Data from: Associations between testosterone and immune activity in alligators depend on bacteria species and temperature

<p>Males often use elaborate sexual traits to enhance reproduction, but can experience trade-offs between investment in these traits and other physiological needs. The immunocompetence handicap hypothesis (ICHH) postulates that testosterone supports development of sexual traits while also suppressing immunity. While the ICHH implicates testosterone-mediated immunosuppression as a key mechanism of honest signaling in males, conflicting patterns across studies suggest that testosterone-immunity interactions are complex. In this study, we test the ICHH in free-ranging alligators and examine how intrinsic (steroid hormone levels) and extrinsic (temperature) factors modulate testosterone-immunity relationships. Specifically, we quantified simultaneous effects of testosterone and dehydroepiandrosterone (DHEA) on killing capacity of plasma against three bacteria species at two challenge temperatures. We found that DHEA was important in predicting the effects of testosterone on killing capacity. Further, testosterone-mediated effects were dependent on both temperature and bacteria species. Our results highlight the context dependency of interactions between testosterone and immunity, and illustrate the importance of evaluating the ICHH in natural systems to identify key intrinsic and extrinsic factors mediating testosterone-immunity trade-offs.</p>

Data from: An experimental test of the relationship between yolk testosterone and the social environment in a colonial passerine

Maternal hormones can be transferred to offspring during prenatal development in response to the maternal social environment, and may adaptively alter offspring phenotype. For example, numerous avian studies show that aggressive competition with conspecifics tends to result in females allocating more testosterone to their egg yolks, and this may cause offspring to have more competitive phenotypes. However, deviations from this pattern of maternal testosterone allocation are found, largely in studies of colonial species, and have yet to be explained. Colonial species may have different life-history constraints causing different yolk testosterone allocation strategies in response to conspecific competition, but few studies have experimentally tested whether colonial species do indeed differ from that of solitary species. To test this, we collected eggs from zebra finches (Taeniopygia guttata), a colonial species, in the presence and absence of conspecific intrusions. Females did not alter the concentration of testosterone deposited in eggs laid during intrusions despite becoming more aggressive. These results suggest that maternal effects are not characterized by a uniform response to the social environment, but rather need to be contextualized with life-history traits.

Data from: Testosterone in ancient hair from an extinct species

Testosterone is a key regulator in vertebrate development, physiology, and behaviour. Whereas technology allows extraction of a wealth of genetic information from extant as well as extinct species, complimentary information on steroid hormone levels may add a social, sexual, and environmental context. Hair shafts have been previously used to sequence DNA from &gt;50,000 14C years old Siberian woolly mammoths (Mammuthus primigenius). Hair-testing has also been used to measure endogenous steroids in multiple extant species. Here we use small quantities of woolly mammoth hair samples to measure testosterone, and a genomics-based approach to determine sex, in permafrost-preserved mammoths dated to circa 10-60 thousand 14C years. Our validated method opens up exciting opportunities to measure multiple steroids in keratinized tissues from extinct populations of mammals. This may be specifically applied to investigating life histories, including the extinct Quaternary megafauna populations whose remains are preserved in the permafrost throughout the northern hemisphere.

Data from: Fighting experience alters brain androgen receptor expression dependent on testosterone status

Contest decisions are influenced by the outcomes of recent fights (winner–loser effects). Steroid hormones and serotonin are closely associated with aggression and therefore probably also play important roles in mediating winner–loser effects. In mangrove rivulus fish, Kryptolebias marmoratus, individuals with higher testosterone (T), 11-ketotestosterone and cortisol levels are more capable of winning, but titres of these hormones do not directly mediate winner–loser effects. In this study, we investigated the effects of winning/losing experiences on brain expression levels of the receptor genes for androgen (AR), oestrogen α/β (ERα/β), glucocorticoid (GR) and serotonin (5-HT1AR). The effect of contest experience on AR gene expression depended on T levels: repeated losses decreased, whereas repeated wins increased AR gene expression in individuals with low T but not in individuals with medium or high T levels. These results lend strong support for AR being involved in mediating winner–loser effects, which, in previous studies, were more detectable in individuals with lower T. Furthermore, the expression levels of ERα/β, 5-HT1AR and GR genes were higher in individuals that initiated contests against larger opponents than in those that did not. Overall, contest experience, underlying endocrine state and hormone and serotonin receptor expression patterns interacted to modulate contest decisions jointly.

