Data for: An AAV-CRISPR/Cas9-strategy for gene editing across divergent rodent species: Targeting neural oxytocin receptors as a proof of concept
<p>A major issue in neuroscience is the poor translatability of research results from preclinical studies in animals to clinical outcomes. Comparative neuroscience can overcome this barrier by studying multiple species to differentiate between species-specific and general mechanisms of neural circuit functioning. Targeted manipulation of neural circuits often depends on genetic dissection, and use of this technique has been restricted to only a few model species, limiting its application in comparative research. However, ongoing advances in genomics make genetic dissection attainable in a growing number of species. To demonstrate the potential of comparative gene editing approaches, we developed a viral-mediated CRISPR/Cas9 strategy that is predicted to target the oxytocin receptor (<em>Oxtr</em>) gene in >80 rodent species. This strategy specifically reduced OXTR levels in all evaluated species (n=6), without causing gross neuronal toxicity. Thus, we show that CRISPR/Cas9-based tools can function in multiple species simultaneously. Thereby, we hope to encourage comparative gene editing and improve the translatability of neuroscientific research.</p>

<p>Various “open” concepts in Computing, such as open standards, open data, open licenses or system interoperability are becoming more important in the professional lives of software engineers. However, students usually do not receive a systematic education about these concepts ; rather they sporadically learn about a subset of these topics. This paper presents the revised version of the Open Computing course in the University of Zagreb, Faculty of Electrical Engineering and Computing (FER), which teaches a clear set of current topics focused on open data and correlating concepts. The new e-learning course is carried out using the “flipped classroom” educational method ; students construct their knowledge in a set of real-world mini-activities throughout the course, instead of passively learning from the official course resources. In this paper, we discuss our flipped classroom activities, their relation to revised Bloom’s taxonomy of educational objectives, and present the evaluation of the first course instance us- ing this model. Students’ feedback shows they welcome this change in approach, finding the course useful and interesting, and preferring this method to the traditional full lecture setting.</p>
Data from: Dynamic balancing of risks and rewards in a large herbivore: Further extending predator-prey concepts to road ecology
<p>Animal behavior is shaped by the ability to identify risks and profitably balance the levels of risks encountered with the payoffs experienced. Anthropogenic disturbances like roads generate novel risks and opportunities that wildlife must accurately perceive and respond to. Basic concepts in predator-prey ecology are often used to understand responses of animals to roads (e.g., increased vigilance, selection for cover in their vicinity). However, prey often display complex behaviors such as modulating space use given varying risks and rewards, and it is unclear if such dynamic balancing is used by animals in the context of road crossings.</p> <p>We tested whether animals dynamically balance risks and rewards relative to roads using extensive field -based and GPS collar data from elk in Yoho National Park (British Columbia, Canada) where a major highway completely bisects their range during most of the year.</p> <p>We analyzed elk behavior by combining hidden Markov movement models with a step-selection function framework. Rewards were indexed by a dynamic map of available forage biomass and risks were indexed by road crossings and traffic volumes.</p> <p>We found that elk generally selected intermediate and high forage biomass and avoided crossing the road. Most of the time, elk modulated their behavior given varying risks and rewards. When crossing the highway compared with not crossing, elk selected for greater forage biomass and this selection was stronger as the number of highway crossings increased. However, with traffic volume, elk only balanced foraging rewards when they crossed a single time during a travel sequence.</p> <p>Using a road ecology system, we empirically tested an important component of predator-prey ecology – the ability to dynamically modulate behavior in response to varying levels of risks and rewards. Such a test articulates how decision-making processes that consider the spatiotemporal variation in risks and rewards allow animals to successfully and profitably navigate busy roads. Applying well-developed concepts in predator-prey theory helps understand how animals respond to anthropogenic disturbances and anticipate the adaptive capacity for individuals and populations to adjust to rapidly changing environments.</p>

Lithium recovery from brines by lithium membrane flow capacitive deionization (Li-MFCDI) – A proof of concept

