Data from: Different genetic basis for ADH activity and plasticity in a novel alcohol environment for Drosophila melanogaster
Phenotypic plasticity is known to enhance population persistence (Wang and Althoff, 2019), facilitate adaptive evolution (Levis et al., 2018), and initiate novel phenotypes in novel environments (Levis and Pfennig, 2016). How plasticity can contribute or hinder adaptation to different environments hinges on its genetic architecture. Even though plasticity in many traits is genetically controlled, whether and how plasticity's genetic architecture might change in novel environments is still unclear. Because much of gene expression can be environmentally influenced, each environment may trigger different sets of genes that influence a trait. Using a quantitative trait loci (QTL) approach, we investigated the genetic basis of plasticity in a classic functional trait, alcohol dehydrogenase (ADH) activity in D. melanogaster, across both historical and novel alcohol environments. Previous research in D. melanogaster has also demonstrated that ADH activity is plastic in response to alcohol concentration in substrates used by both adult flies and larvae. We found that across all environments tested, ADH activity was largely influenced by a single QTL encompassing the Adh coding gene and its known regulatory locus, delta-1. After controlling for the allelic variation of the Adh and delta-1 loci, we found additional but different minor QTLs in the 0% and 14% alcohol environments. In contrast, we discovered no major QTL for plasticity itself, including the Adh locus, regardless of the environmental gradients. This suggests that plasticity in ADH activity is likely influenced by many loci with small effects and that the Adh locus is not environmentally sensitive to dietary alcohol.

Preconception alcohol exposure increases the susceptibility to diabetes in the offspring
<p>Preconception alcohol exposure in mothers has recently been shown to increase stress responses and anxiety behaviors in their offspring during the adult period. One hypothesis is that alcohol-induced genetic modifications in germ cells of the mother could have been transmitted to the offspring to manifest the endophenotypes. In this study, transcriptome analysis of germ cells of female rats given binge-like alcohol identified altered stress gene regulation networks involving glucose metabolism and diabetes mellitus. Endocrine tests involving oral glucose tolerance, glucose-stimulated insulin secretion, and insulin tolerance in the male and female offspring of mothers given binge-like alcohol during the preconception period showed significant hyperglycemia and hypoinsulinemia. These offspring also showed increased levels of inflammatory cytokines and cellular apoptosis in the pancreas and altered insulin production and action on glucose metabolism, particularly insulin signaling molecules, in the liver. These animals, when subjected to a high-fat diet and streptozotocin injection for induction of type 2 diabetes, showed a higher response than control offspring. Preconception alcohol exposed offspring also showed reduced number of stress and glucose regulatory proopiomelanocortin (POMC) producing neurons in the hypothalamus, Replenishment of POMC neurons in the hypothalamus of these animals normalized glucose homeostasis and reduced the susceptibility to diabetes. These data suggest that preconception alcohol exposures alter glucose homeostasis and increase the susceptibility to diabetes by inducing POMC neuronal functional abnormalities, increasing pancreatic inflammation, and consequently lowering insulin production and action.</p>
Data from: Predictors of alcohol consumption among in-school adolescents in the Central Region of Ghana: a baseline information for developing cognitive-behavioural interventions
