Responses of a resident group to an outsider in the blue-breasted quail: a paradigm for studying social resettlement of dispersers

<p>Dispersal is an individual life history trait that can influence the ecological and evolutionary dynamics of both the source and recipient populations. Current studies of animal dispersal have paid little attention to how the responses of residents in a recipient population affect the social resettlement of dispersers into a new habitat. We addressed this question in the blue-breasted quail Synoicus chinensis by designing an outsider introduction experiment to simulate a scenario of interaction between residents and dispersers. In the experiment, we introduced an unfamiliar quail into a group of three differently ranked residents and then examined their behavioral responses to the arrival of the outsider. We found that all residents made negative responses by pecking at the outsider to maintain their pecking order, in which high-ranked residents displayed significantly greater intensity than those of lower ranks. This result highlighted that adverse behavioral responses of residents would prevent outsiders from obtaining hierarchical dominance in the recipient group. Moreover, the residents' sex ratio, their relative ages to the outsiders, and whether outsiders counter-pecked at the residents all influenced the probability of outsiders prevailing against the residents. Those outsiders that displayed counter-peck courage were more likely to gain higher dominance and hence resettle into the recipient group successfully. Our findings suggest that resident groups may impose a selection among dispersers via adverse behavioral responses. Therefore, social factors that can influence the resettlement step of dispersers in a new habitat should be accounted for in future studies of animal dispersal.</p>

Data from: Sex-specific effects of dietary restriction on physiological variables in Japanese quails

<p>Nutritional limitation is a common phenomenon in nature that leads to trade-offs among processes competing for limited resources. These trade-offs are mediated by changes in physiological traits such as growth factors and circulating lipids. However, studies addressing the sex-specific effect of nutritional deficiency on these physiological variables are limited in birds. We used dietary restriction to mimic the depletion of resources to various degrees and investigated sex-specific effects on circulating levels of insulin-like growth factor 1 (IGF-1) and triglycerides in Japanese quails (<em>Coturnix japonica</em>) subjected to <em>ad libitum</em>, 20%, 30% or 40% restriction of their daily requirement, for two weeks. We also explored the association of both physiological variables with body mass and egg production. While dietary restriction showed no effects on circulating IGF-1, this hormone exhibited a marked sexual difference, with females having 64.7% higher IGF-1 levels than males. Dietary restriction significantly reduced plasma triglyceride levels in both sexes. Females showed more than six-fold higher triglyceride levels than males. Triglyceride levels were positively associated with body mass in females while showed not association in males. Overall, our findings revealed sex-specific expression of physiological variables under dietary restriction conditions, which coincide with body size.</p>

Dietary fat supplements influence weight gain and egg production but not offspring sex ratios in Japanese quail

<p>Sex ratio theory suggests that females should bias offspring sex ratios based on maternal condition and the availability of critical food resources. Work in birds indicates that females do, indeed, bias sex ratios according to maternal condition and food quality and quantity. Yet it is unknown whether these sex ratio skews occur due to fluctuations in particular micro- or macro-nutrients, caloric content overall, or even the perception of food availability. We hypothesized that dietary fats may drive biases in offspring sex ratios, because measures of maternal condition often reflect fat reserves, and fats are critical for the process of egg-laying in birds. To test this, we provided breeding Japanese quail, a species that biases sex ratios in response to maternal condition, with either a control breeding diet or a diet supplemented with two oils (safflower oil and flaxseed oil). These oils were chosen for their high omega-3 and omega-6 fatty acid content as well as their importance in mammalian sex allocation. We then measured influences of these diets on the sex ratio of offspring, the change in maternal weight, and the laying rates of female quail. The dietary oil supplements increased weight gains in quail but decreased the number of eggs laid during the experiment. There was no influence of the oil supplements on offspring sex ratios. This indicates that fat may not be a macro-nutrient involved in the process of sex ratio adjustment in quail.</p>

A custom-made AAV1 variant (AAV1-T593K) enables efficient transduction of Japanese quail neurons in vitro and in vivo

<p><span>The widespread use of rodents in neuroscience has prompted the development of optimized viral variants for transduction of brain cells, <em>in vivo</em>. However, many of the viruses developed are less efficient in other model organisms, with birds being among the most resistant to transduction by current viral tools. Resultantly, the use of genetically-encoded tools and methods in avian species is markedly lower than in rodents; likely holding the field back. We sought to bridge this gap by developing custom viruses towards the transduction of brain cells of the Japanese quail. We first develop a protocol for culturing primary neurons and glia from quail embryos, followed by characterization of cultures via immunostaining, single-cell mRNA sequencing, patch clamp electrophysiology and calcium imaging. We then leverage the cultures for the rapid screening of various viruses, only to find that all yielded poor to no infection of cells <em>in vitro</em>. However, few infected neurons were obtained by AAV1 and AAV2. Scrutiny of the sequence of the AAV receptor found in quails led us to rationally design a custom-made AAV variant (AAV1-T593K; AAV1*) that exhibits improved transduction efficiencies <em>in vitro</em> and <em>in vivo</em> (eight- and five-fold, respectively). Together, we present a unique culturing method, transcriptomic profiles of quail's brain cells and a custom-tailored AAV1 for transduction of quail neurons <em>in vitro </em>and<em> in vivo.</em></span></p>

