard for nutritional assessment.
Cumulative scoring: Individual criteria may not reach critical levels, but the combination of mild symptoms across multiple parameters indicates significant distress. Cumulative thresholds catch animals that would be missed by single-criterion endpoints.
Automatic endpoints: Some conditions require immediate euthanasia regardless of cumulative score: >= 20% body weight loss, inability to reach food/water, moribund state, tumor ulceration with necrosis, or respiratory distress.
Documentation: Every monitoring observation must be recorded with date, time, animal ID, individual scores, and cumulative score. This creates the audit trail required by IACUC and regulatory agencies.
