Multi-omics analysis of innate and adaptive responses to BCG vaccination reveals epigenetic cell states that predict trained immunity
<p>This repository contains personal immune profiles of 323 healthy individuals (300BCG) subjected to Bacillus Calmette-Guérin (BCG) with blood samples collected immediately before (day 0), and 14 and 90 days after the vaccination. Uptake of Human Papilloma Virus vaccine among young women living in fishing communities in Wakiso and Mukono districts, Uganda
<p>Human Papilloma Virus (HPV) is a preventable cause of cervical cancer, the most common cancer among women in Uganda. The Uganda Ministry of Health included the HPV vaccine in the free routine immunization schedule since 2015. Five years after this policy, we assessed the uptake of the HPV vaccine and associated socio-demographic factors among young women living in fishing communities in Central Uganda in 2020.</p> <p>We analyzed secondary data from 94 young women aged 9–25 years who were recruited from the two fishing communities (Kasenyi landing site and Koome Island) in a primary implementation study that aimed to promote awareness of maternal and childhood vaccines. We assessed uptake of the HPV vaccine as the proportion of participants who self-reported to have ever received at least one dose of the HPV vaccine. We assessed the socio-demographic factors associated with HPV vaccine uptake using a modified Poisson regression model adjusted for clustering by study site in STATA version 17.</p> <p>The mean (standard deviation) age of study participants was 21.1 (3.1) years and most (81.9%) of them were from Kasenyi landing site. The uptake of the HPV vaccine was 10.6% [95% Confidence Interval (CI) 5.6, 18.9]. After adjusting for covariates, being 20–25 years old (adjusted prevalence ratio [aPR] 0.18, 95%CI 0.06, 0.59) and of Catholic religion (aPR 5.55, 95%CI 1.53, 20.16) were significantly associated with HPV vaccine uptake. The HPV vaccine uptake was very low, despite the reported 99% national coverage of HPV vaccination program for the first dose at the end of 2019. Age and religion were shown to be important determinants of the HPV vaccine uptake. Reasons for very low uptake of HPV vaccinations need to be carefully assessed to find effective strategies to improve it.</p>
COVID-19 vaccine uptake data set
<p>The novel Coronavirus Disease 19 (COVID-19) caused devastating effects globally, and healthcare workers were among the most affected by the pandemic. Despite healthcare workers being prioritized in COVID-19 vaccination globally and in Ghana, hesitancy to receive the vaccines resulted in delayed control of the pandemic. In Ghana, healthcare workers had a vaccine acceptance of 39.3% before the vaccine rollout. Consequently, this study assessed the uptake of COVID-19 vaccination and associated factors among healthcare workers in Ghana in the post-vaccine roll-out period. This was an analytical cross-sectional study that used a semi-structured questionnaire to collect data on COVID-19 vaccination uptake and influencing factors. 256 healthcare workers were selected in Ayawaso West Municipality of Ghana using a stratified random sampling approach. Descriptive statistics were used to examine socio-demographic factors and Likert scale responses. Bivariable and Multivariable logistic regression were performed using IBM SPSS version 22 to identify predictors of vaccine uptake and a statistical significance was declared at p<0.05. More than three-fourths of participants 220 (85.9%) had received at least one dose of the COVID-19 vaccination, while 36 (14.9%) were hesitant. More than half 139 (54.3%) had adequate knowledge about COVID-19 vaccination and the majority 188 (73.4%) had positive perceptions about its effectiveness. Moreover, 218 (85.2%) of HCWs had a positive attitude towards COVID-19 vaccination. Positive attitude towards COVID-19 vaccination (AOR = 4.3; 95% CI: 1.4, 13.0) and high cues to action (AOR = 5.7; 95% CI: 2.2, 14.8) were the factors that significantly predicted uptake of COVID-19 vaccination among healthcare workers. COVID-19 vaccination among HCWs in Ghana is promising. However, hesitancy to receive the vaccination among a significant proportion of HCWs raises concerns. To ensure vaccination of all healthcare workers, interventions to promote vaccination should target key determinants of vaccination uptake, such as attitude towards the vaccination and cues to action.</p>