Saif et al., <a href="https://www.sciencedirect.com/journal/journal-of-membrane-science-letters">Journal of Membrane Science Letters</a>, <a href="https://www.sciencedirect.com/journal/journal-of-membrane-science-letters/vol/3/issue/2">Volume 3, Issue 2</a>, November 2023, 100059</p> <p>The demand of lithium for electric vehicles and energy storage devices is increasing rapidly, thus new sources of<br> lithium (such as seawater and natural or industrial brines), as well as sustainable methods for its recovery, will<br> need to be explored/developed soon. This work presents a novel electromembrane process, called Lithium<br> Membrane Flow Capacitive Deionization (Li-MFCDI), which was tested to recover lithium from a synthetic<br> geothermal brine containing a much higher mass concentration of sodium than lithium (more than 650 times).<br> Specifically, a ceramic lithium-selective membrane was integrated into a flow capacitive deionization (FCDI)<br> cell, which was specifically designed, and 3D printed, to allow simultaneous charging and regeneration of the<br> employed flow electrodes. Despite the extremely high Na+/Li+ mass ratio in the feed stream, 99.98% of the<br> sodium was rejected and the process selectivity for lithium over other monovalent cations was 141 ± 5.85 for<br> Li+/Na+ and 46 ± 1.46 for Li+/K+. The Li-MFCDI process exhibited a stable behaviour over a 7-day test period,<br> and the estimated energy consumption was 16.70 ± 1.63 kWh/kg of Li+ recovered in the draw solution. These<br> results demonstrate promising potential of the Li-MFCDI for the sustainable lithium recovery from saline streams.</p>

Diagnostic of <em>Plasmodium</em> species is important for administering the appropriate treatment. The gold-standard diagnosis from accurate species identification remains the thin blood smear. Nevertheless, this method is time-consuming and requires highly skilled and trained microscopists. To overcome these issues, new diagnostic tools based on deep learning are emerging. This study aimed to evaluate the performances of a RT-DETR (Real-Time Detection Transformer)object detection algorithm to discriminate <em>Plasmodium</em> species on thin blood smears images. The algorithm was trained and validated on a dataset consisting in 24,720 images from 475 thin blood smears corresponding to 2,002,597 labels. Performances were calculated with a test dataset of 4,508 images from 170 smears corresponding to 358,825labels coming from six French university hospital. At the patient level, the RT-DETR algorithm exhibited an overall accuracy of 79.4% (135/170) with a recall of 74% (40/54) and 81.9% (95/116) for negative and positive smears, respectively. Among <em>Plasmodium </em>positive smears, the global sensitivity was 82.7% (91/110) with a sensitivity of 90% (38/42), 81.8% (18/22) and 76.1% (35/46) for <em>P. falciparum</em>, <em>P. malariae </em>and <em>P. ovale/vivax,</em> respectively. The YOLOv5 model achieved a World Health Organization (WHO) competence level 2 for species identification. Besides, the RT-DETR algorithm may be run in real-time on low-cost devices such as a smartphone and could be suitable for deployment in low-resource setting areas where microscopy experts are lacking.</p> <p><strong>Data collection:</strong></p> <p>The training and validation dataset included 24,720 pictures taken from 475 manually May Grunwald-Giemsa (MGG)-stained thin blood smears from the Montpellier University Hospital collection and for a smaller part from the Toulouse University Hospital collection. In Montpellier, the pictures were taken with a Flexcam C1 microscope camera (Leica) attached to a Leica DM 2000 microscope and Leica DF450C microscope camera adapted with a Leica DM2500 microscope at X1000 magnification. Labelling of pictures was performed manually, and then automatically with manual correction with a Computer Visual Annotation Tools (CVAT) free software. Nine categories of labels were used: white blood cells (n=3,338), red blood cells (n=1,887,781), platelets (n=48,520), <em>Trypanosoma brucei </em>(n=2,773), and red blood cells infected by <em>P. falciparum </em>(n=43,545), <em>P. ovale </em>(n=4,651), <em>P. vivax </em>(n=4,115), <em>P. malariae </em>(n=2,849) and <em>Babesia divergens</em> (n=5,142).</p> <p>The test dataset included 4,508 pictures taken from 170 thin blood smears from the same number of patients from the Parasitology laboratories of University Hospitals of Montpellier, Toulouse, Rouen, Lille, Nantes and Saint-Louis in Paris (Table 1). Among these 170 patients, 54 were not infected, including two patients with Howell-Jolly bodies, and 116 were infected with hematozoa. For each patient, between 20 and 30 photos were taken from one thin blood smear with at least one hematozoan parasite per picture for infected patients.</p> <p>Accurate species diagnostic was made by a senior parasitologist, and for recent smears, it was confirmed by specific PCR, either performed locally (Toulouse) or at the Malaria French National Reference Center (Montpellier, Saint Louis, Rouen, Lille, Nantes).</p>
