Primary hepatocyte culture from Oreochromis niloticus fish as a tool for environmental toxicology

<p>Accumulated chemicals have been impairing water quality over decades of human activities. Organisms such as fish are often used for risk assessment, as chemical analysis is insufficient to assess the impact on health. Consequently, researchers have faced challenges in ecotoxicology, concerning the use of animals. Primary and cell line cultures are good alternatives for reducing animal use. This study aims to evaluate the use of primary hepatocytes culture from Oreochromis niloticus as an in vitro tool for screening environmental contaminants and pollutants. Metal cadmium (Cd) is the pollutant model.</p>

Phenotypic screening using waveform analysis of synchronized calcium oscillations in primary cortical cultures

<p><span>At present, <em>in</em> <em>vitro</em> phenotypic screening methods are widely used for drug discovery. In the field of epilepsy research, measurements of neuronal activities have been utilized for predicting efficacy of anti-epileptic drugs (AEDs). Fluorescence measurements of calcium oscillations in neurons are commonly used for measurement of neuronal activities, and some anti-epileptic drugs have been evaluated using this assay technique. However, changes in waveforms were not quantified in previous reports. Here, we have developed a high-throughput screening system containing a new analysis method for quantifying waveforms, and our method has successfully enabled simultaneous measurement of calcium oscillations in a 96-well plate. Features of waveforms were extracted automatically and allowed the characterization of some anti-epileptic drugs using principal component analysis. Moreover, we have shown that trajectories in accordance with the concentrations of compounds in principal component analysis plots were unique to the mechanism of anti-epileptic drugs. We believe that an approach that focuses on the features of calcium oscillations will lead to better understanding of the characteristics of existing anti-epileptic drugs and allow prediction of the mechanism of action (MoA) of novel drug candidates.</span></p>

Data from: Exogenous α-synuclein hinders synaptic communication in cultured cortical primary rat neurons

Amyloid aggregates of the protein α-synuclein (αS) called Lewy Bodies (LB) and Lewy Neurites (LN) are the pathological hallmark of Parkinson's disease (PD) and other synucleinopathies. We have previously shown that high extracellular αS concentrations can be toxic to cells and that neurons take up αS. Here we aimed to get more insight into the toxicity mechanism associated with high extracellular αS concentrations (50-100 μM). High extracellular αS concentrations resulted in a reduction of the firing rate of the neuronal network by disrupting synaptic transmission, while the neuronal ability to fire action potentials was still intact. Furthermore, many cells developed αS deposits larger than 500 nm within five days, but otherwise appeared healthy. Synaptic dysfunction clearly occurred before the establishment of large intracellular deposits and neuronal death, suggesting that an excessive extracellular αS concentration caused synaptic failure and which later possibly contributed to neuronal death.
