Data_ Integrating torrefaction of pulp industry sludge with anaerobic digestion to produce bioenergy and biochemicals: Techno-economic and environmental feasibility analysis

<p>In order to improve the economic competitiveness of bioenergy carriers and biochemicals, they must be produced<br> from low-cost or no-cost feedstock and at a reduced operational expenses. In that regard, this study<br> focused on understanding the techno-economic feasibility of using pulp sludge as a low-cost alternative feedstock<br> to forestry biomass in torrefaction. Economic feasibility of further integrating pulp sludge torrefaction with<br> anaerobic digestion to produce energy, biomethane and volatile fatty acids (VFA) was also studied. The operational<br> expenses were around 8.2 and 2.04 M&euro; and the minimum selling price of torrefied pellets was 407 and<br> 189 &euro;/t for forestry biomass torrefaction and pulp sludge torrefaction respectively. In case of integrated approaches,<br> VFA production showed higher economic feasibility with a torrefied pellets selling price of 163 &euro;/t<br> compared with biomethane production (213 &euro;/t). The biomethane and VFA selling price can be reduced by 90<br> and 64% compared with current market price at torrefied pellets selling price of 260 &euro;/t. Sensitivity analysis<br> revealed that, moisture content of the sludge is the main influencing parameter on the overall economic feasibility<br> of the pulp sludge torrefaction. The environmental analysis showed that pulp sludge torrefaction has higher<br> emissions compared with forestry biomass torrefaction.</p>

Data and software for: Temporal novelty detection and multiple timescale integration drive Drosophila orientation dynamics in temporally diverse olfactory environments

<p>To survive, insects must effectively navigate odors plumes to their source. In natural plumes, turbulent winds break up smooth odor regions into disconnected patches, so navigators encounter brief bursts of odor interrupted by bouts of clean air. The timing of these encounters plays a critical role in navigation, determining the direction, rate, and magnitude of insects' orientation and speed dynamics. Disambiguating the specific role of odor timing from other cues, such as spatial structure, is challenging due to natural correlations between plumes' temporal and spatial features. Here, we use optogenetics to isolate temporal features of odor signals, examining how the frequency and duration of odor encounters shape the navigational decisions of freely-walking <em>Drosophila</em>. We find that fly angular velocity depends on signal frequency and intermittency – fraction of time signal can be detected – but not directly on durations. Rather than switching strategies when signal statistics change, flies smoothly transition between signal regimes, by combining an odor offset response with a frequency-dependent novelty-like response. In the latter, flies are more likely to turn in response to each odor hit only when the hits are sparse. Finally, the upwind bias of individual turns relies on a filtering scheme with two distinct timescales, allowing rapid and sustained responses in a variety of signal statistics. A quantitative model incorporating these ingredients recapitulates fly orientation dynamics across a wide range of environments and shows that temporal novelty detection, when combined with odor motion detection, enhances odor plume navigation.</p>

Here, we identify gene-regulatory landscapes in a wide breadth of functional and dysfunctional CD8<sup>+</sup> TIL states covering four cancer entities using single-cell chromatin profiling. We map enhancer-promoter interactions in human TILs by integrating single-cell chromatin accessibility with single-cell RNA-seq data from tumor-entity-matching samples and prioritize cell-state-specific genes by super-enhancer analysis. Besides revealing entity-specific chromatin remodeling in exhausted TILs, our analyses identify a common chromatin trajectory to TIL dysfunction and determine key enhancers, transcriptional regulators, and deregulated genes involved in this process. Finally, we validate enhancer regulation at immunotherapeutically relevant loci by targeting non-coding regulatory elements with potent CRISPR activators and repressors. In summary, our study provides a framework for understanding and manipulating cell-state-specific gene-regulatory cues from human tumor-infiltrating lymphocytes.</p>

Data from: Displacement experiments provide evidence for path integration in Drosophila

<p>Like many other animals, insects are capable of returning to previously visited locations using path integration, a memory of travelled direction and distance. Recent studies suggest that <em>Drosophila</em> can also use path integration to return to a food reward. However, the existing experimental evidence for path integration in <em>Drosophila</em> has a potential confound: pheromones deposited at the site of reward might enable flies to find previously rewarding locations even without memory. Thus, we designed an experiment to determine if flies can use path integration memory despite potential pheromonal cues by displacing the flies shortly after an optogenetic reward. We found that rewarded flies returned to the location predicted by a memory-based model. Several analyses are consistent with path integration as the mechanism by which flies returned to the reward. We conclude that while pheromones may often be important in fly navigation and must be carefully controlled in future experiments, <em>Drosophila</em> may indeed be capable of performing path integration.</p>