Background and purpose: Despite a recent shift in school going adolescents' engagement in health compromising behaviours has serious socio-economic implications on developing societies, it is surprising that baseline information through research that would influence planned interventions is sparse. The purpose of this study was to investigate the prevalence of alcohol drinking behaviours among in-school adolescents in the Junior High Schools (JHS) in the Central Region of Ghana. Methods and results: Using a descriptive cross-sectional design, multistage sampling procedures were used to sample 1400 school going adolescents in JHS in the Central Region. Preliminary results using simple frequencies and percentages revealed 42% alcohol drinking prevalence in the region. High prevalence of drunkenness (73%, n = 405) and early exposure to alcohol drinking when students were in primary school (52%, n = 286) were noted. Community festivals and use of alcohol as a form of medicine were enabling factors of alcohol consumption in the region. Binary logistic regression analysis also showed that geographical location was a significant predictor of alcohol drinking among school going adolescents, with students in the southern and central part of the region at greater risks of drinking alcohol than those from the northern part (OR = .696, 95% CI = 0.52-926, p = .013). However, no statistical significant variations were found in the odds of drinking alcohol within age (OR = 1.13, 95% CI = 0.86-1.48, p = .370), gender (OR = .81, 95% CI = 0.65-1.01, p = .06), religious affiliation (OR = 1.33, 95% CI = 0.94-1.89, p = .10), parental communication (OR = .86, 95% CI = 0.66-1.06, p = .13), academic performance (OR = 1.07, 95% CI = 0.79-1.45, p = .05) and socioeconomic status (OR = 1.20, 95% CI = 0.95-1.53, p = .12). Conclusions: With this baseline data, it was recommended that schools' curricula should include preventive cognitive-behavioural interventions that teach drug resistance skills and anti-drug norms. These interventions would foster the development of requisite knowledge and social skills (e.g., developing competence) for resisting social and peer influences that may trigger alcohol use and perhaps other drugs. Potentially, the motivation for alcohol use among school going adolescents in the region would be minimized, if not prevented.
Data from: 8-year trends in physical activity, nutrition, TV viewing time, smoking, alcohol and BMI: a comparison of younger and older Queensland adults
Lifestyle behaviours significantly contribute to high levels of chronic disease in older adults. The aims of the study were to compare the prevalence and the prevalence trends of health behaviours (physical activity, fruit and vegetable consumption, fast food consumption, TV viewing, smoking and alcohol consumption), BMI and a summary health behaviour indicator score in older (65+ years) versus younger adults (18-65 years). The self-report outcomes were assessed through the Queensland Social Survey annually between 2007-2014 (n=12,552). Regression analyses were conducted to compare the proportion of older versus younger adults engaging in health behaviours and of healthy weight in all years combined and examine trends in the proportion of younger and older adults engaging in health behaviours and of healthy weight over time. Older adults were more likely to meet recommended intakes of fruit and vegetable (OR=1.43, 95%CI=1.23-1.67), not consume fast food (OR=2.54, 95%CI=2.25-2.86) and be non-smokers (OR=3.02, 95%CI=2.53-3.60) in comparison to younger adults. Conversely, older adults were less likely to meet the physical activity recommendations (OR=0.86, 95%CI= 0.78-0.95) and watch less than 14 hours of TV per week (OR=0.65, 95%CI=0.58-0.74). Overall, older adults were more likely to report engaging in 3, or at least 4 out of 5 healthy behaviours. The proportion of both older and younger adults meeting the physical activity recommendations (OR=0.97, 95%CI=0.95-0.98 and OR=0.94, 95%CI=0.91-0.97 respectively), watching less than 14 hours of TV per week (OR=0.96, 95%CI=0.94-0.99 and OR=0.94, 95%CI=0.90-0.99 respectively) and who were a healthy weight (OR=0.95, 95%CI=0.92-0.99 and OR=0.96, 95%CI=0.94-0.98 respectively) decreased over time. The proportion of older adults meeting the fruit and vegetable recommendations (OR=0.90, 95%CI=0.84-0.96) and not consuming fast food (OR=0.94, 95%CI=0.88-0.99) decreased over time. Although older adults meet more health behaviours than younger adults, the decreasing prevalence of healthy nutrition behaviours in this age group needs to be addressed.
