Data from: Stimulation-induced cytokine polyfunctionality as a dynamic concept
<p>Cytokine polyfunctionality is a well-established concept in immune cells, especially T cells, and their ability to concurrently produce multiple cytokines has been associated with better immunological disease control and subsequent effectiveness during infection and disease. To date, only little is known about the secretion dynamics of those cells, masked by the widespread deployment of mainly time-integrated endpoint measurement techniques that do not easily differentiate between concurrent and sequential secretion. Here, we employed a single-cell microfluidic platform capable of resolving secretion dynamics of individual PBMCs. To study the dynamics of poly-cytokine secretion, as well as the dynamics of concurrent and sequential polyfunctionality, we analyzed the response at different time points after <em>ex vivo </em>activation. Firstly, we observed simultaneous secretion of cytokines over the measurement time for most stimulants in a subpopulation of cells only. Secondly, polyfunctionality generally decreased with prolonged stimulation times and revealed no correlation with the concentration of secreted cytokines in response to stimulation. However, we observed a general trend towards higher cytokine secretion in polyfunctional cells, with their secretion dynamics being distinctly different from mono-cytokine secreting cells. This study provided insights into the distinct secretion behavior of heterogenous cell populations after stimulation with well-described agents and such a system could provide better understanding for various immune dynamics in therapy and disease.</p>

Mg(OH)2 Recovery from Real Bitterns: a Proof of Concept at Pilot Scale
<p>Water, energy, and minerals are fundamental pillars for the future of humankind. Sustainable and renewable productive processes and resources are the only possibility to face the continuously growing global population and high living standards requirements. The present work introduces a proof of concept for the pilot scale production of magnesium hydroxide, Mg(OH)<sub>2</sub>, from waste saltworks bitterns located in the district of Trapani, Italy. Mg(OH)<sub>2</sub> was produced by adopting a proprietary “Magnesium Crystals Granulometry Controlled Reactor”, Mg-CGCR. The influence of several parameters was investigated on Mg(OH)<sub>2</sub> suspensions and powders characteristics: (i) the bittern flow rate, (ii) alkaline sodium hydroxide, NaOH, concentrations solutions (adopted as the precipitant agent) and (iii) the final suspension pH value (stoichiometric or OH<sup>-</sup> excess amounts). A Mg<sup>2+</sup> recovery >99 % can be achieved thanks to the adoption of a product recycling strategy in the reactor. Furthermore, highly pure Mg(OH)<sub>2</sub> powders, addressed by cationic purity, were synthesized. Results demonstrate the possibility of producing highly pure Mg(OH)<sub>2</sub> products from waste-concentrated saline solutions, thus turning waste into valuable compounds.</p>

Medical Concept Normalization in Social Media Posts with Recurrent Neural Networks
<p>Text mining of scientific libraries and social media has already proven itself as a reliable tool for<br> drug repurposing and hypothesis generation. The task of mapping a disease mention to a concept<br> in a controlled vocabulary, typically to the standard thesaurus in the Unified Medical Language<br> System (UMLS), is known as medical concept normalization. This task is challenging due to the<br> differences in medical terminology between health care professionals and social media texts coming<br> from the lay public. To bridge this gap, we use sequence learning with recurrent neural networks<br> and semantic representation of one- or multi-word expressions: we develop end-to-end architectures<br> directly tailored to the task, including bidirectional Long Short-Term Memory and Gated Recurrent<br> Units with an attention mechanism and additional semantic similarity features based on UMLS.<br> Our evaluation over a standard benchmark shows that recurrent neural networks improve results<br> over an effective baseline for classification based on convolutional neural networks. A qualitative<br> examination of mentions discovered in a dataset of user reviews collected from popular online health<br> information platforms as well as quantitative evaluation both show improvements in the semantic<br> representation of health-related expressions in social media.</p>

Bringing the concept of ammonia critical levels into managing cork-oak woodland for conservation
<p>Agricultural activities often emit excessive nitrogen concentrations to nearby habitats, impacting ecosystems structure and functioning. To protect them, critical levels were established as the concentration above which direct adverse effects on ecosystems may arise. However, due to lack of tools, critical levels are seldom applied in management strategies for conservation. Our objective was to provide a spatial-explicit tool to incorporate the concept of ammonia critical levels in management practices of atmospheric nitrogen impacts. To do that, lichens diversity was sampled in 19 plots at a cork oak woodland (Montado), a High Nature Value Farmland where multiple agricultural activities, surrounded by high-intensity agricultural activities, coexist. We inferred and selected the best performing trait-based metrics and determined the main environmental drivers of change. The most responsive metric was the abundance of oligotrophic lichen species, which was likely responding to nitrogen incoming from nearby high-intensity agricultural areas. The low intensity agricultural activities, cattle grazing within the study area, had no effect. Using the known ammonia critical levels for that ecosystem, we modelled and mapped the long-term critical levels exceedance for the entire study area. This allowed us to signal high intensity agriculture as the source of impact and to map the areas where management practices need further attention under a conservation perspective: either decreasing fertilization in nearby fields or by considering these as buffer areas, outside the scope of conservation.</p>

Ivermectin as an adjuvant to anti-epileptic treatment in persons with onchocerciasis-associated epilepsy: a randomized proof-of-concept clinical trial
<p>Open dataset for a proof-of-concept randomized clinical trial with a four month follow-up period was conducted to investigate whether ivermectin had an added value in decreasing the frequency of seizures in onchocerciasis-infected persons with epilepsy (PWE). The 90 PWE with epilepsy were randomly allocated to the treatment arms: phenobarbital + ivermectin (n = 44) or phenobarbital alone (n = 46). Two weeks after randomization, a follow-up visit was scheduled to assess the early treatment outcomes and potential side effects of AED. After the second week visit, PWE had to report to the health center for further follow-up consultations by the project nurses and medical doctors, scheduled at 1, 2, 3 and 4 months. By March 2018, 89 (99%) participants in both arms attended the final follow-up visit. The trial was conducted in five onchocerciasis-endemic villages within the Logo health zone in the Ituri province, DRC: Draju, Kanga, Wala, Ulyeko and Thedeja. </p> <p>At baseline, information was collected on seizure semiology, seizure frequency, epilepsy risk factors, relevant medical history, previous AED and ivermectin use. Weight and height measurements were carried out on all participants, followed by a complete physical and neurological examination performed by one of the medical doctors who had been trained in epilepsy, or by the neurologist. Cognitive impairment was assessed by determining whether the participant was well oriented in time and place, whether he/she could remember his/her name, was coherent in speech, and was obedient to orders; abnormal behaviour such as unexplained aggressive attitudes and/or wandering episodes were also noted.</p> <p> </p> <p> </p>
