Reservoir dynamics of rabies in Southeast Tanzania and the roles of cross-species transmission and domestic dog vaccination

<p>Understanding the role of different species in the transmission of multi-host pathogens, such as rabies virus, is vital for effective control strategies. Across most of sub-Saharan Africa domestic dogs (Canis familiaris) are considered the reservoir for rabies, but the role of wildlife has been long debated. Here we explore the multi-host transmission dynamics of rabies across southeast Tanzania.</p> <p>Between January 2011 and July 2019 data on probable rabies cases were collected in the regions of Lindi and Mtwara. Hospital records of animal-bite patients presenting to healthcare facilities were used as sentinels for animal contact tracing. The timing, location and species of probable rabid animals was used to reconstruct transmission trees to infer who infected whom and the relative frequencies of within- and between-species transmission.</p> <p>During the study, 688 probable human rabies exposures were identified, resulting in 47 deaths. Of these exposures, 389 were from domestic dogs (56.5%) and 262 from jackals (38.1%). Over the same period 549 probable animal rabies cases were traced: 303 in domestic dogs (55.2%) and 221 in jackals (40.3%).</p> <p>Although dog-to-dog transmission was most commonly inferred (40.5% of transmission events), a third of inferred events involved wildlife-to-wildlife transmission (32.6%) and evidence suggested some sustained transmission chains within jackal populations.</p> <p>A steady decline in probable rabies cases in both humans and animals coincided with the implementation of widespread domestic dog vaccination during the first six years of the study. Following the lapse of this programme dog rabies cases began to increase in one of the northernmost districts.</p> <p>Synthesis and applications: In southeast Tanzania, despite a relatively high incidence of rabies in wildlife and evidence of wildlife-to-wildlife transmission, domestic dogs remain essential to the reservoir of infection. Continued dog vaccination alongside improved surveillance would allow a fuller understanding of the role of wildlife in maintaining transmission in this area. Nonetheless, dog vaccination clearly suppressed rabies in both domestic dog and wildlife populations, reducing both public health and conservation risks and, if sustained, has potential to eliminate rabies from this region.</p>

COVID19 vaccination choice among Iraqi students at Al-Zahraa University for women

<p>198 first-year students in the Department of Anesthesia at University of Al-Zahra participated in this cross-sectional questionnaire-based study. A Google Classroom event was attended by 198 students resulting in a response rate of (100 %). This research study was analytical in nature. The questionnaire was created by the researcher, and a few questions were adapted from other studies and incorporated into the questionnaire. Ethical approval was granted by the independent ethics committee of Al-Zahra University in Karbala-Iraq prior to the research. The period of research and data collection was between Feb. 2021 and August 2021. Questionnaires were distributed among the students using Google Classroom and were returned by the students after completion via the same method. The responses were retrieved as an excel file from the google form a questionnaire and imported into SPSS version.23 to be analyzed and frequencies and percentages determined. The questionnaire is related to COVID19 vaccination preference. The questionnaire is comprised of four tables with eight questions. Statistical tests used frequency and rates.</p>

Survey data of the health literacy on COVID-19 and COVID-19 vaccination in Indonesia

<p><span><strong>Introduction</strong>: </span><span>Health literacy on COVID-19 and COVID-19 vaccination is valuable during the pandemic. The objective of this study was to determine the levels of health literacy about the COVID-19 vaccine and vaccination (Vaccine and Vaccination literacy—VL) in the Indonesian adult general population, assessing the perceptions of the respondents/interviewees about current adult immunization and beliefs about vaccination in general, and analyzing correlations of these variables with the VL levels.</span><span> </span></p> <p><span><strong>Methods</strong>: </span><span>A rapid survey was administered via the web. Data were analyzed using descriptive and inferential stats; the internal consistency of the VL scales was assessed through Cronbach's alpha coefficient, and a Principal Component Analysis (PCA) was conducted to investigate how the questions of the functional and interactive-critical VL scales were related to one another and whether the underlying components (factors) and each question's load on the components could be identified as anticipated. An alpha level lesser than 0.05 was considered significant.</span></p> <p><span><strong>Results</strong>: </span><span>Answers to functional- and interactive/ critical- VL questions showed good/ acceptable internal consistency (Cronbach's alpha = 0.817 and 0.699, respectively), lowest values observed were 0.806 for functional scale and 0.640 for the interactive-critical scale. PCA showed two components accounting for 52.45% of the total variability. Approximately 60% of respondents were females (n=686). Almost all respondents used the internet to seek information regarding COVID-19 and COVID-19 vaccination. Many used at least one social media actively with 74.4% of respondents sometimes believing the validity of this information.</span></p> <p><span><strong>Conclusions:</strong> </span><span>High scores were observed in both functional- and interactive/ critical-VL, and were quite balanced between genders in the prior VL and higher in females for the latter; these were also closely related to the educational level and age group. It is crucial to increase public health literacy on managing the pandemic.</span></p>

