<p>Human induced pluripotent stem cell (iPSC)-derived cardiomyocytes are an established model for testing potential chemical hazards. Inter-individual variability in toxicodynamic sensitivity has also been demonstrated <i>in vitro</i>; however, quantitative characterization of the population-wide variability has not been fully explored. We sought to develop a method to address this gap by combining a population-based iPSC-derived cardiomyocyte model with Bayesian concentration-response modeling. A total of 136 compounds, including 44 pharmaceuticals and 82 environmental chemicals, were tested in iPSC-derived cardiomyocytes from 43 non-diseased humans. Hierarchical Bayesian population concentration-response modeling was conducted for five phenotypes reflecting cardiomyocyte function or viability. Toxicodynamic variability was quantified through the derivation of chemical- and phenotype-specific variability factors (TDVF). Toxicokinetic modeling was used for probabilistic <i>in vitro</i>-to-<i>in vivo </i>extrapolation in order to derive population-wide margins of safety (MOS) for pharmaceuticals and margins of exposure (MOE) for environmental chemicals. Pharmaceuticals were found to be active across all phenotypes. Over half of tested environmental chemicals showed activity in at least one phenotype, most commonly positive chronotropy. TDVF estimates for the functional phenotypes were greater than those for cell viability, usually exceeding the generally-assumed default of ~3. Population variability-based MOS for pharmaceuticals were correctly predicted to be relatively narrow, between 10-100; however, MOE for environmental chemicals, based on population exposure estimates, generally exceeded 1000, suggesting they pose little risk at general population exposures even to sensitive sub populations. This study represents a first of its kind human <i>in vitro</i> model that can be used to characterize toxicodynamic population variability in cardiotoxic risk.</p>
Data from: Natural hazards and wildlife health: the effects of a volcanic eruption on the Andean condor
<p>Volcanic eruptions produce health changes in animals that may be associated with emitted gases and deposited ashes. We evaluated whether the Puyehue–Cordón Caulle volcanic eruption in 2011 produced health changes in the threatened Andean condor (<i>Vultur gryphus</i>) living in the area most affected by the eruption, north-western Patagonia. We studied clinical and biochemical parameters of condors examined before and after the eruption. We also examined concentrations of different metals and metalloids in the blood of individuals sampled after the eruption. The most common clinical abnormality associated with the eruptive process was irritating pharyngitis. In condors sampled after the eruption, blood concentrations of albumin, calcium, carotenoids and total proteins decreased to levels under the reference values reported for this species. We found different chemical elements in the blood of these condors after the eruption, such as arsenic and cadmium, with the potential to produce health impacts. Thus, the health of Andean condors was affected in different ways by the eruption; remaining in the affected area appears to have been costly. However, in comparison to other animal species, the health impacts were not as strong and were mainly related to food shortages due to the decrease in availability of livestock carcasses linked to the eruption. This suggests that condors dealt relatively well with this massive event. Future research is needed to evaluate if the health changes we found reduce the survival of this species, and if the cost of inhabiting volcanic areas has any ecological or evolutionary influence on the condor's life history.</p>
Data from: Comprehensive diagnosis of PCDD/F emission from three hazardous waste incinerators
Comprehensive diagnosis of polychlorinated dibenzo-p-dioxin and dibenzofuran (PCDD/F) emissions was systematically conducted on three hazardous waste incinerators (HWIs.) Results indicated that PCDD/F mainly existed in the solid phase before the bag filter. This was especially true for higher chlorinated dioxin and furan congeners (hexa-, hepta- and octa-.) The aged bag filters tended to increase the gas-phase PCDD/F. Emissions also increased due to PCDD/F desorption from circulated scrubbing solution and plastic packing media used in the wet scrubber. The PCDD/F concentrations were elevated during start-up process, reaching up to 5.4 times higher than those measured during the normal operating period. The ratios of PCDFs/PCDDs revealed that the surface-catalyzed de novo synthesis was the dominant pathway of PCDD/F formation. Installation of more efficient fabric filters, intermittent replacement of circulated scrubbing solution will result in reduced PCDD/F emission. Additionally, 2,3,4,7,8-PeCDF correlated well with the international toxic equivalent quantity (I-TEQ) value, which suggests that 2,3,4,7,8-PeCDF could act as an I-TEQ indicator.
