<p>This repository contains scripts and datasets associated with the manuscript "tttrlib: modular software for integrating fluorescence spectroscopy and imaging". The software is designed to facilitate the analysis of fluorescence spectroscopy and imaging data to enable integration with molecular modeling.</p> <p>The provided scripts encompass various functionalities, including:</p> <ul> <li>Single-molecule FRET (smFRET) analysis for studying conformational dynamics, particularly focusing on human guanylate binding protein 1 (hGBP1).</li> <li>Comprehensive image spectroscopy workflows applicable to murine guanylate binding protein 2 (mGBP2), featuring intensity and time-resolved analyses.</li> <li>Tools for burst variance analysis (BVA), multiparameter fluorescence detection, and correlative analysis of fluorescence lifetime and intensity.</li> </ul> <p>Datasets for both hGBP1 and mGBP2 are included to exemplify the software's capabilities in real-world applications.</p>

Functional near-infrared spectroscopy visually evoked measurements and Autism Questionnaire score in adults and children

<p><strong>Functional near-infrared spectroscopy (fNIRS) visually-evoked data collected from healthy adults&nbsp;and children. One of the keys for developing appropriate fabrication methods is determination of the composition and phase. Here we explore the FeGe system and establish reference Raman signatures for the distinction between the FeGe hexagonal and cubic structures, as well as FeGe<sub>2</sub> and Fe<sub>2</sub>Ge<sub>3</sub> phases. The experimental results are substantiated by first principles lattice dynamics calculations as well as by complementary structural characterizations such as transmission electron microscopy and X-Ray diffraction, along with the magnetic measurements.</p>

Also included is an .xlsx document with fitted timescales for all included molecules and Matalb structure titled &#39;FinalDataAndStatistics.mat&#39;, which includes the final data, and statistics for the data used within the paper.</p>

The inner mechanics of rhodopsin guanylyl cyclase during cGMP-formation revealed by real-time FTIR spectroscopy

<p>Enzymerhodopsins represent a recently discovered class of rhodopsins which includes histidine kinase rhodopsin, rhodopsin phosphodiesterases and rhodopsin guanylyl cyclases (RGCs). The regulatory influence of the rhodopsin domain on the enzyme activity is only partially understood and holds the key for a deeper understanding of intra-molecular signaling pathways. Here we present a UV-Vis and FTIR study about the light-induced dynamics of a RGC from the fungus <em>Catenaria anguillulae</em>, which provides insights into the catalytic process. After the spectroscopic characterization of the late rhodopsin photoproducts, we analyzed truncated variants and revealed the involvement of the cytosolic N-terminus in the structural rearrangements upon photo-activation of the protein. We tracked the catalytic reaction of RGC and the free GC domain independently by UV-light induced release of GTP from the photolabile NPE-GTP substrate. Our results show substrate binding to the dark-adapted RGC and GC alike and reveal differences between the constructs attributable to the regulatory influence of the rhodopsin on the conformation of the binding pocket. By monitoring the phosphate rearrangement during cGMP and pyrophosphate formation in light-activated RGC, we were able to confirm the M state as the active state of the protein. The described setup and experimental design enable real-time monitoring of substrate turnover in light-activated enzymes on a molecular scale, thus opening the pathway to a deeper understanding of enzyme activity and protein-protein interactions.</p>