Influence of Volatile Anesthesia on the Release of Glutamate and other Amino Acids in the Nucleus Accumbens of Alcohol-Withdrawn Rats
<p>An alcohol withdrawal syndrome is a potentially life-threatening condition, which may occur when patients with alcohol use disorders have to undergo general anesthesia. Excitatory amino acids, such as glutamate, act as neurotransmitters and play a key role in alcohol withdrawal. We investigated the influence of isoflurane, sevoflurane, and desflurane anesthesia on the profile of excitatory and inhibitory amino acids in the Nucleus accumbens (NAcc) of alcohol-withdrawn rats (AWR).</p> <p>80 Wistar rats were randomized into two groups of 40, pair-fed with alcoholic or non-alcoholic nutrition. Nutrition was withdrawn and microdialysis trials were performed to measure the activity of amino acids in the NAcc. The withdrawal onset time was determined in a first trial with 20 rats. 60 rats then received isoflurane, sevoflurane, or desflurane anesthesia for three hours during withdrawal period, followed by one hour of elimination. Amino acid concentrations were obtained by chromatography. Quantification was done by integration and analysis of the area under the curve (AUC).</p>

PREVALENCE OF ALCOHOL USE, DEPRESSIVE AND ANXIETY DISORDERS AMONG PATIENTS OF OPIOID AGONIST TREATMENT PROGRAMMES IN UKRAINE
<p>Prevalence data collection was organised based on Opioid Assisted Treatment centres in three Ukrainian cities, Kyiv (one centre, centre of Ukraine), Sumy (one centre, east), and Lviv (one centre, west), during the period from October 2021 to January 2022 (before the full-scale Russian invasion). </p><p>The staff of OAT centres did all the data collection. Doctors, psychologists, or social workers who previously obtained detailed instructions made individual screenings for medication during patients' appointed visits. All the questionaries (AUDIT for alcohol use patterns, PHQ-9 for depression symptoms, GAD-7 for anxiety symptoms) were printed out in the Ukrainian language and filled in together with each patient. In addition, the number of missed visits on site for substitute medication for the last month was counted, and blood alcohol levels were recorded with the breath-alcohol measuring instrument (Drager Alcotest 6820). </p><p>The dataset is based on the results of the program of the Global Fund to Fight AIDS, Tuberculosis and Malaria, "Accelerating Progress in Reducing the Burden of Tuberculosis and HIV Infection in Ukraine", in accordance with the Grant Agreement between the Public Health Center of Ministry of Health of Ukraine and the Global Fund to Fight AIDS, Tuberculosis and Malaria No. 1936 of December 04, 2020. The donor was not involved in the study design, the collection, analysis and interpretation of data or the preparation of the article.</p>

<p>The first set of data comprise FTIR spectra of Ca-alginate, Ag/Ca-alginate, pure PVA, PVA/Ca-alginate, and PVA/Ag/Ca-alginate fibers. The fibers were obtained by extrusion and gelation of the aqueous solutions with following compositions: for Ag/Ca-alginate fibers - 1.27 ± 0.08 w/v Na-alginate and AgNPs at 2.6 mM, for Ca-alginate fibers - 1.27 ± 0.08 w/v Na-alginate, for PVA fibers - 5.7 % w/v PVA, for PVA/Ca-alginate fibers - 5.7 % w/v PVA and 1.27 ± 0.08 w/v Na-alginate, and for PVA/Ag/Ca-alginate fibers - 5.7 % w/v PVA, 1.27 ± 0.08 w/v Na-alginate and AgNPs at 2.6 mM.</p> <p>The second set of data are UV-Visible absorption spectra of the initial PVA/Ag/Na-alginate colloid solution (5.7 % w/v PVA, 1.27 ± 0.08 w/v Na-alginate, 2.6 mM nominal silver concentration) and resulting fibers produced after gelling of alginate after dissolution in 2.28 % w/v sodium citrate solution.</p> <p>The third set of data are force and stroke values over time measured by using a Universal Testing Machine for Ca-alginate fibers obtained from the 1.27 ± 0.08 % w/v Na-alginate solution and PVA/Ca-alginate fibers, obtained from the solution containing 5.67 % w/v PVA and 1.27 ± 0.08 % w/v Na-alginate at the initial time, and after drying and rehydration for 24 h and 48 h in physiological saline solution. </p>