<p>SARS‐CoV‐2 vaccine is considered the primary health strategy able to end the current COVID‐19 pandemic. This viral infection impacts more severely solid organ transplant recipients (SOTRs) than general population, but the effect of vaccination in this subgroup of immunosuppressed patients is not known due to their exclusion from vaccination trials. Preliminary reports suggest a lower antibody production after BNT162b2 Pfizer/BioNTech mRNA‐vaccine,<a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8222937/#ajt16702-bib-0001"> 1 </a>, <a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8222937/#ajt16702-bib-0002">2 </a>, <a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8222937/#ajt16702-bib-0003">3 </a>, <a href="https://www.ncbi.nlm.nih.gov/pmc/articles/PMC8222937/#ajt16702-bib-0004">4 </a>but no data are currently available on the elicited virus‐specific T cell responses.</p>
Protective effect conferred by prior infection and vaccination on COVID-19 in a healthcare worker cohort in South India
<p><span>The emergence of newer variants with the immune escape potential raises concerns about breakthroughs and re-infections resulting in future waves of infection. We examined the protective effect of prior COVID-19 disease and vaccination on infection rates among a cohort of healthcare workers (HCW) in South India during the second wave driven mainly by the delta variant. </span><span>Symptomatic HCWs were routinely tested by RT-PCR as per institutional policy. Vaccination was offered to all HCWs in late January, and the details were documented. We set up a non-concurrent cohort to document infection rates and estimated protective efficacy of prior infection and vaccination between 16th Apr to 31st May 2021, using a Cox proportional hazards model with time-varying covariates adjusting for daily incidence.</span></p> <p><span>Between June 2020 and May 2021, 2735 (23.9%) of 11,405 HCWs were infected, with 1412, including 32 re-infections, reported during the second wave. 6863 HCWs received two doses of vaccine and 1905 one dose. The protective efficacy of prior infection against symptomatic infection was 86.0% (95% CI 76.7% - 91.6%). Vaccination combined with prior infection provided 91.1% (95% CI 84.1% - 94.9%) efficacy. In the absence of prior infection, vaccine efficacy against symptomatic infection during the second wave was 31.8% (95% CI 23.5% – 39.1%).</span></p> <p><span>Prior infection provided substantial protection against symptomatic re-infection and severe disease during a delta variant-driven second wave in a cohort of health care workers.</span></p>

Misconceptions about COVID-19 vaccine among adults in Saudi Arabia and their associated factor
<p>It is of the utmost importance for the elements that influence public compliance to the COVID-19 vaccine to be assessed, including misconceptions, rumors, and conspiracy theories. Hence our study aimed to estimate the distribution of the most common misconceptions and their predictors in Saudi Arabia. We distributed an online questionnaire to participants who were 18 years or older at the time of the survey, which included four sections: demographic characteristics of participants, level of their education, their sources of information, and perception of participants regarding eleven of the most common misconceptions in the community.</p> <p>An evaluation of the relationships between different predictors and acceptance of these misconceptions was conducted using ordinal logistic regression; the most widely accepted misconception was that the vaccine has serious side effects, with 34.8% of participants reporting this.</p> <p><strong>Accepting this misinformation</strong> Factors that were significantly associated with acceptance or refusal of misconceptions were:</p> <p><span>Gender</span>, as females in this survey significantly accept rumors more than males (p<0.001);</p> <p><span>Educational level</span>, especially secondary school, was associated with a significant decrease in acceptance of misconceptions (p=0.001);</p> <p><span>Information source</span>. 60.5% of participants use social media as their primary source of information, and it was also a significant positive predictor of acceptance of misconceptions (p=0.034).</p> <p>It is of utmost importance to increase assurance in the safety of the vaccine, as it was the most accepted misconception, and to address the elements that affect believing rumors among the population.</p>

Twitter vaccine misinformation data
