Serum response factor utilizes distinct promoter- and enhancer-based mechanisms to regulate cytoskeletal gene expression in macrophages.
<p>Cells of the monocyte/macrophage lineage play essential roles in tissue homeostasis and immune responses, but mechanisms underlying the coordinated expression of cytoskeletal genes required for specialized functions of these cells, such as directed migration and phagocytosis, remain unknown. Here, using genetic and genomic approaches, we provide evidence that serum response factor (SRF) regulates both general and cell type-restricted components of the cytoskeletal gene expression program in macrophages. Genome-wide location analysis of SRF in macrophages demonstrates enrichment of SRF binding at ubiquitously expressed target gene promoters, as expected, but also reveals that the majority of SRF binding sites associated with cell type-restricted target genes are at distal inter- and intragenic locations. Most of these distal SRF binding sites are established by the prior binding of the macrophage- and the B cell-specific transcription factor PU.1 and exhibit histone modifications characteristic of enhancers. Consistent with this, representative cytoskeletal target genes associated with these elements require both SRF and PU.1 for full expression. These findings suggest that SRF uses two distinct molecular strategies to regulate programs of cytoskeletal gene expression: a promoter-based strategy for ubiquitously expressed target genes and an enhancer-based strategy at target genes that exhibit cell type-restricted patterns of expression.</p>

<p><span>This study used a quantitative research design to examine the impact of ChatGPT utilization on student laziness and productivity. Quantitative research allows the collection of numerical data and facilitates statistical analysis to test the proposed hypothesis and draw empirical conclusions. Data was collected through an online survey distributed to undergraduate students from various universities. The survey included questions about ChatGPT usage patterns, perceived laziness (measured through procrastination and task avoidance scales), and self-reported productivity metrics.</span></p>

Utilization of high-performance concrete mixtures for advanced manufacturing technologies
<p><span>The presented experimental program focuses on the design of high-performance dry concrete mixtures, which could find application in advanced manufacturing technologies, for example additive solutions. The combination of high-performance concrete (HPC) with advanced or additive technologies provides new possibilities for constructing architecturally attractive buildings with high material requirements. The purpose of this study was to develop a dry mixture made from high-performance concrete that could be distributed directly in a advanced or additive technologies of solutions in the pre-prepared condition with all input materials (except for water) in order to reduce both financial and labor costs. This research specifically aimed to improve the basic strength characteristics, including mechanical (assessed using compressive strength, tensile splitting strength, and flexural strength tests) and durability properties (assessed using tests of resistance to frost, water, and defrosting chemicals), of hardened mixtures, with partial insight into the rheology of fresh mixtures (consistency as assessed using the slump-flow test). Additionally, the load-bearing capacity of the selected mixtures in the form of specimens with concrete reinforcement were tested using a three-point bending test. A reference mixture with two liquid plasticizers—the first based on polycarboxylate and polyphosphonate and the second based on polyether carboxylate—was modified using a powdered plasticizer, based on the polymerization product Glycol, to create a dry mixture; the reference mixture was compared with the developed mixtures with respect to the above-mentioned properties. In general, the results show that the replacement of the aforementioned liquid plasticizers by a powdered plasticizer based on the polymerization product Glycol in the given mixtures is effective up to 5 % (of the cement content) with regard to the mechanical and durability properties. The presented work provides an over-view of the compared characteristics, which will serve as a basis for future research into the development of additive manufacturing technologies in the conditions of the Czech Republic while respecting the principles of sustainable construction.</span></p>
Utilizing Digital Information for Personalized Cholesterol Management
<p><span>Patients with diseases due to high cholesterol levels in Indonesia can be said to be high because, in 2022, sufferers of the disease reached 28%, with 7.9% of people dying. The problem in this study is the need for more understanding, awareness, and ability of the community to manage and maintain cholesterol through digital information, especially social media. This research uses a questionnaire to understand the community's needs and supporting factors in digitally managing cholesterol through social media. This research gathered data using purposive sampling through a Google Form questionnaire. The data collection was conducted in April 2024 in various cities in Indonesia and successfully collected 221 responses. The method used to analyze the data in this study is the Structural Equation Model (SEM) using Smart Partial Least Square (Smart-PLS) 4.0 as the analysis approach. In this study, there are ten variables with five indicators each, and there are 11 hypotheses. The outcomes of this study show that all of the indicators were valid and reliable, with all hypotheses having a significant effect. Indonesian people must be able to utilize digital information to increase knowledge and understanding about health, especially about Cholesterol Management, so that the number of people affected by disease and who die from cholesterol can be reduced.</span></p>

On the utilization of Disruptive Technologies for Municipality-wide Policy Making
<p>Disruptive technologies (DTs) aim to introduce innovative elements with the immediate goal of changing citizens' behavior and the optimization of certain processes in either the private or public sector. The sector, especially "e-governance" has adopted several DT-based systems and is open to exploring new technologies that could be potentially useful for the sector's administration. This paper proposes a DT-based framework for the early detection of misinformation, the identification and prioritization of emerging and significant issues that refer to the municipality, and the recommendation of counter-actions that could be applied for each identified issue. The proposed framework aims to be applied initially at the municipal level as this is a more limited community for the framework's study and evaluation. News feeds and information posted on social media platforms will be the key information included in the proposed framework, aiming at sensing the public opinion and extracting issues and topics that concern a municipality's citizens. Through social media platforms, users tend to post, among other, concerns or issues they face within the local community, and thus useful content can be extracted and used for the identification of emerging and significant issues. The main advantage of this framework is the utilization of information from different data sources and their processing in order to immediately inform the relevant authority about the most important issues, allowing them to act in a proactive way. The expected outcome of the proposed framework is to raise the citizens' awareness of the issues of the local society and restrict criminality and violence in the municipality.</p>

Space resource utilization of dominant species integrates abundance- and functional-based processes for better predictions of plant diversity dynamics
<p>Sustainable ecosystem management relies on our ability to predict changes in plant diversity and to understand the underlying mechanisms. Empirical evidence demonstrates that abundance- and functional-based processes simultaneously explain the loss of plant diversity in response to human activities. Recently, a novel indicator based on percent cover (CoverD) and maximum height (HeightD) of the dominant plant species – Space Resource Utilization (SRUD) – has proven to give robust and better predictions of plant diversity dynamics than community biomass. Whether the superior predictive ability of SRUD is due to its capacity to simultaneously capture abundance- and functional-based processes remains unknown. Here, we tested this hypothesis by quantifying mechanistic links between changes in SRUD and biodiversity in response to nutrients and herbivores. Furthermore, we assessed the relative contribution of dominant, intermediate, and rare species to reduced density of individuals by combining null model analysis with field experiments. We found that SRUD successfully captured changes in ground-level light availability and changes in the number of individuals to predict plant diversity dynamics, and each of CoverD and HeightD partly and independently contributed to both processes. Comparative results from null model analysis and field experiments confirmed that individual losses of dominant, intermediate, and rare species followed non-random processes. Specifically, compared with random loss process, rare species lost proportionally more individuals and thus disproportionately contributed to species loss, while dominant and intermediate species lost less. Our results demonstrate that SRUD captures both abundance- and functional-based processes thus explaining why SRUD provides more accurate predictions of changes in species diversity. Given that rare species can play an important role in shaping community structure, resisting against invasion, impacting higher trophic levels, and providing multiple ecosystem functions, reducing the SRU of dominant species could alleviate the risk of exclusion of rare species by mitigating abundance- and functional-based competition processes.</p>
