Azole resistance mechanisms and population structure of Aspergillus fumigatus on retail plant products

<p><em>Aspergillus fumigatus </em>is a ubiquitous saprotroph and human-pathogenic fungus that is life-threatening to the immunocompromised. Triazole-resistant <em>A. fumigatus</em><em> </em>was found in patients without prior treatment with azoles, leading researchers to conclude that resistance had developed in agricultural environments where azoles are used against plant pathogens. Previous studies have documented azole-resistant <em>A. fumigatus </em>across agricultural environments, but few have looked at retail plant products. Our objectives were to determine if azole-resistant <em>A. fumigatus </em>is prevalent<em> </em>in retail plant products produced in the United States (U.S.), as well as to identify the resistance mechanism(s) and population genetic structure of these isolates. Five hundred twenty-five isolates were collected from retail plant products and screened for azole resistance. Twenty-four isolates collected from compost, soil, flower bulbs, and raw peanuts were pan-azole resistant. Resistant isolates had the TR<sub>34</sub>/L98H, TR<sub>46</sub>/Y121F/T289A, G448S, and H147Y <em>cyp51A </em>alleles, all known to underly pan-azole resistance, as well as  WT alleles, suggesting that non-cyp51A-mechanisms contribute to pan-azole resistance in some isolates. Minimum spanning networks showed two lineages containing isolates with TR alleles or the F46Y/M172V/E427K allele, and discriminant analysis of principle components (DAPC) identified three primary clusters. This is consistent with previous studies detecting three clades of <em>A. fumigatus</em> and identifying pan-azole-resistant isolates with TR alleles in a single clade. We found pan-azole resistance in U.S. retail plant products, particularly compost and flower bulbs, which indicates the risk of exposure to these products for susceptible populations and that highly resistant isolates are likely distributed worldwide on these products.</p>

Data for: Local adaptation of a generalist hemiparasitic plant to one of its potential host plants

<p>Coevolution is often found in parasite-host interactions but has not yet been described for hemiparasitic plants and their hosts. Root hemiparasites like <em>Rhinanthus alectorolophus</em> perform photosynthesis but also parasitize other plant species, some of which (e.g. <em>Plantago lanceolata</em>) may defend themselves against parasite attack by blocking the haustoria of the parasites. We grew seedlings of the hemiparasite <em>Rhinanthus alectorolophus</em> and the potential host <em>Plantago lanceolata</em> from seven grassland sites in a factorial design. To detect differences in host defence, we also included hosts from two 'naïve' populations from regions where the parasite does not occur.<em> R. alectorolophus</em> grew consistently larger and had higher fitness with sympatric than with allopatric hosts, suggesting parasite adaptation to local host populations. Moreover, <em>R. alectorolophus</em> remained smallest with allopatric hosts from the same region and reached intermediate sizes with allopatric hosts from other regions or naïve hosts, suggesting host adaptation to parasites at the regional scale. Parasite presence reduced the size of the host plants already after four weeks, but only that of hosts with 'experience' of the parasite, suggesting an early host response. Follow-up experiments confirmed that parasites attach to hosts already after four weeks and hosts respond by changing belowground allocation patterns. However, parasite roots did not preferentially grow towards sympatric hosts. Our results suggest that local adaptation to hosts can occur even in generalist parasites and does not require specialization on individual hosts. We discuss the role of potential mechanisms, including variation in chemical signalling (early) and in host defence (late effects).</p>

Data from: neglected puzzle pieces of urban green infrastructure: richness, cover, and composition of insect-pollinated plants in traffic-related green spaces

