SpaceX Inspiration4 Oral, Nasal, and Skin Metagenomic and Metatranscriptomic Microbial Swabs
The SpaceX Inspiration4 mission was a 3-day mission with four private astronauts to low Earth orbit that occurred in September 2021. The crew collected biospecimen samples before, during, and after flight. One of these biospecimen collections included oral, nasal, and skin microbial swabs collected from ten locations (oral, nasal cavity, post-auricular, axillary vault, volar forearm, occiput, umbilicus, gluteal crease, glabella, and toe web space). Skin swabs were first dipped in nuclease-free water before swabbing each region for 30 seconds. Oral and nasal swabs did not include this swab-wetting step. Nuclease-free water samples were sequenced as controls. Swabs were collected at three pre-flight timespoints (L-92, L-44, L-3), two in-flight timepoints (flight day 2 (FD2), and flight day 3 (FD3)), and three post-flight timepoints (R+1, R+45, R+82). Swabs were stored in 400uL of Zymo Research DNA/RNA Shield at 4C until nucleic acid extraction. DNA and RNA were extracted from the same swab to generate metagenomic and metatranscriptomic profiles.

The SpaceX Inspiration4 mission was a 3-day mission with four private astronauts to low Earth orbit that occurred in September 2021. The crew collected biospecimen samples before, during, and after flight. One of these biospecimen collections included microbial swabs collected from nine surfaces in the Dragon capsule (execute button, G-meter button, control touch screen - left, control touch screen - right, side hatch mobility aid, lid of waste locker, seat 2, commode panel, viewing dome) and one open air control. Swabs were collected twice during flight (flight day 2 (FD2), and flight day 3 (FD3)) and twice pre-flight in the crew training capsule in Hawthorne, CA (L-92, L-44). Swabs were stored in 400uL of Zymo Research DNA/RNA Shield at 4C until nucleic acid extraction. DNA and RNA were extracted from the same swab to generate metagenomic and metatranscriptomic profiles.

<p>The database includes the raw data of the article “Comparison of two RNA Extraction Methods for the Molecular Detection of SARS-CoV-2 from nasopharyngeal swab samples”.</p> <p>Aim of this work was to compare a two commercially available kit for viral RNA extraction from human swab samples that differ mainly for time consuming (9 min. vs 35 min.) in order to improve SARS-CoV-2 diagnostic workflow.</p> <p>The database contains the data obtained by the following evaluations:</p> <p>a) RNA analysis for the evaluation of quality and quantity by nanodrop instrument</p> <p>b) Real Time PCR analysis of SARS-CoV-2 genes and internal control.</p> <p>The two procedures evaluated gave similar analytical results in terms of extracted RNA analysis and Real Time PCR data for gene expression.</p>
