Advancing the automation of plant nucleic acid extraction for rapid diagnosis of plant diseases in space
Human space exploration missions will continue the development of sustainable plant cultivation in what are obviously novel habitat settings. Effective pathology mitigation strategies are needed to cope with plant disease outbreaks in any space-based plant growth system. However, few technologies currently exist for space-based diagnosis of plant pathogens. Therefore, we developed a method of extracting plant nucleic acid that will facilitate the rapid diagnosis of plant diseases for future spaceflight applications. The microHomogenizer from Claremont BioSolutions, originally designed for bacterial and animal tissue samples, was evaluated for plant–microbial nucleic acid extractions. The microHomogenizer is an appealing device in that it provides automation and containment capabilities that would be required in spaceflight applications. Two different plant pathosystems were used to assess the versatility of the extraction process. Tomato and lettuce plants were respectively inoculated with a fungal plant pathogen, an oomycete pathogen, and a plant viral pathogen. The microHomogenizer, along with the developed protocols, proved to be an effective mechanism for producing DNA from two pathosystems, in that PCR and sequencing of the resulting samples demonstrated clear DNA-based diagnoses. Thus, this investigation advances the efforts to automate nucleic acid extraction for future plant disease diagnosis in space.

Data for: Proteomics reveal biomarkers for diagnosis, disease activity and long-term disability outcomes in multiple sclerosis
<p>Protein levels were measured in cerebrospinal fluid samples (CSF; n = 186) and plasma samples (n = 165) from persons with multiple sclerosis and healthy controls. CSF samples and plasma samples were taken from 92 persons with CIS or RRMS at Linköping University Hospital, Sweden and 51 persons with CIS or RRMS at the Karolinska University Hospital, Sweden. Everyone fulfilled the revised McDonald criteria from 2010 and 2017 for CIS or Multiple sclerosis (MS). Age-matched healthy controls (HC) were recruited from healthy blood donors (23 at the Linköping University hospital and 20 at the Karolinska University Hospital). The concentration of 1463 proteins were measured using the Olink Explore platform which uses Proximity Extension Assay (PEA) technology. The proteins were preselected from four Olink panels: Explore 384 Cardiometabolic, Explore 384 Inflammation, Explore 384 Neurology, and Explore 384 Oncology. The protein concentrations are given as Olink’s relative protein quantification unit on log2 scale: Normalized Protein Expression (NPX). The NPX values were intensity normalized by Olink.</p>

PMID: 30795994.</p> <p>This is the abstract:</p> <p><strong>Background and aims: </strong>Adults with congenital heart disease (ACHD) are at risk of overweight and obesity, two major health problems, though underweight can be a negative prognostic factor too. Awareness of the body mass index (BMI) in ACHD is very limited. The present study describes the use and prevalence of BMI in Italian symptomatic hospitalized ACHD patients in relation to complexity by Bethesda system classification, diagnosis, sex and age.</p> <p><strong>Methods and results: </strong>We classified 1388 ACHD patients, aged 18-69 years, on the basis of their BMI, and compared them to the Italian reference population. In our total ACHD population we found a significantly higher prevalence of underweight compared to the Italian reference population (6.34% vs 3.20%). ACHD women were more underweight than men. Underweight decreased with age. Overweight was significantly less frequent in the total ACHD population (26.73% compared to 31.70%) in the Italian reference population. Men were more likely to be overweight than women. In statistical terms obesity was similar in the Italian reference population (10.50%) and our ACHD population (9.58%). Both overweight and obesity increased with age. Results were comparable using a diagnostic anatomical-functional classification and the Bethesda system classification.</p> <p><strong>Conclusions: </strong>In our cohort of ACHD the prevalence of underweight was double that of the Italian reference population. The prevalence of overweight was lower, while obesity was similar. Since BMI does not account for differences in body fat distribution, a future aim will be to quantify the visceral component of the adipose tissue in ACHD patients and examine their body composition in order to reflect their risk of acquired cardiovascular disease better, and either to maintain or achieve an adequate visceral component.</p>
