NGFS Climate Scenarios Data Set

<h1><strong>Notice to users of NGFS long-term climate scenarios</strong></h1> <p>The NGFS informs users that the academic paper underpinning the physical risk estimates in Phase V of its long-term scenarios, Kotz et al. (2024), has received critiques in a post-publication review at <em>Nature</em>. The authors have revised their analysis, with limited impacts on results. The updated paper still has to undergo peer-review.</p> <p>The NGFS closely monitors the academic process and will incorporate any necessary updates in future iterations of its long-term scenarios.</p> <p>Users are reminded that neither the NGFS, nor its member institutions, nor any person acting on their behalf, is responsible or liable for any reliance on, or for any use of the NGFS scenarios and/or supplementary documentation. Data set of article : Using hypnosis in competition period to enhance self-confidence and to reduce anxiety among volleyball players

<p>Abstract: The aim of our research was to test the influence of hypnosis on the evolution of self-confidence and anxiety. Our sample included (N = 7 volleyball players) of a Tunisian Volleyball Team, aged 16 to 24, were followed daily for four weeks to enjoy their training and followed a program practicing the use of hypnosis. To assess the evolution of the psychological variables, participants completed the following questionnaire based on three measurement times (at the beginning of the experiment, after two weeks and at the end of the four weeks): The Competitive State Anxiety&nbsp;Inventory-2 (CSAI-2). The questionnaire (CSAI-2) was obtained statistically significant from the three times analysis of variance (Alpha = .78). A one-way model ANOVA revealed that hypnosis improved for the athletes (p &lt; .01); self-confidence increased (p &lt; .01) and cognitive anxiety decreased (p &lt; .05). Correlation analysis revealed that changes in hypnosis were moderately related to changes in self-confidence (p &lt; .05).The results of the study showed that hypnosis can enhance self-confidence and reduce cognitive anxiety. These observations allow us to guide future interventions with athletes to strengthen them psychologically.</p> <p>Key words: hypnosis, self-confident, anxiety&hellip;</p>

Data set related to the article: "Gender Differences Associated with the Prognostic Value of BPIFB4 in COVID-19 Patients: A Single-Center Preliminary Study."

<p><strong>Abstract:</strong> In the ongoing global COVID-19 pandemic, male sex is a risk factor for severe disease and death, and the reasons for these clinical discrepancies are largely unknown. The aim of this work is to study the influence of sex on the course of infection and the differences in prognostic markers between genders in COVID-19 patients. Our cohort consisted of 64 adult patients (n = 34 men and n = 30 women) with PCR-proven SARS-CoV-2 infection. Further, a group of patients was characterized by a different severity degree (n = 8 high- and n = 8 low-grade individuals for both male and female patients). As expected, the serum concentrations of LDH, fibrinogen, CRP, and leucocyte count in men were significantly higher than in females. When serum concentrations of the inflammatory cytokines, including IL-6, IL-2, IP-10 and IL-4 and chemokines like MCP-1, were measured with multiplex ELISA, no significant differences between male and female patients were found. In COVID-19 patients, we recently attributed a new prognostic value to BPIFB4, a natural defensin against dysregulation of the immune responses. Here, we clarify that BPIFB4 is inversely related to the disease degree in men but not in women. Indeed, higher levels of BPIFB4 characterized low-grade male patients compared to high-grade ones. On the contrary, no significant difference was reported between low-grade female patients and high-grade ones. In conclusion, the identification of BPIFB4 as a biomarker of mild/moderate disease and its sex-specific activity would open an interesting field for research to underpin gender-related susceptibility to the disease.</p>

<p>As we age, our body experiences chronic, systemic inflammation contributing to the morbidity and mortality of the elderly. The senescent immune system has been described to have a causal role in driving systemic aging and therefore may represent a key therapeutic target to prevent pathological consequences associated with aging and extend a healthy lifespan. Previous studies from our group associated a polymorphic haplotype variant in the BPIFB4 gene (LAV-BPIFB4) with exceptional longevity. Transfer of the LAV-BPIFB4 in preclinical models halted the progression of cardiovascular diseases (CVDs) and frailty by counterbalancing chronic inflammation. In the present study, we aimed to delineate the action of systemic adeno-associated viral vector-mediated LAV-BPIFB4 gene transfer (AAV-LAV-BPIFB4) on the deleterious age-related changes of the immune system and thereby the senescence-associated events occurring in C57BL/6J mice aged 26 months. Our in vivo data showed that 26-months-old mice had a higher frequency of CD45+SA-beta Gal+ immune cells in peripheral blood than young (4-months-old) C57BL/6J mice. Notably, AAVLAV-BPIFB4 gene transfer in aged mice reduced the pool of peripheral immunosenescent cells that were shown to be enriched in the spleen. In addition, the proper tuning of the immune secretory phenotype (IL1&beta;low, IL6low, IL10high) associated with a significant reduction in SA-beta Gal-positive area of aorta from AAV-LAV treated mice. At the functional level, the reduction of senescenceassociated inflammation ensured sustained NAD+ levels in the plasma of AAV-LAV-BPIFB4 old mice by preventing the NADase CD38 increase in F4/80+ tissue-resident macrophages and Ly6Chigh pro-inflammatory monocytes of the spleen and bone marrow. Finally, to validate the clinical implication of our findings, we showed that Long-living-individuals (LLIs, &gt;95 years), which delay CVDs onset, especially if LAV-carriers, were characterized by high NAD+ levels. In conclusion, the new senotherapeutic action of LAV-BPIFB4 may offer a valuable therapeutic tool to control aging and reduce the burden of its pathophysiological disorders, such as CVDs.</p>

