Hypoxia inducible factor-1α knockout does not impair acute thermal tolerance or heat hardening in zebrafish
The rapid increase in critical thermal maximum (CTmax) in fish (or other animals), previously exposed to critically high temperature is termed 'heat hardening', which likely represents a key strategy to cope with increasingly extreme environments. The physiological mechanisms that determine acute thermal tolerance, and the underlying pathways facilitating heat hardening, remain debated. It has been posited, however, that exposure to high temperature is associated with tissue hypoxia and may be associated with increased expression of hypoxia-inducible factor-1 (Hif-1). We studied acute thermal tolerance in zebrafish lacking functional Hif-1α paralogs (Hif-1aa and Hif-1ab double knockout; Hif-1α-/-), which are known to exhibit markedly reduced hypoxia tolerance. We hypothesised that Hif-1α-/- zebrafish would suffer reduced acute thermal tolerance relative to wild-types and that the heat hardening ability would be lost. On the contrary, we observed that Hif-1α-/- and wild-type fish did not differ in CTmax, and both genotypes exhibited heat hardening of a similar degree when CTmax was re-tested 48 h later. Despite exhibiting impaired hypoxia tolerance (based on loss of equilibrium experiments), Hif-1α-/- zebrafish display unaltered thermal tolerance, suggesting that these traits are not necessarily functionally associated. Hif-1α is accordingly not required for short-term acclimation in the form of heat hardening.
Data from: Thyroid hormone receptor beta mutations alter photoreceptor development and function in Danio rerio (zebrafish)
<p>We investigate mutations in <i>trβ2</i>, a splice variant of <i>thrb</i>, identifying changes in function, structure, and behavior in larval and adult zebrafish retinas. Two N-terminus CRISPR mutants were identified. The first is a <i>6BP+1</i> insertion deletion frameshift resulting in a truncated protein. The second is a <i>3BP </i>in frame deletion with intact binding domains. ERG recordings of isolated cone signals showed that the <i>6BP+1</i> mutants did not respond to red wavelengths of light while the <i>3BP </i>mutants did respond. <i>6BP+1 </i>mutants lacked optomotor and optokinetic responses to red/black and green/black contrasts. Both larval and adult <i>6BP+1</i> mutants exhibit a loss of red-cone contribution to the ERG and an increase in UV-cone contribution. Transgenic reporters show loss of cone <i>trβ2</i> activation in the <i>6BP+1 </i>mutant but increase in the density of cones with active blue, green, and UV opsin genes. Antibody reactivity for red-cone LWS1 and LWS2 opsin was absent in the <i>6BP+1</i> mutant, as was reactivity for arrestin3a. Our results confirm a critical role for <i>trβ2</i> in long-wavelength cone development.</p>
Vape flavorants dull sensory perception and cause hyperactivity in developing zebrafish embryos
<p>E-cigarette use (vaping) during pregnancy has been increasing, and the potential exists for the developing brain <i>in utero</i> to be exposed to chemical constituents in the vape. Vapes come in over 7,000 unique flavors with and without nicotine, and while nicotine is a known neurotoxicant, the effects of vape flavoring alone, in the absence of nicotine, on brain function are not well understood. Here we performed a screen of vape aerosol extracts (VAEs) to determine the potential for prenatal neurotoxicity using the zebrafish embryo photomotor response (PMR) – a translational biosensor of neurobehavioral effects. We screened three commonly used aerosolized vape liquids (flavored and flavorless) either with or without nicotine. No neurobehavioral effects were detected in flavorless, nicotine-free VAEs, while the addition of nicotine to this VAE dulled sensory perception. Flavored nicotine-free VAEs also dulled sensory perception and caused hyperactivity in zebrafish embryos. The combination of flavor and nicotine produced largely additive effects. Flavored VAEs without nicotine had similar neuroactive potency as nicotine. Similar effects were also seen in embryos exposed to the pure flavoring compound, cinnamaldehyde. Together, using zebrafish PMR as a high throughput translational behavioral model for prenatal exposure, our results demonstrate that e-cigarette flavorants that we screened elicit neurobehavioral effects worthy of further investigation for long-term neurotoxic potential, and also have the potential to modulate nicotine impact on the developing brain.</p>