Assessment of plasma testosterone, estradiol, and progesterone levels in Vietnam female patients with moderate to severe acne vulgaris

<p><strong>Introduction</strong>: Acne is a chronic inflammatory disorder of the pilosebaceous unit with differential pathogenesis. To elucidate the roles of hormones in acne pathogenesis, we conducted a study to evaluate the plasma testosterone, estradiol, and progesterone levels in women with acne vulgaris.</p> <p><strong>Methods</strong>: We conducted a cross-sectional descriptive study, and 1<span>40</span> women with acne vulgaris were examined; their plasma estradiol, progesterone, and testosterone were analyzed by chemiluminescence technique and compared with the healthy control group.</p> <p><span><strong>Results</strong>:</span><span> </span>Increased plasma hormone levels in women with acne vulgaris accounted for 33.57%, and hyperandrogenism accounted for 20.71% of cases. We found significant differences in testosterone levels (mean value, 56.92 ± 27.64ng/dL, 60.71± 25.85ng/dL versus 38.35 ± 10.09ng/dL, p&lt;0.05), respectively, in the moderate to severe acne group and the control group. However, the estradiol level of the moderate and severe acne groups (325.12 ± 91.79 and 305.26 ± 83.01pmol/l) was lower than the control group (368.6 ± 58.34pmol/l), with p-value under 0.05. No statistically significant differences were found for progesterone levels. </p> <p><span><strong>Conclusion</strong>:</span><span> Our study showed that female patients with moderate to severe acne vulgaris had abnormalities in plasma testosterone and estradiol levels. These abnormalities might be part roles in the pathogenesis of acne vulgaris, even when the mean levels were in the normal range. Female patients suffering from premenstrual acne flares might be examined for plasma testosterone levels.</span></p>

Data from: Female MHC type affects male testosterone levels and sperm number in the horse (Equus caballus)

Odours of vertebrates often contain information about the major histocompatibility complex (MHC), and are used in kin recognition, mate choice or female investment in pregnancy. It is, however, still unclear whether MHC-linked signals can also affect male reproductive strategies. We used horses (Equus caballus) to study this question under experimental conditions. Twelve stallions were individually exposed either to an unfamiliar MHC-similar mare and then to an unfamiliar MHC-dissimilar mare, or vice versa. Each exposure lasted over a period of four weeks. Peripheral blood testosterone levels were determined weekly. Three ejaculates each were collected in the week after exposure to both mares (i.e. in the ninth week) to determine mean sperm number and sperm velocity. We found high testosterone levels when stallions were kept close to MHC-dissimilar mares and significantly lower ones when kept close to MHC-similar mares. Mean sperm number per ejaculate (but not sperm velocity) was positively correlated to mean testosterone levels and also affected by the order of presentation of mares: sperm numbers were higher if MHC-dissimilar mares were presented last than if MHC-similar mares were presented last. We conclude that MHC-linked signals influence testosterone secretion and semen characteristics, two indicators of male reproductive strategies.

Data from: Pair bonding prevents reinforcing effects of testosterone in male California mice in an unfamiliar environment

Testosterone (T) can be released by stimuli such as social interactions, and thereby influence future social behaviours. Because the reinforcing effects of T can induce preferences for specific environmental locations, T has the potential to alter behaviour through space use. In a monogamous species, this T pulse may contribute differently to space use in sexually naive (SN) and pair-bonded (PB) males: SN males may be more likely to explore new areas to set up a territory than PB males, which are more likely to defend an existing, established territory. In this study, we test for variation in T-driven space use by examining variation in the formation of conditioned place preferences (CPPs) in SN and PB male California mice. In the three-chambered CPP apparatus, subcutaneous administrations of physiological levels of T were used to repeatedly condition SN and PB males to a side chamber, which is an unfamiliar/neutral environment. The final tests revealed that T-induced CPPs in the side chamber are developed in SN, but not PB males. This study fills a gap in our knowledge about plasticity in the rewarding nature of T pulses, based on past social experience.