<p>Insect-pollinated vascular plants in spontaneous vegetation provide essential ecosystem services and benefit wildlife. However, floral communities associated with traffic-related green spaces are rarely considered valuable elements of urban green infrastructure (UGI). The dataset contains information on species-based floral communities of vascular insect-pollinated plants in traffic-related green spaces in three highly populated Finnish cities. Those are Helsinki (665 558 inhabitants), Tampere (244 029 inhabitants), and Turku (175 645 inhabitants). Data were collected during the mean flowering phenophase of vascular plants in July-August 2022 from two types of locations: (i) urban (city centers) and (ii) suburban (city outskirts), and from three types of traffic-related green spaces: (i) traffic islands, (ii) parking lots, (iii) road verges. The dataset contains information for the 93 vascular insect-pollinated plant species flowering during the survey. Sampling campaign was conducted in 90 sampling sites, and the dataset contains information on the location coordinates. In addition, the dataset possesses information on the amount of garbage pieces (cigarette filters, plastic boxes, or scraps) revealed for each sampling point in traffic-related green spaces.</p>

Acclimation of thermal tolerance in juvenile plants from three biomes is supressed when extremes co-occur

<p>Given the rising frequency of thermal extremes (heatwaves and cold snaps) due to climate change, comprehending how a plant's origin affects its thermal tolerance breadth becomes vital. We studied juvenile plants from three biomes: temperate coastal rainforest, desert, and alpine. In controlled settings, plants underwent hot days and cold nights in a factorial design to examine thermal tolerance acclimation. We assessed thermal thresholds (<em>T</em><sub>crit-hot</sub> and <em>T</em><sub>crit-cold</sub>) and thermal tolerance breadth (TTB). We hypothesised that: 1) desert species would show the highest heat tolerance, alpine the greatest cold tolerance, with temperate species intermediate; 2) all species would increase heat tolerance post hot days and cold tolerance after cold nights; 3) combined exposure would broaden TTB more than individual conditions, especially in desert and alpine species. We found that biome responses were minor compared to the responses to the extreme temperature treatments. All plants increased thermal tolerance in response to hot 40°C days (<em>T</em><sub>crit-hot</sub> increased by ~3.5°C) but there was minimal change in <em>T</em><sub>crit-cold</sub> in response to the cold -2°C nights. In contrast, when exposed to both hot days and cold nights, on average plants exhibited an antagonistic response in TTB, where cold tolerance decreased and heat tolerance was reduced, and so we did not see the bi-directional expansion we hypothesised. There was, however, considerable variation among species in these responses. As climate change intensifies, plant communities, especially in transitional seasons, will regularly face such temperature swings. Our results shed light on potential plant responses under these extremes, emphasizing the need for deeper species-specific thermal acclimation insights, ultimately guiding conservation efforts.</p>

Insect root feeders incur negative density-dependent damage across plant species in an alpine meadow

<p>Although herbivores are well known to incur positive density-dependent damage and mortality, thereby likely shaping plant community assembly, the response of belowground root feeders to changes in plant density has seldom been addressed. Locally rare plant species (with lower plant biomass per area) are often smaller with shallower roots than common species (with higher plant biomass per area) in competition-intensive grasslands. Likewise, root feeders are often distributed in the upper soil layers. We hypothesized therefore that root feeders would incur a negative density (biomass) dependent damage across plant species. To test this hypothesis, we investigated the diversity and abundance of plant and root feeder species in an alpine meadow, and determined the diet of the root feeders using metabarcoding. Across all species, root feeder load decreased with increasing aboveground plant biomass, root biomass, and total plant biomass per area, indicating negative density dependence of damage across plant species. Aboveground plant biomass per area increased with increasing individual plant biomass and root depth per area across species, suggesting that rare plant species were smaller in size and had shallower root systems compared to common plant species. Both root biomass per area and root feeder biomass per area decreased with soil depth, but the root feeder biomass decreased disproportionally faster compared to root biomass with increasing root depth. Root feeder load decreased with increasing root depth, but was not correlated with the feeding preference of root feeder species. Moreover, the prediction derived from a random process incorporating vertical distributions of root biomass and root feeder biomass significantly accounted for interspecific variation in root feeder load. In conclusion, the data indicate that root feeders incur negative density-dependent damage across plant species. On this basis, we suggest that manipulative experiments should be conducted to determine the effect of the negative density-dependent damage on plant community structure, and that different types of plant-animal interactions should be concurrently examined to fully understand the effect of plant density on overall herbivore damage across plant species.</p>
