Data from: Evaluating the impact of genomic data and priors on Bayesian estimates of the angiosperm evolutionary timescale

The evolutionary timescale of angiosperms has long been a key question in biology. Molecular estimates of this timescale have shown considerable variation, being influenced by differences in taxon sampling, gene sampling, fossil calibrations, evolutionary models, and choices of priors. Here, we analyze a data set comprising 76 protein-coding genes from the chloroplast genomes of 195 taxa spanning 86 families, including novel genome sequences for 11 taxa, to evaluate the impact of models, priors, and gene sampling on Bayesian estimates of the angiosperm evolutionary timescale. Using a Bayesian relaxed molecular-clock method, with a core set of 35 minimum and two maximum fossil constraints, we estimated that crown angiosperms arose 221 (251-192) Ma during the Triassic. Based on a range of additional sensitivity and subsampling analyses, we found that our date estimates were generally robust to large changes in the parameters of the birth-death tree prior and of the model of rate variation across branches. We found an exception to this when we implemented fossil calibrations in the form of highly informative gamma priors rather than as uniform priors on node ages. Under all other calibration schemes, including trials of seven maximum age constraints, we consistently found that the earliest divergences of angiosperm clades substantially predate the oldest fossils that can be assigned unequivocally to their crown group. Overall, our results and experiments with genome-scale data suggest that reliable estimates of the angiosperm crown age will require increased taxon sampling, significant methodological changes, and new information from the fossil record.

Data from: Evaluation of the impact of RNA preservation methods of spiders for de novo transcriptome assembly

With advances in high-throughput sequencing technologies, de novo transcriptome sequencing and assembly has become a cost-effective method to obtain comprehensive genetic information of a species of interest, especially in non-model species with large genomes such as spiders. However, high-quality RNA is essential for successful sequencing and sample preservation conditions require careful consideration for the effective storage of field-collected samples. To this end, we report a streamlined feasibility study of various storage conditions and their effects on de novo transcriptome assembly results. The storage parameters considered include temperatures ranging from room temperature to -80°C; preservatives, including ethanol, RNAlater, TRIzol, and RNAlater-ICE; and sample submersion states. As a result, intact RNA was extracted and assembly was successful when samples were preserved at low temperatures regardless of the type of preservative used. The assemblies as well as the gene expression profiles were shown to be robust to RNA degradation, when 30 million 150 bp paired-end reads are obtained. The parameters for sample storage, RNA extraction, library preparation, sequencing, and in silico assembly considered in this work provide a guideline for the study of field-collected samples of spiders.

Data from: Mindfulness online: a preliminary evaluation of the feasibility of a web-based mindfulness course and the impact on stress

OBJECTIVES: Stress has been shown to have a number of negative effects on health over time. Mindfulness interventions have been shown to decrease perceived stress but access to interventions is limited. Therefore, the effectiveness of an online mindfulness course for perceived stress was investigated. DESIGN: A preliminary evaluation of an online mindfulness course. PARTICIPANTS: This sample consisted of 100 self-referrals to the online course. The average age of participants was 48 years and 74% were women. INTERVENTIONS: The online programme consisted of modules taken from Mindfulness Based Stress Reduction and Mindfulness Based Cognitive Therapy and lasted for approximately 6 weeks. PRIMARY AND SECONDARY OUTCOME MEASURES: Participants completed the Perceived Stress Scale (PSS) before the course, after the course and at 1-month follow-up. Completion of formal (eg, body scan, mindful movement) and informal (eg, mindful meal, noticing) mindfulness activities was self-reported each week. RESULTS: Participation in the online mindfulness course significantly reduced perceived stress upon completion and remained stable at follow-up. The pre-post effect size was equivalent to levels found in other class-based mindfulness programmes. Furthermore, people who had higher PSS scores before the course reported engaging in significantly more mindfulness practice, which was in turn associated with greater decreases in PSS. CONCLUSIONS: Because perceived stress significantly decreased with such limited exposure to mindfulness, there are implications for the accessibility of mindfulness therapies online. Future research needs to evaluate other health outcomes for which face-to-face mindfulness therapies have been shown to help, such as anxiety and depressive symptoms.

Stepping into the Wildeverse: Evaluating the impact of augmented reality mobile gaming on pro-conservation behaviours

<p>Promoting pro-conservation behaviours has become a priority for conservation organisations worldwide. Yet, current engagement strategies still face a number of barriers to creating successful interventions at the scale needed to meet global sustainability challenges.</p> <p>Online and mobile games enjoy immense worldwide popularity, tapping into an audience not normally reached through conventional conservation outreach channels. Despite this potential to be a new, high impact and scalable platform for promoting pro-environmental behaviours, the opportunities within digital games for conservation have thus far been little explored and organisations have called for robust impact evaluations for this medium. Therefore, we investigated the effectiveness of the augmented reality game Wildeverse, which seeks to generate support for ape conservation and encourage pro-environmental behaviours.</p> <p>We conducted a randomised control trial to experimentally compare the impacts of this game against watching a documentary, a conventional conservation outreach intervention. We compared changes in participants' knowledge, attitudes and revealed donation behaviours across the two groups and found that games performed as well as documentaries in supporting positive environmental knowledge and attitudes.</p> <p>The results gathered from this study provide experimental insight into the potential for the broader use of digital games for conservation outreach and also provide evidence against the argument that gaming can detract from real-world environmental problems creating a disconnect with these issues. However, results from this study could not provide any evidence that Wildeverse was successful in achieving its additional aim of encouraging players to donate to conservation, which provides evidence for the existence of an Environmental Values-Behaviour (EVB) gap in conservation gaming.</p> <p>The gaming industry is global and rapidly growing and should no longer be ignored as an avenue for conservation outreach. We recommend several ways in which future studies can expand on this work to better understand how to effectively harness the power of digital games to drive tangible change that benefits biodiversity. </p>
