Extracellular vesicles from Fusarium graminearum contain protein effectors expressed during infection of corn

<p><em>Fusarium</em> <em>graminearum</em> (<em>Fgr</em>) is a devastating filamentous fungal pathogen that causes diseases in cereals, while producing mycotoxins that are toxic for humans and animals, and render grains unusable. Low efficiency in managing <em>Fgr</em> poses a constant need for identifying novel control mechanisms. Evidence that fungal extracellular vesicles (EVs) from pathogenic yeast have a role in human disease led us to question whether this is also true for fungal plant pathogens. We separated EVs from <em>Fgr</em> and performed a proteomic analysis to determine if EVs carry proteins with potential roles in pathogenesis. We revealed that protein effectors, which are crucial for fungal virulence, were detected in EV preparations and some of them did not contain predicted secretion signals. Furthermore, a transcriptomic analysis of corn (<em>Zea</em> <em>mays</em>) plants infected by <em>Fgr</em> revealed that the genes of some of the effectors were highly expressed in vivo, suggesting that the <em>Fgr</em> EVs are a mechanism for the unconventional secretion of effectors and virulence factors. Our results expand the knowledge on fungal EVs in plant pathogenesis and cross-kingdom communication, and may contribute to the discovery of new antifungals.</p> <p>The following are available online at www.mdpi.com/xxx/s1, Figure S1. <p>Mastodon project containing the cell tracks of a developing <em>Tribolium Castaneum </em>embryo.&nbsp;</p> <p>To open it in Mastodon, download both the <em>.mastodon </em>and the <em>.xml </em>files and put them in the same folder.</p> <p>The image data is stored on a BigDataServer in the Institut Pasteur, Paris, provided by the <a href="https://research.pasteur.fr/en/team/image-analysis-hub/">Image Analysis Hub</a>. When you open this <em>.mastodon </em>file with Mastodon, it will stream the image from the server without you having to download it. Shotgun metagenomic sequencing dataset of a synthetic mock community containing 20 genomes spiked-in at even and staggered concentrations.

<p>Shotgun metagenomics (SM) sequencing is a popular method used in microbial ecology to obtain insights on microbial community structure and function potential in a given biological system without the need to cultivate microorganisms. The dataset described in this article describes technical triplicates of shotgun metagenomic sequence libraries generated from two purified and titrated mixes of 20 distinct reference bacterial genomes for which key characteristics such as genome size, sequence and spiked-in concentrations are known. In one of the genomic DNA mix, each genome is spiked-in at similar concentrations (representing an even microbial community) and in the other, genomes are spiked-in at different concentrations with some genomes highly abundant and other in low quantity, mimicking an uneven microbial community DNA extract. In order to be interpretable, SM sequencing data needs to be properly analyzed by complex analytical bioinformatic pipelines. Environments investigated with this method can range from simple to very complex. Typically, microbial communities contain microbes that are ubiquitous and some others much rarer. Analysis of rare microbes in a complex microbial community are challenging to perform as their sequencing signals get submerged by the microbial genomes that are more abundant. In this context, it is critical to have access to sequencing data of simple mock communities of mixes of well characterized genomes in order to develop and validate bioinformatic methods that aim to accurately analyze microbial communities.</p>

Data for: Higher floral richness promotes rarer bee communities across remnant and reconstructed tallgrass prairies, though remnants contain higher abundances of a threatened bumble bee (Bombus Latreille)

<p>Managing and restoring tallgrass prairie ecosystem is an important form of pollinator conservation in the Midwestern United States. Prairie reconstruction has been found to enhance native bee diversity and abundance, but it is less clear if prairie reconstruction conserves species thought to be at-risk. We reanalyze a previously published dataset on the bee communities of reconstructed and remnant prairie in the US state of Minnesota to investigate how the abundance of at-risk species respond to local factors, such as floral diversity and prairie type (reconstructed or remnant), and landscape factors, in the form of surrounding agricultural production. We defined at-risk species in two ways. For bumble bees, we used the IUCN red list of bumble bees for North America. As other species in the bee community have not been systematically evaluated, we used an independent data set to calculate a community-level measure of rarity as a proxy for at-risk species. We calculated community rarity metrics using a Species Weighted Mean (SWM) approach, with species-level rarity (relative abundance and site occurrence) derived from a regional dataset comprised of over 30,000 specimens from across the US state of Minnesota. We found that the declining bumble bee <em>Bombus</em> <em>fervidus</em> had higher abundances in remnant rather than reconstructed prairies. Floral richness was associated with rarer bee communities (lower SWM values) across remnant and reconstructed prairies. We show that planting and managing prairies for floral diversity promotes bee communities with rarer species, but that remnants better support some at-risk species such as <em>Bombus</em> <em>fervidus</em>. </p>
