Large scale across-breed genome-wide association study reveals a variant in HMGA2 associated with inguinal cryptorchidism risk in dogs

<p class="MsoNormal"><span>Cryptorchidism is the most common congenital sex development disorder in dogs. Despite this, little progress has been made in understanding its genetic background. Extensive genetic testing of dogs through consumer and veterinary channels using a high-density SNP genotyping microarray coupled with links to clinical records presents the opportunity for a large-scale genome-wide association study to elucidate the molecular risk factors associated with cryptorchidism in dogs. Using an inter-breed genome-wide association study approach, a significant statistical association on canine chromosome 10 was identified, with the top SNP pinpointing a variant of <em>HMGA2 </em>previously associated with adult weight variance. In further analysis we show that incidence of cryptorchidism is skewed towards smaller dogs in concordance with the identified variant's previous association with adult weight. This study represents the first putative variant to be associated with cryptorchidism in dogs.</span></p>

Data from: Metabolic changes associated with two endocrine abnormalities in dogs: elevated fructosamine and low thyroxine

<p>Metabolomics studies in canine endocrine abnormalities are sparse and basic information on these abnormalities must be generated. This dataset includes the 1H NMR metabolic profiling results of 151 dog serum samples. The samples have been collected in an ethical way, being leftovers of clinical diagnostic samples. The sample set includes 25 control samples based on unremarkable routine clinical chemistry and hematology, 79 samples exhibiting high fructosamine which reflects poor glycemic control, and 47 samples with low thyroxine, a metabolically important thyroid hormone.</p> <p>The metabolite data and metadata were utilized in the study: Ottka et al. Metabolic changes associated with two endocrine abnormalities in dogs: elevated fructosamine and low thyroxine.</p> <p>The control group results were utilized also in: Ottka et al. Serum NMR metabolomics uncovers multiple metabolic changes in phenobarbital-treated dogs.</p>

Data from: Grassland type and seasonal effects have a bigger influence on plant functional and taxonomical diversity than prairie dog disturbances in semi-arid grasslands

<p>Prairie dogs (Cynomys sp.) are considered keystone species and ecosystem engineers for their grazing and burrowing activities (summarized here as disturbances). As climate changes and its variability increases, the mechanisms underlying organisms' interactions with their habitat will likely shift. Understanding the mediating role of prairie dog disturbance on vegetation structure, and its interaction with environmental conditions through time, will increase knowledge on the risks and vulnerability of grasslands. Here, we compared how plant taxonomical diversity, functional diversity metrics and community-weighted trait means (CWM) respond to prairie dog C. mexicanus disturbance across grassland types and seasons (dry and wet) in a priority conservation semiarid grassland of Northeast Mexico. Our findings suggest that functional metrics and CWM analyses responded to interactions between prairie dog disturbance, grassland type and season, whilst species diversity and cover measures were less sensitive to the role of prairie dog disturbance. We found weak evidence that prairie dog disturbance has a negative effect on vegetation structure, except for minimal effects on C4 and graminoid cover, but which depended mainly on season. Grassland type and season explained most of the effects on plant functional and taxonomic diversity as well as CWM traits. Furthermore, we found that leaf area as well as forb and annual cover increased during the wet season, independent of prairie dog disturbance. Our results provide evidence that grassland type and season have a stronger effect than prairie dog disturbance on the vegetation of this short-grass, water restricted grassland ecosystem. We argue that focusing solely on disturbance and grazing effects is misleading, and attention is needed on the relationships between vegetation and environmental conditions which will be critical to understand semi-arid grassland dynamics under future climate change conditions in the region. </p>

Life history data for: High investment into reproduction is associated with reduced lifespan in dogs

<p><span>Prominent differences in aging among and within species present an evolutionary puzzle. The theories proposed to explain evolutionary differences in aging are based on the axiom that selection maximizes fitness, not necessarily lifespan. This implies trade-offs between investment into self-maintenance and investment into reproduction, where high investment into growth and current reproduction are associated with short lifespans. Fast growth and large adult size are related with shorter lifespans in the domestic dog, a bourgeoning model in aging research, however, whether reproduction influences lifespan in this system remains unknown. Here we test the relationship between reproduction and differences in lifespan among dog breeds, controlling simultaneously for shared ancestry and recent gene flow. We found that shared ancestry explains a higher proportion of the among-breed variation in life history traits, in comparison with recent gene flow. Our results also show that reproductive investment negatively impacts lifespan, and more strongly so in large breeds, an effect that is not merely a correlated response of adult size. These results suggest that basic life history trade-offs are apparent in a domestic animal whose diversity is the result of artificial selection and that among-breed differences in lifespan are due to a combination of size and reproduction.</span></p>

