Data from: Increased soluble IL-7 receptor concentrations associate with improved IL-7 therapy outcomes in SIV-infected ART-treated Rhesus macaques
The use of interleukin-7 (IL-7) as an immunorestorative therapeutic has proven effective in HIV infection, cancer and bone marrow transplantation. Mediating its activity through membrane-bound IL-7 receptor α (mCD127), IL-7 therapy increases T-cell numbers and survival. A soluble form, sCD127, is found in plasma, and we have previously identified increased plasma sCD127 concentrations in HIV infection. Furthermore, patients with high sCD127 exhibited the best viral control, implicating a role for IL-7 or sCD127 directly in improved virologic/immunologic outcomes. The role of the cytokine IL-7 in elevating sCD127 levels was addressed here through assessment of retrospective samples obtained from SIV-infected antiretroviral (ART)-treated Rhesus macaques. IL-7 was administered in clustered weekly doses, allowing for an assessment prior, during and following IL-7 administration. The levels of sCD127 remained relatively unchanged during both early SIV infection and following initiation of ART. However, treatment with IL-7 increased sCD127 concentrations in most animals, transiently or persistently, paralleling increased T-cell numbers, correlating significantly with CD8+ T-cell levels. In addition, proliferating CD4+ or CD8+ T-cells (measured by Ki67) increased in association with elevated sCD127 concentrations. Finally, a high concentration of sCD127 in IL7-treated animals was associated with increased retention of T-cells (measured by BrDU). In addition, a lack, or loss of viral control was associated with more pronounced and frequent elevations in plasma sCD127 concentrations with IL-7 therapy. In summary, plasma sCD127 levels in SIV-infected ART-treated macaques was associated with therapeutic IL-7 administration, with higher sCD127 levels in macaques demonstrating the best T-cell responses. This study furthers our knowledge regarding the interrelationship between increased IL-7 levels and elevated sCD127 levels that may have implications for future IL-7 immunotherapeutic approaches in HIV-infected patients.

Integration of single-cell transcriptome and chromatin accessibility of early gonads development among goats, pigs, macaques, and humans
<p>The early gonads of mammals contain primordial germ cells (PGCs) and somatic cell precursors that are essential for sex determination and gametogenesis. Although it is extensively documented in mice, the development of early gonads in non-rodents remains to be delineated. Because molecular differences between mouse and human gonadal cells have been reported, it warrants the study of the key markers and regulatory features that are conserved or divergent between non-rodent species and human. Here, we integrate single-cell transcriptome and chromatin accessibility analysis to identify regulatory signatures of PGCs and somatic cells in the early gonads of goats, pigs, macaques, and humans. We identify the evolutionarily conserved and species-specific events, including genes expression, signaling pathways, and cell-cell interactions. We also uncover potential cis-regulatory elements and key transcription factors in PGCs and somatic cells. Our datasets provide important resources for better understanding the evolutionary programs of PGCs and gonadal somatic cell development in mammals.</p>
Dataset for Influence of ecological and social factors on huddling behaviour and cluster organisation in Japanese macaques (Macaca fuscata)
<p>Dataset for Influence of ecological and social factors on huddling behaviour and cluster organisation in Japanese macaques (Macaca fuscata)</p> <p><span>Huddling behaviour is present in many animal species. This behaviour involves maintaining close physical contact with conspecifics to minimise heat loss and, in general, reduce energy expenditure. Additionally, this behaviour also facilitates complex social interactions within a population. In Japanese macaques, this behaviour is observed in many populations across Japan, including Shodoshima, where huddling clusters can reach up to 100 individuals in winter. Based on several studies on this species, it appears that huddling, or <span> </span>in Japanese, is influenced by both meteorological factors and social relationships between individuals. The objective of this study is to understand the determinants that drive the expression, variation, and organisation of huddling behaviour. Two hypotheses were formulated. The first hypothesis posits that the formation and variations in the size and number of clusters are influenced by meteorological factors, while the second hypothesis suggests that the number and position of individuals within a cluster is related to existing relationships between individuals. To test these, data on the number, size, and individuals composing a cluster were collected, allowing building huddling social networks. Simultaneously, meteorological measurements were taken, along with observations on dominance and grooming interactions between individuals. This allowed us to create several statistical models and social networks for comparison. Our results suggest that the initiation of huddling is mainly related to solar radiation energy, while variations in number and size could be explained by temperature. Moreover, the organisation within a cluster is not random but reflects relationships between individuals. However, it is not a single parameter but rather a combination of social relationships that underpin the organisation of individuals within clusters.</span></p>