Vaccinating badgers in a post-cull landscape; insights from the field

<p>In 2010 the BadgerBCG vaccine was licensed for use in badgers in the UK to reduce the severity of Mycobacterium bovis infection, and hence the risks of onward transmission to cattle. To date badger vaccination in the UK has been deployed at a relatively limited spatial scale (compared to the large scale badger culls) and almost exclusively in high density badger populations which have not been recently culled. UK Government policy direction has moved towards the wider roll-out of badger vaccination as an exit strategy following culling. Field resources required to carry out vaccination in undisturbed badger populations are well documented, but levels of effort are unlikely to be directly transferable to previously culled populations where badger density and social behaviour may be markedly different. We present an evidence based assessment of the likely effort required to vaccinate badger populations that have recently been culled, drawing on data from past culling operations, vaccination operations in a previously culled area and the practical field experiences of expert badger trappers. Trapping efficiency declined over successive years of industry-led culling, however this effect was not consistently noted in the government-run Randomised Badger Culling Trial (RBCT). Fewer badgers were removed using cage-trapping (compared to 'controlled shooting') in the latter years of intensive industry-led culls. When trapping badgers for vaccination in areas that had previously been culled, highly experienced government field staff adapted their practices in response to the lower density and likely more mobile residual population. A longer and more variable pre-baiting period was expected and there was a greater reliance on higher levels of skill and experience in interpreting field signs and trapping effectively in such populations.</p>

Data for: Oligonucleotide mapping via mass spectrometry to enable comprehensive primary structure characterization of an mRNA vaccine against SARS-CoV-2

<p>Oligonucleotide mapping via liquid chromatography mass spectrometry mass spectrometry (LC-MS/MS) was recently developed to support development of Comirnaty®, the world's first commercial mRNA vaccine which immunizes against the SARS-CoV-2 virus. Analogous to peptide mapping of therapeutic protein modalities, oligonucleotide mapping described here provides direct primary structure characterization of mRNA, through enzymatic digestion, accurate mass determinations, and optimized collisionally-induced fragmentation. Sample preparation for oligonucleotide mapping is a rapid, one-pot, one-enzyme digestion. The digest is analyzed via LC-MS/MS with an extended gradient and resulting data analysis employs semi-automated software. In a single method, oligonucleotide mapping readouts include a highly reproducible and completely annotated UV chromatogram with &gt;98% sequence coverage and a microheterogeneity assessment of 5´ terminus capping and 3´ terminus poly(A) tail length. Oligonucleotide mapping was pivotal to ensure the quality, safety, and efficacy of mRNA vaccines by providing: confirmation of construct identity and primary structure and assessment of product comparability following manufacturing process changes. More broadly, this technique may be used to directly interrogate the primary structure of RNA molecules in general.</p>

Raw Data for the article: Analysis of the Specific Immune Response after the Third Dose of mRNA COVID-19 Vaccines in Organ Transplant Recipients: Possible Spike-S1 Reactive IgA Signature in Protection from SARS-CoV-2 Infection

<p><strong>Background:</strong>&nbsp;Several studies have indicated that anti-SARS-CoV-2 mRNA vaccinations are less effective in inducing robust immune responses among solid organ transplant recipients (SOTRs) compared with the immunocompetent. The third dose of vaccine in SOTRs showed promising results of immunogenicity, even though clinical studies have suggested that immunocompromised subjects are less likely to build a protective immune response against SARS-CoV-2 resulting in lower vaccine efficacy for the prevention of severe COVID-19.&nbsp;<strong>Methods:</strong>&nbsp;Serological IgG and IgA were analyzed through CLIA or ELISA, respectively, while Spike-specific T cells were detected by ELISpot assay after the second and third dose of vaccine in 43 SOTRs.&nbsp;<strong>Results:</strong>&nbsp;The third dose induced an improvement in antibody response against SARS-CoV-2. We also reported a strong correlation between specific humoral and cellular responses after the third dose, even though we did not see significant changes in the magnitude of the SARS-CoV-2-specific T cell response. SOTRs who contracted the SARS-CoV-2 infection after the third dose, despite eliciting a positive IgG response, failed to mount an anti-Spike-S1 IgA response, both after the third dose and after SARS-CoV-2 infection.&nbsp;<strong>Conclusions:</strong>&nbsp;We can conclude that serum IgA detection can be helpful, along with IgG detection, for the evaluation of vaccine efficacy, principally in fragile subjects at high risk of infection.</p>

The tegumentary <em>Sm</em>-TSP-2 antigen was selected based on its unique recognition by cytophilic antibodies in putatively immune individuals living in areas of ongoing <em>S</em>. <em>mansoni</em> transmission in Brazil, and preclinical studies in which vaccination with <em>Sm</em>-TSP-2 protected mice following infection challenge. </p>