Data from: Evaluating the importance of individual heterogeneity in reproduction to Weddell seal population dynamics using integral projection models

<ol> <li>Identifying and accounting for unobserved individual heterogeneity in vital rates in demographic models is important for estimating population-level vital rates and identifying diverse life-history strategies, but much less is known about how this individual heterogeneity influences population dynamics.</li> <li>We aimed to understand how the distribution of individual heterogeneity in reproductive and survival rates influenced population dynamics using vital rates from a Weddell seal population by altering the distribution of individual heterogeneity in reproduction, which also altered the distribution of individual survival rates through the incorporation of our estimate of the correlation between the two rates and assessing resulting changes in population growth.</li> <li>We constructed an integral projection model (IPM) structured by age and reproductive state using estimates of vital rates for a long-lived mammal that has recently been shown to exhibit large individual heterogeneity in reproduction. Using output from the IPM, we evaluated how population dynamics changed with different underlying distributions of unobserved individual heterogeneity in reproduction.</li> <li>Results indicate that the changes to the underlying distribution of individual heterogeneity in reproduction cause very small changes in the population growth rate and other population metrics. The largest difference in the estimated population growth rate resulting from changes to the underlying distribution of individual heterogeneity was less than 1%.</li> <li>Our work highlights the differing importance of individual heterogeneity at the population level compared to the individual level. Although individual heterogeneity in reproduction may result in large differences in the lifetime fitness of individuals, changing the proportion of above- or below-average breeders in the population results in much smaller differences in annual population growth rate. For a long-lived mammal with stable and high adult-survival that gives birth to a single offspring, individual heterogeneity in reproduction has a limited effect on population dynamics. We posit that the limited effect of individual heterogeneity on population dynamics may be due to canalization of life-history traits.</li> </ol>

<p>This dataset was generated within the research&nbsp;thesis of Axel Hutomo, under the supervision of Leonardo Alfonso and Ioana Popescu at IHE Delft, and it is published as supplementary data for the article <em>Integrating Data-Driven and Hydraulic Modelling with Acoustic Sensor Information for Improved Leak Location in Water Distribution Networks, </em>currently under review.&nbsp;</p> <p>The Excel sheet provides information about the datasets produced to integrate&nbsp;acoustic sensor data and hydraulic&nbsp;model output data, to be used by&nbsp;the Machine Learning model.&nbsp;The acoustic sensor data were obtained by extracting several features in&nbsp;time and frequency domains from each audio file coming from acoustic sensors, whereas hydraulic model data was obtained by modelling these leaks using a pressure-independent analysis.</p> <p>The Python code shows the building of the ANN for leakage modelling prediction, integrating the two datasets above, for different leak rates.</p>

Data from: Sex-dependent integration of ornamentation, personality, morphology and life history

<p>Phenotypic integration can be defined as the patterns and strength of the covariance between traits in an organism. The pace of life syndrome (POLS) hypothesis provides a testable case of phenotypic integration as it predicts that traits that mediate the trade-off between current and future reproduction, should have coevolved with the slow-fast life-history continuum, and may thus covary across individuals of a population. Although the POLS hypothesis has received increasing attention over the last decade, there is still a need for investigating whether POLS are sex-specific or if ornamental traits can be included within the POLS framework. We used nine years of data to describe the integration patterns of ornamental coloration, personality, morphology and life history in blue tits (<em>Cyanistes caeruleus</em>) and whether they differed between males and females. With that aim, we fitted multivariate mixed models separating the among- from the within-individual covariances. We found that the overall integration between the studied traits was weak, but our results suggested some sex-specific covariances at the among- and within-individual levels. Besides, we found several covariances between the ornamental colors and personality in males but not in females. Finally, using structural equation models, we tested for the presence of trait modules (i.e. covariances between traits involved in the same biological function). Our results support the presence of a morphological module, not of ornamental or behavioral modules in males or females. Also, our results do not align with the POLS hypothesis, however they highlight the importance of considering sex when studying phenotypic integration.</p>