<p>Anti-vaccine content is rapidly propagated via social media, fostering vaccine hesitancy, while pro-vaccine content has not replicated the opponent's successes. Despite this disparity in the dissemination of anti- and pro-vaccine posts, linguistic features that facilitate or inhibit the propagation of vaccine-related content remain less known. Moreover, most prior machine-learning algorithms classified social-media posts into binary categories (e.g., misinformation or not) and have rarely tackled a higher-order classification task based on divergent perspectives about vaccines (e.g., anti-vaccine, pro-vaccine, and neutral). Our objectives are (1) to identify sets of linguistic features that facilitate and inhibit the propagation of vaccine-related content and (2) to compare whether anti-vaccine, pro-vaccine, and neutral tweets contain either set more frequently than the others. To achieve these goals, we collected a large set of social media posts (over 120 million tweets) between Nov. 15 and Dec. 15, 2021, coinciding with the Omicron variant surge. A two-stage framework was developed using a fine-tuned BERT classifier, demonstrating over 99 and 80 percent accuracy for binary and ternary classification. Finally, the Linguistic Inquiry Word Count text analysis tool was used to count linguistic features in each classified tweet. Our regression results show that anti-vaccine tweets are propagated (i.e., retweeted), while pro-vaccine tweets garner passive endorsements (i.e., favorited). Our results also yielded the two sets of linguistic features as facilitators and inhibitors of the propagation of vaccine-related tweets. Finally, our regression results show that anti-vaccine tweets tend to use the facilitators, while pro-vaccine counterparts employ the inhibitors. These findings and algorithms from this study will aid public health officials' efforts to counteract vaccine misinformation, thereby facilitating the delivery of preventive measures during pandemics and epidemics.</p>
The metabolic hormone adiponectin affects the correlation between nutritional status and pneumococcal vaccine response in vulnerable indigenous children
<p class="MsoNoSpacing"><strong><span>Background:</span></strong><span> Almost 200 million children worldwide are either undernourished or overweight</span><span>. </span><span>Only a few studies have addressed the effect of variation in nutritional status on vaccine response</span><span>. </span><span>We previously demonstrated an association between stunting and an increased post-vaccination 13-valent pneumococcal conjugate vaccine (PCV13) response. In this prospective study, we assessed to what extent metabolic hormones may be a modifier in the association between nutritional status and PCV13 response.</span></p> <p class="MsoNoSpacing"><strong><span>Methods: </span></strong><span>Venezuelan children aged 6 weeks to 59 months were vaccinated with a primary series of PCV13. Nutritional status and serum levels of leptin, adiponectin and ghrelin were measured upon vaccination and their combined effect on serum post-vaccination antibody concentrations was assessed by generalized estimating equations multivariable regression analysis.</span></p> <p class="MsoNoSpacing"><strong><span>Results:</span></strong><span> A total of 210 children were included, of whom 80 were stunted, 81 had a normal weight and 49 were overweight. Overweight children had lower post-vaccination antibody concentrations than normal weight children </span><span>(regression coefficient -1.15, 95% CI -2.22 – -0.072)</span><span>. Additionally, there was a significant adiponectin-nutritional status interaction. In stunted children, higher adiponectin serum concentrations were associated with lower post-PCV13 antibody concentrations </span><span>(regression coefficient -0.19, 95% CI -0.24 – -0.14) </span><span>while the opposite was seen in overweight children </span><span>(regression coefficient 0.14, 95% CI 0.049 – 0.22)</span><span>. </span></p> <p><strong><span>Conclusion:</span></strong><span> Metabolic hormones, in particular adiponectin, may modify the effect of nutritional status on pneumococcal vaccine response. These findings emphasize the importance of further research to better understand the immunometabolic pathways underlying vaccine response and enable a future of optimal personalized vaccination schedules.</span></p>

Assessing Vaccine-Related Content for Journalistic Quality: A large-scale dataset and article repository