Data-set of CO2, CH4, N2O dissolved concentrations and ancillary data in Lake Victoria (March 2018, October 2018, and June 2019)

<p>The data-base contains 4 xlsx files:</p> <ul> <li>CTD_Lavigas_123 contains vertical profiles of CTD data (water temperature, specific conductivity, pH, oxygen saturation level (%O2)), partial pressure of CO2 measured with headspace equilibration with a Licor, CH4 and N2O measured with headspace equilibration by gas chromatography on samples collected with a Niskin bottle, obtained in March-April 2018, October-November 2018, and June 2019.</li> <li>UW_Lavigas_1 contains the continuous measurements in surface waters of partial pressure of CO2 and CH4 measured by equilibration with a Los Gatos Research portable analyzer, plus CTD data and bottom depth, obtained in March-April 2018.</li> <li>UW_Lavigas_2 contains the continuous measurements in surface waters of partial pressure of CO2 and CH4 measured by equilibration with a Los Gatos Research portable analyzer, plus CTD data and bottom depth, obtained in October-November 2018.</li> <li>UW_Lavigas_3 contains the continuous measurements in surface waters of partial pressure of CO2 and CH4 measured by equilibration with a Los Gatos Research portable analyzer, plus CTD data and bottom depth, obtained in June 2019.</li> </ul> <p>For vertical profiles (CTD_Lavigas_123) water was collected with a Niskin bottle and two 60 mL borosilicate serum bottles (Wheaton) for the determination of CH4 and N2O were filled with a silicone tubing, allowed to overflow, poisoned with a saturated solution of HgCl2 (100 &micro;L), sealed with butyl stoppers, crimped with aluminum caps, and stored at ambient temperature in the dark. Samples for the determination of pCO2 were collected from the Niskin bottle in 4 polypropylene 60 mL syringes where 30 mL of sample water were equilibrated with 30 mL of ambient air (5 min of vigorous manual shaking). The pCO2 in ambient air and in the equilibrated gas phase were determined with a Li-840 calibrated with N2 and a suite of CO2:N2 commercial mixtures (Air Liquide Belgium) with mixing ratios of 388, 813, 3788, and 8300 ppm CO2. The overall precision of pCO2 measurements was &plusmn;2.0%. Concentrations of CH4 and N2O were determined via the headspace equilibration technique (20 mL N2 headspace in 60 mL serum bottles) and measured by GC with flame ionization detection (GC-FID) and electron capture detection (GC-ECD) with a SRI 8610C GC-FID-ECD calibrated with CH4:CO2:N2O:N2 mixtures (Air Liquide Belgium) of 1, 10, 30, 509 and 2011 ppm CH4 and of 0.2, 2.0 and 6.0 ppm N2O, and using the solubility coefficients of CH4 (Yamamoto et al. 1976) and N2O (Weiss &amp; Price 1980). The precision of measurements was &plusmn;3.9% for CH4 and &plusmn;3.2% for N2O.</p> <p>In surface waters, pCO2 and the partial pressure of CH4 (pCH4) were measured with a Los Gatos Research (LGR) off-axis integrated cavity output spectroscopy analyzer (Ultraportable Greenhouse Gas Analyzer with extended range for CH4) coupled to an equilibrator (Frankignoulle et al. 2001). The reading of pCO2 and pCH4 from the LGR was compared in the home laboratory to the values from CH4:CO2:N2 mixtures (Air Liquide Belgium) of 1, 10, 30, 509 and 2011 ppm CH4 and 431, 1088, 4,385 and 22,029 ppm CO2. The instrument did not show detectable drift between the start and the end of the fieldwork, and the factory calibration gave satisfactory readings against standards.</p> <p>Both approaches to measure pCO2 and CH4 show excellent agreement (Borges et al. 2022). Water temperature, conductivity, pH, and dissolved O2 were measured with Yellow Spring Instrument (YSI) multiprobe (YSI EXO-2). <p>Abstract:</p> <p>SARS-CoV-2 infection causes a considerable inflammatory response coupled with impaired platelet reactivity, which can lead to platelet disorders recognized as negative prognostic factors in COVID-19 patients. The virus may cause thrombocytopenia or thrombocytosis during the different disease stages by destroying or activating platelets and influencing platelet production. While it is known that several viruses can impair megakaryopoiesis by generating an improper production and activation of platelets, the potential involvement of SARS-CoV-2 in affecting megakaryopoiesis is poorly understood. To this purpose, we explored, in vitro, the impact of SARS-CoV-2 stimulation in the MEG-01 cell line, a human megakaryoblastic leukemia cell line, considering its spontaneous capacity of releasing platelet-like particles (PLPs). We interrogated the effect of heat-inactivated SARS- CoV-2 lysate in the release of PLPs and activation from MEG-01, the signaling pathway influenced by SARS-CoV-2, and the functional effect on macrophagic skewing. The results highlight the potential influence of SARS-CoV-2 in the early stages of megakaryopoiesis by enhancing the production and activation of platelets, very likely due to the impairment of STATs signaling and AMPK activity. Overall, these findings provide new insight into the role of SARS-CoV-2 in affecting megakaryocyte&ndash; platelet compartment, possibly unlocking another avenue by which SARS-CoV-2 moves.</p>