<p>The behavior of the comprehensive circulating renin‐angiotensin system (RAS) in dogs with myxomatous mitral valve disease (MMVD) before the onset of congestive heart failure remains largely unexplored.</p> <p>Hypothesis/Objectives: The classical and alternative RAS activity and aldosterone concentrations will be significantly higher in dogs with American College of Veterinary Internal Medicine (ACVIM) stage B2 MMVD compared to normal dogs and dogs with ACVIM stage B1 MMVD.</p> <p>Animals: One hundred seventeen client‐owned dogs (normal = 60; B1 = 31; B2 = 26).</p> <p>Methods: Prospective observational study. Angiotensin peptides (AP) and aldosterone concentrations were measured using liquid chromatography and mass spectrometry. Angiotensin converting enzymes 1 and 2 (ACE, ACE2) and renin activity surrogates were calculated from AP concentrations. Equilibrium dialysis (ED) and immediate protease inhibition (PI) methods of AP quantification were compared in 14 healthy dogs.</p> <p>Results: Core RAS activity and aldosterone concentrations did not differ among the 3 groups. However, the balance between the alternative and classical RAS differed, with dogs with stage B2 MMVD having significantly higher ACE2 activity surrogate (ACE2surr) when compared to normal dogs (adjusted P = .02; ratio of medians for ACE2surr [B2:normal], 1.89; 95% confidence interval [CI]: 1.4‐2.6). The ED and PI methods of AP quantification were highly correlated (AngI, r = .9, P &lt; .0001; AngII, r = .8, P = .001).</p> <p>Conclusions and Clinical Importance: Circulating alternative RAS activity, specifically the surrogate measure of ACE2 activity, was increased in dogs with stage B2 MMVD as compared to normal dogs. Equilibrium dialysis results are analogous to immediate protease inhibition in dogs.</p>

Immune-mediated hematological disease in dogs is associated with alterations of the fecal microbiota: a pilot study

<div> <h3>Background</h3> <p>The dog is the most popular companion animal and is a valuable large animal model for several human diseases. Canine immune-mediated hematological diseases, including immune-mediated hemolytic anemia (IMHA) and immune thrombocytopenia (ITP), share many features in common with autoimmune hematological diseases of humans. The gut microbiome has been linked to systemic illness, but few studies have evaluated its association with immune-mediated hematological disease. To address this knowledge gap, 16S rRNA gene sequencing was used to profile the fecal microbiota of dogs with spontaneous IMHA and ITP at presentation and following successful treatment. In total, 21 affected and 13 healthy control dogs were included in the study.</p> <h3>Results</h3> <p>IMHA/ITP is associated with remodeling of fecal microbiota, marked by decreased relative abundance of the spirochete <em>Treponema</em> spp., increased relative abundance of the pathobionts <em>Clostridium septicum</em> and <em>Escherichia coli</em>, and increased overall microbial diversity. Logistic regression analysis demonstrated that <em>Treponema</em> spp. were associated with decreased risk of IMHA/ITP (odds ratio [OR] 0.24&ndash;0.34), while Ruminococcaceae UCG-009 and Christensenellaceae R-7 group were associated with increased risk of disease (OR&thinsp;=&amp;thinsp;6.84 [95% CI 2&ndash;32.74] and 8.36 [95% CI 1.85&ndash;71.88] respectively).</p> <h3>Conclusions</h3> <p>This study demonstrates an association of immune-mediated hematological diseases in dogs with fecal dysbiosis, and points to specific bacterial genera as biomarkers of disease. Microbes identified as positive or negative risk factors for IMHA/ITP represent an area for future research as potential targets for new diagnostic assays and/or therapeutic applications.</p> </div>

Long-term data reveal fitness costs of anthropogenic prey depletion for a subordinate competitor, the African wild dog (Lycaon pictus

<p>Within carnivore guilds, dominant competitors (e.g., lions, <em>Panthera leo</em>) are limited primarily by the density of prey, while subordinate competitors (e.g., African wild dogs, <em>Lycaon pictus</em>) have been limited by the density of dominant competitors. Historically, the fitness and population density of subordinate competitors have not been tightly linked to prey density. However, populations of large herbivores have declined substantially across sub-Saharan Africa due to human impacts, and where prey depletion is severe, fitness costs for competitive subordinates may begin to outweigh the benefits of competitive release. Using long-term intensive monitoring of African wild dogs in Zambia's Luangwa Valley Ecosystem (LVE), we tested the effects of prey depletion on survival and reproduction. Our study area included four contiguous regions that varied in protection, prey density, and lion density. We fit Bayesian Cormack-Jolly-Seber and closed-capture models to estimate effects on survival and population density, and generalized linear models to estimate effects on reproductive success. We found that the LVE is a stronghold for wild dogs, with an estimated median density of 4.0 individuals/100 km<sup>2</sup>. Despite this high density, survival and reproduction differed among regions, and both components of fitness were substantially reduced in the region with the lowest prey density. Anthropogenic prey depletion is becoming an important limiting factor for African wild dogs. If prey depletion (or any other form of habitat degradation) becomes severe enough that its fitness costs outweigh the benefits of competitive release, such changes can fundamentally alter the balance between limiting factors for competitively subordinate species.</p>
