Utilizing Hydrothermal Processing to Align Structure and In Vitro Digestion Kinetics between Three Different Pulse Types

Consequently, digestion is anticipated to emerge from the combined effect of intrinsic (matrix-dependent) and extrinsic (processed-induced) factors. In this work, we aimed to investigate the interrelated effect of intrinsic and extrinsic factors on pulses&rsquo; structural architecture and resulting digestive consequences. Three commercially relevant pulses (chickpea, pea, black bean) were selected based on reported differences in macronutrient and cell wall composition. Starch and protein digestion kinetics of hydrothermally processed whole pulses were assessed along with microstructural and physicochemical characteristics and compared to the digestion behavior of individual cotyledon cells isolated thereof. Despite different rates of hardness decay upon hydrothermal processing, the pulses reached similar residual hardness values (40 N). Aligning the pulses at the level of this macrostructural property translated into similar microstructural characteristics after mechanical disintegration (isolated cotyledon cells) with comparable yields of cotyledon cells for all pulses (41&ndash;62%). We observed that processing to equivalent microstructural properties resulted in similar starch and protein digestion kinetics, regardless of the pulse type and (prolonged) processing times. This demonstrated the capacity of (residual) hardness as a food structuring parameter in pulses. Furthermore, we illustrated that the digestive behavior of isolated cotyledon cells was representative of the digestion behavior of corresponding whole pulses, opening up perspectives for the incorporation of complete hydrothermally processed pulses as food ingredients.</p>

Data set from 'Sequential Feature Selection for Power System Event Classification Utilizing Wide-Area PMU Data'

<p>The increasing penetration of intermittent, nonsynchronous<br> generation has led to a reduction in total power<br> system inertia. Low inertia systems are more sensitive to sudden<br> changes, and more susceptible to secondary issues that can result<br> in large scale events. Due to the short time frames involved,<br> automatic methods for power system event detection and diagnosis<br> are required. Wide-area monitoring systems can provide<br> the data required to detect and diagnose events; however due to<br> the increasing quantity of data it is next to impossible for power<br> system operators to manually process raw data. The important<br> information is required to be extracted and presented to system<br> operators for real/near-time decision making and control. This<br> paper demonstrates an approach for the wide-area classification<br> of a number of power system events. A mixture of sequential<br> feature selection and linear discriminant analysis is adopted<br> to reduce the dimensionality of PMU data. Successful event<br> classification is obtained by employing quadratic discriminant<br> analysis on wide-area synchronized frequency, phase angle and<br> voltage measurements. The reliability of the proposed method is<br> evaluated using simulated case studies and benchmarked against<br> other classification methods.</p>

Cloud computing is one of the most popular and sophisticated technologies adopted by organizations worldwide. Some world-leading organizations enhance their efficiency and effectiveness by using cloud computing technology. Working from home (WFH) has been a popular trend among organizations during the coronavirus (COVID-19) pandemic. The COVID-19 saw a breakthrough in work cultures and environments where working from home was a remarkable success in remote working environments, despite being a rare phenomenon in Sri Lanka. Yet, it is argued that the deployment of work from home has not been effective among Sri Lankan business organizations due to a lack of IT infrastructure, facilities, and knowledge. The purpose of the study is to investigate the impact of cloud computing, embracing the service models (Infrastructure as a Service, Platform as a Service, and Software as a Service) as theoretical lenses and testing the COVID-19 as the moderator. The study has been conducted based on a deductive approach and adopted a stratified random sampling method. The sample consisted of 384 IT employees among those who had experienced working from home. The study utilized multiple regression and found that cloud computing service models significantly impact work from home with the moderating effect of COVID-19.

