Early onset of adult deafness in the Rhodesian Ridgeback is associated with in-frame deletion in the EPS8L2 gene

<p>Domestic dogs exhibit diverse types of both congenital and non-congenital hearing losses. Rhodesian Ridgebacks can suffer from a progressive hearing loss in the early stage of their life, a condition known as early onset of adult deafness (EOAD), where they lose their hearing ability within 1-2 years after birth. In order to investigate the genetic basis of this hereditary hearing disorders, we performed a genome-wide association study (GWAS) by using a sample of 23 affected and 162 control Rhodesian Ridgebacks. We identified a genomic region on canine chromosome 18 (CFA18) that is strongly associated with EOAD, and our subsequent targeted Sanger sequencing analysis identified a 12-bp inframe deletion in EPS8L2 (CFA18:25,868,739-25,868,751 in the UMICH_Zoey_3.1/canFam5 reference genome build). Additional genotyping confirmed a strong association between the 12-bp deletion and EOAD, where all affected dogs were homozygous for the deletion, while none of the control dogs was a deletion homozygote. A segregation pattern of this deletion in a 2-generation nuclear family indicated an autosomal recessive mode of inheritance. Since EPS8L2 plays a critical role in the maintenance and integrity of the inner ear hair cells in humans and other mammals, the inframe deletion found in this study represents a strong candidate causal mutation for EOAD in Rhodesian Ridgebacks. Genetic and clinical similarities between childhood deafness in humans and EOAD in Rhodesian Ridgebacks emphasizes the potential value of this dog breed in translational research in hereditary hearing disorders.</p>

Outputs of Numerical experiments verifying the diverse interannual variability of Asian summer monsoon onset process

<p><strong>Introduction</strong></p> <p>We designed two sets of numerical experiments using the Community Earth System Model (CESM 1.2.2) released by NCAR to verify the SSTAs&#39; effect on the interannual modes of ASMOP in April and May. In the atmospheric general circulation model (AGCM) experiments, we solely forced the Community Atmosphere Model version 5.3 (CAM5.3), the atmospheric component of the CESM 1.2.2, by the specific SSTAs in April&ndash;May. The AGCM control run included a 40-yr integration forced by the climatological sea surface temperature (SST) with the annual cycle. We attached the specific SSTAs to the climatological SST in each sensitivity experiment and integrated them for 40 years. The ensemble results in the last 30 years were used for analysis. By contrast, in the PACEMAKER experiments, we first integrated the CESM 1.2.2 for 200 years as a control run to exclude the evident climate shift. Afterward, the sensitivity experiments were conducted on the last 100-yr outputs of the control run, in which we nudged the April&ndash;May SSTAs in the specific domain but left the rest of the model&#39;s coupled climate system free to evolve. The last 30 members were ensembled for analysis in each PACEMAKER experiment.&nbsp;</p> <p><strong>Numerical Experiment Design</strong></p> <p>In the AGCM category, we forced CAM 5.3 by warm SSTAs in the southwestern Indian Ocean (20&deg;S-0&deg;, 40&deg;-80&deg;E) in the ASMOP-PC2 experiment but by cold SSTAs in the western North Pacific (0&deg;-20&deg;N, 120&deg;-160&deg;E) in the ASMOP-PC3 experiment&nbsp;from April to May, respectively. Similar SSTAs were nudged in the two oceanic domains from April to May in the PACEMAKER category.</p> <p><strong>File Description</strong></p> <ol> <li>Output files were saved in the NetCDF format.</li> <li>The &quot;AGCM.tar.gz&quot; and &quot;PACEMAKER.tar.gz&quot; was the compressed files of the atmospheric outputs in May in the AGCM and PACEMAKER experiments, respectively.</li> <li>In each compressed file, the results of the control run (in the subfolder named &quot;Control_Run&quot;), ASMOP-PC2 (in the subfolder named &quot;ASMOP-PC2&quot;), and ASMOP-PC3 (in the subfolder named &quot;ASMOP-PC3&quot;) sensitivity experiments&nbsp;were included for comparison.</li> </ol>

Intermediate-onset colorectal cancer: a clinical and familial boundary between both early and late-onset colorectal cancer

<p>Comparative studies of colorectal cancer (CRC) according to the age of onset<br> have found differences between early-onset CRC (EOCRC) and late-onset CRC<br> (LOCRC). Using this as a starting point, we wished to determine whether intermediateonset<br> CRC (IOCRC) might also be considered as an independent group within CRC.<br> We performed a retrospective comparative study of the clinicopathological and familial<br> features, as well as of the symptoms and their duration, of a total of 272 subjects<br> diagnosed with CRC classified into three groups according to the age-of-onset (98<br> EOCRC, 83 IOCRC and 91 LOCRC). The results show that from a clinicopathological<br> point of view, IOCRC shared certain features with EOCRC (gender, prognosis), and<br> with LOCRC (multiple primary CRCs), whereas it also had characteristics that were<br> specific for IOCRC (mean number of associated polyps). A gradual progression was<br> observed from EOCRC to LOCRC from a greater family aggregation to sporadic cases,<br> in parallel with a change of Lynch Syndrome cases to the sporadic microsatellite<br> instability pathway, with the IOCRC being a boundary group that is more related to<br> EOCRC. With respect to symptoms, duration and correlation with stages, IOCRC<br> appeared more similar to EOCRC. Clinically, IOCRC behaves as a transitional group<br> between EOCRC and LOCRC, with features in common with both groups, but also<br> with IOCRC-specific features. Excluding cases with familial cancer history, the<br> awareness for EOCRC diagnosis should be extended to IOCRC.</p>

Monolithic FAPbBr3 Photoanode for Photoelectrochemical Water Oxidation with Low Onset-potential and Enhanced Stability

<p>Despite considerable research efforts on photoelectrochemical water splitting over the past decades, practical application faces challenges by the absence of efficient, stable, and scalable photoelectrodes. Herein, we report a metal-halide perovskite-based photoanode for photoelectrochemical water oxidation. With a planar structure using mesoporous carbon as a hole-conducting layer, the precious metal-free FAPbBr<sub>3</sub> photovoltaic device achieves 9.2% solar-to-electrical power conversion efficiency and 1.4 V open-circuit voltage. The photovoltaic architecture successfully applies to build a monolithic photoanode with the FAPbBr<sub>3</sub> absorber, carbon/graphite conductive protection layers, and NiFe catalyst layers for water oxidation. The photoanode delivers ultralow onset-potential below 0 V versus the reversible hydrogen electrode and high applied-bias photon-to-current efficiency of 8.5%. Stable operation exceeding 100&thinsp;h under solar illumination by applying ultraviolet-filter protection. The photothermal investigation verifies the performance boost in perovskite photoanode by photothermal effect. This study is significant in guiding the development of photovoltaic material-based photoelectrodes for solar fuel applications.</p>
