Data from: Is high-sensitivity troponin, alone or in combination with copeptin, sensitive enough for ruling out NSTEMI in very early presenters at admission? A post-hoc analysis performed in emergency departments

Objectives: Copeptin and high-sensitivity cardiac troponin (HS-cTn) assays improve the early detection of NSTEMI. Their sensitivities may however be reduced in very early presenters. Setting: We performed a post-hoc analysis of three prospective studies that included patients who presented to the Emergency Department for chest pain onset (CPO) of less than 6 hours. Participants: 449 patients were included, in whom 12% had NSTEMI. CPO occurred <2h from ED presentation in 160, between 2-4h in 143, and >4h in 146 patients. The prevalence of NSTEMI was similar in all groups (9%, 13% and 12%, respectively, p=0.281). Measures: Diagnostic performances of HS-cTn and copeptin at presentation were examined according to CPO. The discharge diagnosis was adjudicated by 2 experts, including cTnI. HS-cTn and copeptin were blindly measured. Results: Diagnostic accuracies of cTnI, cTnI+copeptin and HS-cTnT (but not HS-cTnT+copeptin) lower through CPO categories. For patients with CPO<2h, the choice of a threshold value of 14 ng/L for HS-cTnT resulted in 3 false negative (Sensitivity 80% [95%CI: 51-95]; specificity 85% [78-90]; 79% of correctly ruled-out patients) and that of 5 ng/L in 2 false negative ( Sensitivity 87% [59-98]; specificity 58% [50-66]; 52% of correctly ruled-out patients). The addition of copeptin to HS-cTnT induced a decrease of misclassified patients to 1 in patients with CPO<2h (Sensitivity 93% [66-100]; specificity 41% [33-50]). Conclusion: A single measurement of HS-cTn, alone or in combination with copeptin at admission, seems not safe enough for ruling out NSTEMI in very early presenters (with CPO<2h). Data from: Evolutionary rescue and local adaptation under different rates of temperature increase: a combined analysis of changes in phenotype expression and genotype frequency in Paramecium microcosms

Evolutionary Rescue (ER) occurs when populations, which have declined due to rapid environmental change, recover through genetic adaptation. The success of this process and the evolutionary trajectory of the population strongly depend on the rate of environmental change. Here we investigated how different rates of temperature increase (from 23°C to 32°C) affect population persistence and evolutionary change in experimental microcosms of the protozoan Paramecium caudatum. Consistent with theory on ER, we found that those populations experiencing the slowest rate of temperature increase were the least likely to become extinct and tended to be the best adapted to the new temperature environment. All high-temperature populations were more tolerant to severe heat stress (35°C, 37°C), indicating a common mechanism of heat protection. High-temperature populations also had superior growth rates at optimum temperatures, leading to the absence of a pattern of local adaptation to control (23°C) and high-temperature (32°C) environments. However, high-temperature populations had reduced growth at low temperatures (5-9°C), causing a shift in the temperature niche. In part, the observed evolutionary change can be explained by selection from standing variation. Using mitochondrial markers, we found complete divergence between control and high-temperature populations in the frequencies of six initial founder genotypes. Our results confirm basic predictions of ER and illustrate how adaptation to an extreme local environment can produce positive as well as negative correlated responses to selection over the entire range of the ecological niche.

Data from: Bayesian phylogenetic analysis of combined data

The recent development of Bayesian phylogenetic inference using Markov chain Monte Carlo (MCMC) techniques has facilitated the exploration of parameter-rich evolutionary models. At the same time, stochastic models have become more realistic (and complex) and have been extended to new types of data, such as morphology. Based on this foundation, we developed a Bayesian MCMC approach to the analysis of combined data sets and explored its utility in inferring relationships among gall wasps based on data from morphology and four genes (nuclear and mitochondrial, ribosomal and protein coding). Examined models range in complexity from those recognizing only a morphological and a molecular partition to those having complex substitution models with independent parameters for each gene. Bayesian MCMC analysis deals efficiently with complex models: convergence occurs faster and more predictably for complex models, mixing is adequate for all parameters even under very complex models, and the parameter update cycle is virtually unaffected by model partitioning across sites. Morphology contributed only 5% of the characters in the data set but nevertheless influenced the combined-data tree, supporting the utility of morphological data in multigene analyses. We used Bayesian criteria (Bayes factors) to show that process heterogeneity across data partitions is a significant model component, although not as important as among-site rate variation. More complex evolutionary models are associated with more topological uncertainty and less conflict between morphology and molecules. Bayes factors sometimes favor simpler models over considerably more parameter-rich models, but the best model overall is also the most complex and Bayes factors do not support exclusion of apparently weak parameters from this model. Thus, Bayes factors appear to be useful for selecting among complex models, but it is still unclear whether their use strikes a reasonable balance between model complexity and error in parameter estimates.

Data from: A national VS30 model for South Korea to combine nationwide dense borehole measurements with ambient seismic noise analysis

<p>The average shear-wave velocity within the top 30 m from the surface, V<sub>S30</sub>, represents site characteristics including the soil classification and site amplification that are essential information for building codes and seismic design. A novel method to determine a V<sub>S30</sub> model based on a composite analysis of borehole standard penetration test numbers (SPT N) and horizontal-to-vertical (H/V) spectral ambient noise ratios is introduced. A national V<sub>S30</sub> model for South Korea is determined using the method. The shear-wave velocity structures beneath 20 nationwide broadband seismic stations are determined using the H/V analysis. The SPT N data are collected from 175,619 nationwide densely-distributed boreholes. The shear-wave velocity models from SPT N values are calibrated for the local reference velocity models from H/V analysis. A representative relationship between the SPT N values and shear-wave velocities is introduced. A national V<sub>S30</sub> model for South Korea is determined using the calibrated SPT N models at the nationwide boreholes. The V<sub>S30</sub> model is verified by comparisons with local field measurements. The proposed model is consistent with the USGS model based on a surface slope analysis. The V<sub>S30</sub> structure presents high correlation with geological and topographic features. The V<sub>S30</sub> values are low in coastal (low topographic) areas, and high in mountain (high topographic) areas. Apparent linear relationship is observed between V<sub>S30</sub> and topography. The western and southeastern coastal regions may be vulnerable to strong seismic shaking.</p>

Data from: Combined analysis of variation in core, accessory and regulatory genome regions provides a super-resolution view into the evolution of bacterial populations

The use of whole-genome phylogenetic analysis has revolutionized our understanding of the evolution and spread of many important bacterial pathogens due to the high resolution view it provides. However, the majority of such analyses do not consider the potential role of accessory genes when inferring evolutionary trajectories. Moreover, the recently discovered importance of the switching of gene regulatory elements suggests that an exhaustive analysis, combining information from core and accessory genes with regulatory elements could provide unparalleled detail of the evolution of a bacterial population. Here we demonstrate this principle by applying it to a worldwide multi-host sample of the important pathogenic E. coli lineage ST131. Our approach reveals the existence of multiple circulating subtypes of the major drug–resistant clade of ST131 and provides the first ever population level evidence of core genome substitutions in gene regulatory regions associated with the acquisition and maintenance of different accessory genome elements.