Risky sexual behaviours and utilization of HIV testing services among the adolescent girls and young women aged between 15-24 years in Kibra Sub County, Nairobi County, Kenya

<p><strong>Introduction: </strong>HIV remains a significant global health challenge, disproportionately affecting adolescent girls and young women (AGYW). HIV testing is crucial in controlling the infection and reducing its prevalence. Understanding risky sexual behaviors among AGYW is pivotal in aligning prevention interventions. Despite global prevention efforts, testing gaps persist among AGYW, linked to risky sexual behavior (RSB). This study explores the association between these behaviors and HIV testing utilization among AGYW (aged 15-24) in Kibra Sub County, Nairobi.</p> <p><strong>Methods: </strong>A cross-sectional study sampled 379 AGYW from three wards in Kibra Sub County in Nairobi County. To be an eligible participant, one must have been a resident for at least one year before the time of the study and aged between 15-24 years, employing standardized structured interviewer-administered questionnaires and statistical analyses. Results were analyzed using Chi-square tests and logistic regression. Data was collected between June to July 2023.</p> <p><strong>Results: </strong>Overall, HIV testing prevalence was 60.7% (n=230). Those aged 20-24 were 71.3% (n=164), with secondary education were 63.5% (n=146) and married 28.7% (66) were more likely to undergo testing. Participants engaging in risky behaviors such as lack of condom use (3.96 times more likely), experiencing gender-based violence (4.65 times more likely), or contracting STIs (2.85 times more likely) had higher odds of seeking HIV testing services.</p> <p><strong>Conclusion: </strong>This study establishes a clear link between risky sexual behaviors and HIV testing among AGYW, with a 60.7% testing prevalence; however, gaps still exist. Efforts to enhance testing rates are vital. Interventions should align with acceptable methods, focusing on this high-risk group to ensure effective HIV care and prevention.</p>

Antigen-specific memory NK cell responses against HIV and influenza utilize the NKG2/HLA-E axis

<p>For over a decade, multiple studies have disputed the notion of natural killer (NK) cells as purely innate lymphocytes by demonstrating that they are capable of putative antigen-specific immunological memory against multiple infectious agents including two critical global health priorities – human immunodeficiency virus (HIV) and influenza. However, the mechanisms underlying antigen specificity remain unknown. Herein, we demonstrate that antigen-specific human NK cell memory develops upon exposure to both HIV and influenza, unified by a conserved and epitope-specific targetable mechanism largely dependent on the activating CD94/NKG2C receptor and its ligand HLA-E, and confirm these findings by three rigorous assays. We validated the permanent acquisition of antigen-specificity by individual memory NK cells by single-cell cloning. We identified biomarkers of antigen-specific NK cell memory through complex immunophenotyping by 30-parameter flow cytometry showing elevated expression of KLRG1, a4b7, and NKG2C. Finally, we show individual HLA-E-restricted peptides that may constitute the dominant NK cell response in HIV-1- and influenza-infected persons in vivo. Our findings clarify the mechanisms behind antigen-specific memory NK cell responses, and suggest they could be targeted for future vaccines, cure strategies, or other therapeutic interventions.</p>

Utility index and vision related quality of life in patients awaiting specialist eye care

<p>Objectives:</p> <p>This study aimed to ascertain utility and vision-related quality of life in patients awaiting access to specialist eye care. A secondary aim was to evaluate the association of utility indices with demographic profile and waiting time.</p> <p>Methods:</p> <p>Consecutive patients that had been waiting for ophthalmology care answered the 25-item National Eye Institute Visual Function Questionnaire (NEI VFQ-25). The questionnaire was administered when patients arrived at the clinics for their first visit. We derived a utility index (VFQ-UI) from the patients' responses, then calculated the correlation between this index and waiting time and compared utility across demographic subgroups stratified by age, sex, and care setting.</p> <p>Results:</p> <p>536 individuals participated in the study (mean age 52.9±16.6 years; 370 women, 69% women). The median utility index was 0.85 (interquartile range [IQR] 0.70–0.92; minimum 0.40, maximum 0.97). The mean VFQ-25 score was 70.88±14.59. Utility correlated weakly and nonsignificantly with waiting time (-0.05, <em>P </em>= 0.24). It did not vary across age groups (<em>P </em>= 0.85) or care settings (<em>P </em>= 0.77). Utility was significantly lower for women (0.84, IQR 0.70–0.92) than men (0.87, IQR 0.73–0.93, <em>P </em>= 0.03), but the magnitude of this difference was small (Cohen's d = 0.13).</p> <p>Conclusion:</p> <p>Patients awaiting access to ophthalmology care had a utility index of 0.85 on a scale of 0 to 1. This measurement was not previously reported in the literature. Utility measures can provide insight into patients' perspectives and support economic health analyses and inform health policies.</p>
