Biosynthesis, Characterization and Anti-dengue vector activity of Silver Nanoparticles prepared by Azadirachta indica and Citrullus colocynthis

<p><span>Here, we report biosynthesis of silver nanoparticles (AgNPs) using aqueous extracts of (i) <i>Azadirachta indica</i> leaves and (ii) <i>Citrullus colocynthis</i> fruit and their larvicidal activity against <i>Aedes aegypti.</i> The Uv-Vis spectroscopy absorption peaks occurred in range 412-416 nm for <i>A. indica</i> AgNPs and 416-431nm for <i>C. </i><i>colocynthis</i> AgNPs indicating the silver nature of prepared colloidal samples. The Scanning electron microscopy examination revealed the spherical morphology of both types of NPs with average size of 17±4 nm (<i>A. indica</i> AgNPs) and 26±5 nm (<i>C. </i><i>colocynthis</i> AgNPs). The x-ray diffraction pattern confirmed the face centred cubic (FCC) structure with crystallite size of 11±1 nm (<i>A. indica</i> AgNPs) and 15±1 nm (<i>C. </i><i>colocynthis</i> AgNPs) while characteristic peaks appearing in Fourier transform infrared spectroscopy analysis indicated the attachment of different biomolecules on AgNPs.. The larvicidal activity at different concentrations of synthesized AgNPs (1-20 </span>mg/L<span>) and extracts (0.5 % - 1.5 %) against <i>Aedes aegypti</i> was examined for 24 h. A concentration dependent larvicidal potential of both types of AgNPs was observed. The LC<sub>50 </sub>values were found to be 0.3 mg/L and 1.25 mg/L for <i>C. colocynthis</i> AgNPs and <i>A. indica</i> AgNPs respectively. However, extracts did not exhibit any notable larvicidal activity.</span></p>

Data from: Reduced survival and reproductive success generates selection pressure for the dengue mosquito Aedes aegypti to evolve resistance against infection by the microsporidian parasite Vavraia culicis

The success and sustainability of control measures aimed at reducing the transmission of mosquito-borne diseases will depend on how they influence the fitness of mosquitoes in targeted populations. We investigated the effects of the microsporidian parasite Vavraia culicis on the survival, blood-feeding behaviour and reproductive success of female Aedes aegypti mosquitoes, the main vector of dengue. Infection reduced survival to adulthood and increased adult female mosquito age-dependent mortality relative to uninfected individuals; this additional mortality was closely correlated with the number of parasite spores they harboured when they died. In the first gonotrophic cycle, infected females were less likely to blood-feed, took smaller meals when they did so, and developed fewer eggs than uninfected females. Even though the conditions of this laboratory study favoured minimal developmental times, the costs of infection were already being experienced by the time females reached an age at which they could first reproduce. These results suggest there will be selection pressure for mosquitoes to evolve resistance against this pathogen if it is used as an agent in a control program to reduce the transmission of mosquito-borne human diseases.

Optimal control approaches for combining medicines and mosquito control in tackling dengue

Dengue is a debilitating and devastating viral infection spread by mosquito vectors, and over half the world's population currently live at risk of dengue (and other flavivirus) infections. Here we use an integrated epidemiological and vector ecology framework to predict optimal approaches for tackling dengue. Our aim is to investigate how vector control and/or vaccination strategies can be best combined and implemented for dengue disease control on small networks, and whether these optimal strategies differ under different circumstances. We show that that a combination of vaccination programmes and the release of genetically modified self-limiting mosquitoes (comparable to sterile insect approaches) is always considered the most beneficial strategy for reducing the number of infected individuals, due to both methods having differing impacts on the underlying disease dynamics. Additionally, depending on the impact of human movement on the disease dynamics, the optimal way to combat the spread of dengue is to focus prevention efforts on large population centres. Using mathematical frameworks, such as optimal control, are essential in developing predictive management and mitigation strategies for dengue disease control.