Framework for virtual education of COVID-19 vaccines for Mandarin-speaking learners: An educational intervention module

<p><strong>Background</strong>: In the United States, patients with limited English proficiency face significant barriers to comprehending and acting upon health-related information, particularly during the COVID-19 pandemic. The ability of health professionals to communicate COVID-19-related information to Mandarin-speaking patients has proved critical in discussions about vaccine efficacy, side effects, and post-vaccine protection.</p> <p><strong>Methods</strong>: The authors created a one-hour educational module to help Mandarin-speaking medical students better convey COVID-19 vaccine information to Mandarin-only speakers. The module is composed of an educational guide, which introduced key terminology and addressed commonly asked questions, and pre- and post-surveys. The authors recruited 59 Mandarin-speaking medical students from 31 U.S. academic medical centers, all of whom had previously completed a medical Mandarin elective. The module and surveys were distributed and completed in August 2021. Data analysis measured the change in aggregate mean for subjective five-point Likert-scale questions and change in percent accuracy for objective knowledge-based questions.</p> <p><strong>Results</strong>: The educational module significantly improved participants' subjective comfort level in discussing the COVID-19 vaccine in English and Mandarin. The largest improvement in both English and Mandarin was demonstrated in the participant's ability to explain differences between the COVID-19 vaccines, with an aggregate mean improvement of 0.39 for English and 1.48 for Mandarin. Survey respondents also demonstrated increased percent accuracy in knowledge-based objective questions in Mandarin.</p> <p><strong>Conclusions</strong>: This module provides Mandarin-learning medical students with skills to deliver reliable information to the general population and acts as a model for the continued development of educational modules for multilingual medical professionals.</p>

Visual policy narrative messaging and COVID-19 vaccine uptake

<p><span>In the face of vaccine hesitancy, public health officials are seeking more effective risk communication approaches to increase vaccination rates. We test the influence of visual policy narratives on COVID-19 vaccination behavior through a panel survey experiment conducted in early 2021 (n=3900) and then eight weeks later (n=2268). We examine the effects of three visual policy narrative messages that test the narrative mechanism of character selection (<em>yourself, your circle, </em>and<em> your community</em>) and a non-narrative control on COVID-19 vaccine behavior. Visual risk messages that use narratives positively influence COVID-19 vaccination through serial mediation of affective response to the messages and motivation to get the COVID-19 vaccination. Additionally, character selection matters, as messages focusing on protecting others (i.e., <em>your</em> <em>circle</em> and <em>your</em> <em>community</em>) perform stronger than that of <em>yourself</em>. Political ideology moderated some of the effects, with conservative respondents in the non-narrative control condition having a higher probability of vaccination in comparison to the <em>protect</em> <em>yourself</em> condition. Taken together, these results suggest that public health officials should use narrative-based visual communication messages that emphasize communal benefits of vaccinations.</span></p>

Macroeconomic Impact of Ebola outbreaks in SSA and potential mitigation of GDP loss with prophylactic vaccination programs

<p>Introduction:&nbsp;Decisions about prevention of and response to Ebola outbreaks require an understanding of the macroeconomic implications of these interventions. Prophylactic vaccines hold promise to mitigate the negative economic impacts of infectious disease outbreaks. The objective of this study was to evaluate the relationship between outbreak size and economic impact among countries with recorded Ebola outbreaks and to quantify the hypothetical benefits of prophylactic Ebola vaccination interventions in these outbreaks.</p> <p>Methods: The synthetic control method was used to estimate the causal impacts of Ebola outbreaks on per capita gross domestic product (GDP) of five countries in sub-Saharan Africa that have previously experienced Ebola outbreaks between 2000 and 2016, where no vaccines were deployed. Using illustrative assumptions about vaccine coverage, efficacy, and protective immunity, the potential economic benefits of prophylactic Ebola vaccination were estimated using the number of cases in an outbreak as a key indicator.</p> <p>Results:&nbsp;The impact of Ebola outbreaks on the macroeconomy of the selected countries led to a decline in GDP of up to 36%, which was greatest in the third year after the onset of each outbreak and increased exponentially with the size of outbreak (i.e., number of reported cases). Over three years, the aggregate loss estimated for Sierra Leone from its 2014-2016 outbreak is estimated at 16.1 billion International$.&nbsp;&nbsp;Prophylactic vaccination could have prevented up to 89% of an outbreak&rsquo;s negative impact on GDP, reducing the outbreak&rsquo;s impact to as little as 1.6% of GDP lost.&nbsp;</p> <p>Conclusion:&nbsp;This study supports the case that macroeconomic returns are associated with prophylactic Ebola vaccination. Our findings support recommendations for prophylactic Ebola vaccination as a core component of prevention and response measures for global health security.&nbsp;</p>
