Similar transcriptomic responses to early and late drought stresses produce divergent phenotypes in sunflower (Helianthus annuus L.)
<p>Cultivated sunflower (<em>Helianthus</em> <em>annuus</em> L.) exhibits numerous phenotypic and transcriptomic responses to drought. However, the ways in which these responses vary with differences in drought timing and severity are insufficiently understood. We used phenotypic and transcriptomic data to evaluate the response of sunflower to drought scenarios of different timing and severity in a common garden experiment. Using a semi-automated outdoor high-throughput phenotyping platform, we grew six oilseed sunflower lines under control and drought conditions. Our results reveal that similar transcriptomic responses can have disparate phenotypic effects when triggered at different developmental time points. Leaf transcriptomic responses, however, share similarities despite timing and severity differences (e.g., 523 differentially expressed genes (DEGs) were shared across all treatments), though increased severity elicits greater differences in expression, particularly during vegetative growth. Across treatments, DEGs were highly enriched for genes related to photosynthesis and plastid maintenance. A co-expression analysis identified a single module (M8) enriched in all drought stress treatments. Genes related to drought, temperature, proline biosynthesis, and other stress responses were overrepresented in this module. In contrast to transcriptomic responses, phenotypic responses were largely divergent between early and late drought. Early-stressed sunflowers responded to drought with reduced overall growth, but became highly water-acquisitive during recovery irrigation, resulting in overcompensation (higher aboveground biomass and leaf area) and a greater overall shift in phenotypic correlations, whereas late-stressed sunflowers were smaller and more water use-efficient. Taken together, these results suggest that drought stress at an earlier growth stage elicits a change in development that enables greater uptake and transpiration of water during recovery, resulting in higher growth rates despite similar initial transcriptomic responses.</p>
Disease parameters following ocular herpes simplex virus type 1 infection are similar in male and female BALB/C mice
<p>Sex-related differences in the incidence or severity of infection have been described for multiple viruses. With herpes simplex viruses, the best example is HSV-2 genital infection where women have a higher incidence of infection and can have more severe infections than men. HSV-1 causes several types of infections including skin and mucosal ulcers, keratitis, and encephalitis in humans that do not appear to have a strong biological sex component. Given that mouse strains differ in their MHC loci it is important to determine if sex differences occur in multiple strains of mice. Our goal was to answer two questions: Are virus-related sex differences present in BALB/C mice and does virulence of the viral strain have an effect? We generated a panel of recombinant HSV-1 viruses with differing virulence phenotypes and characterized multiple clinical correlates of ocular infection in BALB/c mice. We found no sex-specific differences in blepharitis, corneal clouding, neurovirulence, and viral titers in eye washes. Sex differences in neovascularization, weight loss, and eyewash titers were observed for some recombinants, but these were not consistent across the phenotypes tested for any recombinant virus. Considering these findings, we conclude that there are no significant sex-specific ocular pathologies in the parameters measured, regardless of the virulence phenotype following ocular infection in BALB/c mice, suggesting that the use of both sexes is not necessary for the bulk of ocular infection studies.</p>
Data from: Geographical variation and discrimination of social calls in male great Himalayan leaf-nosed bats: Do functionally similar calls have similar patterns?
<p><span>Geographical variation in animals' acoustic signals has received much attention. However, few studies have compared the patterns and underlying selective forces driving geographical divergence of vocalizations with similar and different functions within the same species. Also, the social consequences of geographical divergence in acoustic signals are still rather poorly understood. Here we recorded three types of social calls of male great Himalayan leaf-nosed bats (<em>Hipposideros armiger</em>) across eight colonies in China. Two calls share similar functions and the third has a function distinct from the other two. We examined the patterns and causes of geographical variation of each of these calls. We found that all three calls had significant geographic variation with similar patterns of spatial variation. Only one of the two social calls with similar functions was found to be affected by genetic drift, while the other two calls</span> <span>were not affected by selection and drift, or morphological constraints. Furthermore, we found that bats could discriminate between vocalizations of their own colony and those of an allopatric colony. Overall, these results suggest that acoustic signals with similar functions may be shaped by different driving forces and acoustic signals with different functions may exhibit similar geographical patterns. This study expands our limited knowledge of the patterns of geographical variation of vocalizations emitted at different emotional states and highlights the importance of comparing simultaneously patterns and causes of geographical divergence of vocalizations with similar and different functions.</span></p>

Seedling performance in a dioecious tree species is similar near female and male conspecific adults despite differences in colonization by arbuscular mycorrhizal fungi
<p>Plant–soil feedbacks (PSFs) are a key driver of species diversity and composition in plant communities worldwide; however, the factors that may cause feedbacks to vary within species are rarely examined. In dioecious species, the strength of feedbacks may differ near female plants that produce seed versus near male plants (which do not) because repeated inputs of seeds and high seedling densities near females may cause accumulation of host‐specific soil microbes that influence seedling performance. To test whether conspecific seedling performance is reduced near seed‐producing female trees relative to male or heterospecific trees, we conducted shadehouse and field experiments with a dioecious tropical tree species, Virola surinamensis (<em>Myristicaceae</em>), on Barro Colorado Island, Panama. The shadehouse experiment isolated the effect of soil microbial communities on seedling growth and allowed us to quantify colonization by mutualistic arbuscular mycorrhizal (AM) fungi, while the field experiment allowed us to assess seedling survival and growth in the presence of nearby conspecific adults and seedlings. In both experiments, seedling performance was similar between seedlings grown in the soil microbial communities and field environments underneath female conspecific, male conspecific, and heterospecific trees. However, contrary to expectation, seedling colonization by AM fungi was higher in male conspecific soil microbial communities than in female or heterospecific soil microbial communities at the end of the shadehouse experiment. Together, our experiments show that while differences among female and male plants in dioecious species may influence the association of conspecific seedlings with AM fungi in their soils, this variation does not necessarily translate directly to differences in seedling performance, at least over the time frame of our experiments. Studies of additional dioecious species are needed to help determine differences in soil microbial communities beneath male and female plants and to assess the role of seed input versus adult root systems in driving PSFs.</p>
Data for: Sourdough starters exhibit similar succession patterns but develop flour-specific climax communities
<p>The microbial fermentation behind sourdough bread is among our oldest technologies, yet there are many opportunities for sourdough science to learn from traditional bakers. We analyzed the microbiome structure and function of 40 starters (grown from 10 types of flour) over 14 days and identified 6 distinct stages of succession. At each stage, bacterial taxa correlate with key functions that determine bread quality, including pH, rise, and aromatic profile. Day 1 starter cultures were dominated by microorganisms commonly associated with plants and flour and by aromas similar to toasted grain/cereal. Bacterial diversity peaked from days 2-6 as taxa shifted from opportunistic/generalist bacteria associated with flour inputs, toward specialized climax bacterial communities (days 10–14) characterized by acid-tolerant taxa and fruity (p<3.03e-03), sour (p<1.60e-01), and fermented (p<1.47e-05) aromas. This collection of traits changes predictably through time, regardless of flour type, highlighting patterns of bacterial constraints and dynamics that are conserved across systems and scales. Yet, while sourdough climax communities exhibit similar markers of maturity (i.e., pH ≤4 and enriched in <em>Lactobacillus</em> [mean abundance 48.1%], <em>Pediococcus</em> [mean abundance 22.7%], and/or <em>Gluconobacter</em> [mean abundance 19.1%],), we also detected specific taxa and aromas associated with each type of flour. Our results address important ecological questions about the relationship between community structure and function and may enable bakers to deliberately select for specific sourdough starter and bread characteristics.</p>