Data from: Evidence of genetic erosion in a peripheral population of a North American game bird: the Montezuma quail (Cyrtonyx montezumae)

Population extirpations are often precursors to species extinctions. Anthropogenic activities often lead to smaller populations that are more prone to extirpations and advocates for active conservation management have recently called for the preservation and monitoring of genetic diversity, particularly with regard to the adaptive potential of vulnerable populations. We used genomics and curated arrays of molecular markers, including those expected to impact key fitness traits, to quantify evidence of genomic erosion in core and peripheral populations of a gallinaceous bird. The Montezuma quail (Cyrtonyx montezumae) is a game species considered vulnerable to extirpation in Texas, but core populations in Arizona and New Mexico are robust and have the potential to serve as genetic reservoirs. We sequenced the Montezuma quail genome then developed a single nucleotide polymorphism (SNP) assay to quantify genetic variation, effective population sizes, signatures of natural selection, and population structure. We genotyped SNPs from gene deserts and from genes associated with fitness traits and found the isolated Texas population exhibits an extremely small effective population size, is genetically distinct from our Arizona and New Mexico samples, and has reduced heterozygosity at the fitness-related markers. Thus, our samples from Texas exhibit symptoms of genetic erosion that could exacerbate future risk of local extirpation. Management agencies must decide if active conservation efforts such as assisted gene flow or genetic rescue are now warranted. This decision may not be straightforward because the current conservation status of the Texas population reflects its isolated geographic locale on the periphery of the species' range.

Data from: Altered embryonic development in northern bobwhite quail (Colinus virginianus) induced by pre-incubation oscillatory thermal stresses mimicking global warming predictions

Global warming is likely to alter reproductive success of ground-nesting birds that lay eggs normally left unattended for days or even weeks before actual parental incubation, especially in already warm climates. The native North American bobwhite quail (Colinus virginianus) is such a species, and pre-incubation quail eggs may experience temperatures &gt;45°C. Yet, almost nothing is known about embryonic survival after such high pre-incubation temperatures. Freshly laid bobwhite quail eggs were exposed during a 12 day pre-incubation period to one of five thermal regimes: low oscillating temperatures (25-40°C, mean=28.9°C), high oscillating temperatures (30-45°C, mean=33.9°C), low constant temperatures (28.85°C), high constant temperatures (mean=33.9°C), or commercially employed pre-incubation temperatures (20°C). After treatment, eggs were then incubated at a standard 37.5°C to determine subsequent effects on embryonic development rate, survival, water loss, hatching, and embryonic oxygen consumption. Both quantity of heating degree hours during pre-incubation and specific thermal regime (oscillating vs. non-oscillating) profoundly affected important aspects of embryo survival and indices of development and growth Pre-incubation quail eggs showed a remarkable tolerance to constant high temperatures (up to 45°C), surviving for 4.5±0.3 days of subsequent incubation, but high oscillating pre-incubation temperature increased embryo survival (mean survival 12.2±1.8 days) and led to more rapid development than high constant temperature (maximum 38.5ºC), even though both groups experienced the same total heating degree-hours. Oxygen consumption was ~200-300 μl O2.egg.min-1 at hatching in all groups, and was not affected by pre-incubation conditions. Oscillating temperatures, which are the norm for pre-incubation quail eggs in their natural habitat, thus enhanced survival at higher temperatures. However, a 5°C increase in pre-incubation temperature, which equates to the predicted long-term increases of 5°C or more, nonetheless reduced hatching rate by approximately 50%. Thus, while pre-incubation bobwhite eggs may be resiliant to moderate oscillating temperature increases, global warming will likely severely impact wild bobwhite quail populations, especially in their strongholds in southern latitudes.

Female preference and adverse developmental effects of glyphosate-based herbicides on ecologically relevant traits in Japanese quail

<p>Controversial glyphosate-based herbicides (GBHs) are the most frequently used herbicides globally. An increasing number of studies have identified GBH residues in soil, water and even human food that may expose non-target organism including wildlife, livestock, and humans to health risks. After a heated debate, European Union allowed the use of GBHs to continue until 2022, after which their risks will be re-evaluated. Thus, decision makers urgently need scientific evidence on GBH residues and their possible effects on ecosystems. An important, yet neglected, aspect is to assess whether animals show preference or avoidance for GBH contaminated food, as it can influence the likelihood of adverse health effects in wildlife. Here, using Japanese quails (<i>Coturnix japonica</i>) as our model, we show that females preferred GBH-contaminated food compared to control food. In females, exposure to GBHs caused delayed plumage development, and GBH residues were present in eggs, muscles and liver. These results indicate that female preference is not adaptive, potentially exposing non-target animals to greater risk of adverse effects of GBHs in natural and agricultural environments. Our results on tissue residues suggest that further studies are needed to understand the risks of such residues in the food chain.</p>