<p>This dataset was produced through a collaboration with the NSF-funded <a href="https://artt.cs.washington.edu/">ARTT project</a> (led by Hacks/Hackers) and <a href="https://overtone.ai/">Overtone</a>. It consists of 1,000 vaccine-related articles, pulled from a wide variety of news media sources, with associated scores based on their journalistic quality. The scores were provided through Overtone’s algorithm, and range from one (low-quality or low informational value add) to five (high-quality or high informational value add). Articles were sourced from traditional journalism outlets (news and news-leaning websites), as well as non-journalistic sources of vaccine information, such as governmental websites, healthcare and NGO websites, and medical journals. Given the algorithm’s focus on editorial content, as opposed to other metrics such as author, outlet, or engagement, analyzing a diverse set of article types allowed the research team to examine how different styles of vaccine-related content measured against traditional journalistic quality standards.<strong> </strong>Therefore, this dataset provides a unique insight into the spectrum of vaccine reporting, and serves as a contribution to the field of automated quality assessment. </p> <p><strong>About ARTT:</strong> The Analysis and Response Toolkit for Trust (ARTT) project is focused on helping people engage in trust-building ways when discussing vaccine efficacy and other topics online. </p> <p><strong>About Overtone:</strong> Overtone has built a Natural Language Processing algorithm that finds and sorts online content by its intrinsic qualities, rather than clicks or shares. Safety of ChAdOx1 nCoV-19 vaccination in patients with end-stage renal disease on hemodialysis
<p>This dataset was the minimal dataset of the research article entitle 'Safety of ChAdOx1 nCoV-19 vaccination in patients with end-stage renal disease on hemodialysis'. The study aims to investigate the localized and systemic adverse effects (AEs) within 30-days after the first dose of ChAdOx1 nCoV19 (Oxford-AstraZeneca) vaccination in end-stage renal disease (ESRD) patients receiving hemodialysis (HD). The first dataset is the raw data for logistic regression analyses examining the risk factors in association with AEs, another dataset contains the second- and the seventh- day incidences of various AEs depicted in bar graphs. It comprises variables recording the status of patient-reported local and systemic AEs in questionnaire on the second and seventh days after vaccination as well as patients' baseline demographic and clinical characteristics.</p>
Data from: A Nonadjuvanted Whole-Inactivated Pneumococcal Vaccine Induces Multiserotype Opsonophagocytic Responses Mediated by Noncapsule-Specific Antibodies
<p><em>Streptococcus pneumoniae</em> (Spn) remains a major cause of global mortality, with extensive antigenic diversity between capsular serotypes that poses an ongoing challenge for vaccine development. Widespread use of pneumococcal conjugate vaccines (PCVs) targeting Spn capsules has greatly reduced infections by vaccine-included serotypes, but has led to increased infections by non-included serotypes. To date, high cost of PCVs has also limited their usefulness in low-income regions where disease burdens are highest. To overcome these limitations, serotype-independent vaccines are being actively researched. We have developed a whole-cell gamma-irradiated Spn vaccine (termed Gamma-PN) providing serotype-independent protection. We demonstrate that Gamma-PN immunization via the clinically relevant intramuscular route induces protein-specific antibodies able to bind numerous non-vaccine encapsulated serotypes, which mediate opsonophagocytic killing and protection against lethal challenges. Gamma-PN induced comparable or superior OPKA responses to serotypes found in the licensed Prevnar13 (PCV13), and a superior response to non-included serotypes, including emergent 22F and 35B. Additionally, despite a lower observed reactogenicity, administration of Gamma-PN without adjuvant resulted in higher OPKA responses and improved protection compared to adjuvanted Gamma-PN. To our knowledge, this has never been demonstrated for a whole-inactivated Spn vaccine. Eliminating the requirement for adjuvant comes with numerous benefits for clinical applications of this vaccine, and poses interesting questions for the inclusion of adjuvant in similar vaccines in development.</p>