We have previously shown that carriers of a longevity-associated variant (LAV) of the BPIFB4 gene enjoy prolonged health</p> <p>spans and lesser cardiovascular complications. Moreover, supplementation of LAV-BPIFB4 via an adeno-associated viral vector im-</p> <p>proves cardiovascular performance in limb ischaemia, atherosclerosis, and diabetes models. Here, we asked whether the LAV-</p> <p>BPIFB4 gene could address the unmet therapeutic need to delay the heart&rsquo;s spontaneous ageing.&nbsp;</p> <p><strong>Methods and results</strong></p> <p>Immunohistological studies showed a remarkable reduction in vessel coverage by pericytes in failing hearts explanted from elderly patients. This defect was attenuated in patients carrying the homozygous LAV-BPIFB4 genotype. Moreover, pericytes isolated from older hearts showed low levels of BPIFB4, depressed pro-angiogenic activity, and loss of ribosome biogenesis. LAV-BPIFB4 supple- mentation restored pericyte function and pericyte-endothelial cell interactions through a mechanism involving the nucleolar protein nucleolin. Conversely, BPIFB4 silencing in normal pericytes mimed the heart failure pericytes. Finally, gene therapy with LAV-BPIFB4 prevented cardiac deterioration in middle-aged mice and rescued cardiac function and myocardial perfusion in older</p> <p>mice by improving microvasculature density and pericyte coverage.</p> <p><strong>Conclusions</strong></p> <p>We report the success of the LAV-BPIFB4 gene/protein in improving homeostatic processes in the heart&rsquo;s ageing. These findings open to using LAV-BPIFB4 to reverse the decline of heart performance in older people.</p>

<p>Abstract: Glioblastoma (GBM) is the most common primary brain cancer with the median age at diagnosis around 64 years, thus pointing to aging as an important risk factor. Indeed, aging, by increasing the senescence burden, is configured as a negative prognostic factor for GBM stage. Furthermore, several anti-GBM therapies exist, such as temozolomide (TMZ) and etoposide (ETP), that unfortunately trigger senescence and the secretion of proinflammatory senescence-associated secretory phenotype (SASP) factors that are responsible for the improper burst of (i) tumorigenesis, (ii) cancer metastasis, (iii) immunosuppression, and (iv) tissue dysfunction. Thus, adjuvant therapies that limit senescence are urgently needed. The longevity-associated variant (LAV) of the bactericidal/permeability-increasing fold-containing family B member 4 (BPIFB4) gene previously demonstrated a modulatory activity in restoring age-related immune dysfunction and in balancing the low-grade inflammatory status of elderly people. Based on the above findings, we tested LAV- BPIFB4 senotherapeutic effects on senescent glioblastoma U87-MG cells and on T cells from GBM patients. We interrogated SA-&beta;-gal and HLA-E senescence markers, SASP factors, and proliferation and apoptosis assays. The results highlighted a LAV-BPIFB4 remodeling of the senescent phenotype of GBM cells, enhancement of their sensitivity to temozolomide and a selective reduction of the T cells&rsquo; senescence from GBM patients. Overall, these findings candidate LAV-BPIFB4 as an adjuvant therapy for GBM.</p>

<p>These data-sets contain information relative to the Paper entitled&nbsp; &ldquo;Light and Perception: Study of the Effects of Daylight and Artificial Light on Affect, Mood and Sleepiness under a Sky-Lighting Machine&rdquo;</p> <p>&nbsp;</p> <p>Data-set A contains Affect, Sleepiness and Mood data divided by participant ID (N = 28) and session number (N=9), summarised by Task (Reading and Game) and Light Condition (Daylight DL; Artificial Light AL). We coded AL as 0 and DL as 1; Reading as 0 and Game as 1.&nbsp;</p> <p>&nbsp;</p> <p>Data-set A includes as factors Gender, Order and Time of Day. Plus these subjective parameters, used as&nbsp; predictors in the models: perceived Color of Light and perceived Level of Light (Brightness).&nbsp;</p> <p>&nbsp;</p> <p>These photometric values were included as predictors in the model: Mean Illuminance, referred to as Room Illuminance; Mean Correlated Color Temperature (CCT); Variation, defined as the Standard Deviation (SD) of the Illuminance values; mEDI means calculated with CIE S 026/E:2018 on the spectral values of the two median Illuminance measurements taken over the time-span of thirty minutes; Illuminance measured by the actigraphs, referred to as Personal Illuminance.
