Insights into metabolic changes during epidermal differentiation as revealed by multiphoton microscopy with fluorescence lifetime imaging

Rapid developments in the field of organotypic cultures has generated a growing need for effective quality control measures during tissue development. In this study, we correlate metabolic changes with epidermal differentiation and demonstrate that multiphoton microscopy with fluorescence lifetime imaging (MPM-FLIM) can be applied as a non-invasive approach to monitor epidermal differentiation of keratinocytes with respect to proliferative and differentiated states. Keratinocytes grown at 1.5 mM Ca2+ exhibited increased expression of differentiation markers KRT1 and KRT10 compared to 60 μM Ca2+, and a metabolic shift from glycolysis to mitochondrial respiration. Fitting the fluorescence decay with a biexponential model revealed a decreased relative fraction of intracellular NADH and FAD after high calcium treatment, consistent with increased oxidative phosphorylation. Using these two parameters, the epidermal differentiation process could be monitored over a 96 h period. Implementing discriminating analysis based on k-means clustering generated clusters that correlated well with culturing time, suggesting that this methodology can be employed as part of an automated pipeline for monitoring keratinocyte differentiation.

IODP Expedition 371 Core composite images
A digital composite image (PNG) is made for each core comprising core sections scanned using a line-scan camera. The composite layout is equivalent to traditional core table photos. Top left is top of core; color and meter rule references are included.
IODP Expedition 371 Closeup images
Close-up images taken by digital cameras as requested by the science party, typically when the section-half image is not sufficient. Close-up photographs of the areas of interest may be taken from whole-round sections, pieces, or section halves in sediments and rock.
IODP Expedition 371 Section-half images
Digital section images were taken of the flat face of split cores on the Section Half Imaging Logger (SHIL) using a linescan camera at a resolution of 20 lines/mm (50 micron pixels). Cores were imaged as soon as possible after splitting to minimize color changes that occur through oxidation and drying. The SHIL produces TIF files as well as reduced-size JPG files. The TIF files are not kept online but users may request them from the IODP-JRSO Data Librarian.
IODP Expedition 371 Thin section images
Hard rock and sediment thin section images were acquired using either the JRSO-developed Petrographic Image Capture and Archival Tool (PICAT) imager or (rarely) an upright microscope and a digital camera. Sample images are acquired in unpolarized, polarized, and/or cross-polarized light. Image files are presented compressed by hole.
High resolution microsection images for: Common juniper, the oldest living non-clonal woody species across the tundra biome and the European continent

Two high resolution images of the stem section are available as .czi files. These images are from a living Juniperus communis L. branch from Abisko (Sweden) sampled in August 2021. These high-resolution photographs (2.89 pixel/μm) were created using Axio Scan 7, Zeiss, Germany.

One high resolution image of the same stem section is archived as a .tif file (49835x25587 pixels). This image is a composition of the two .czi images created using Axio Scan 7, Zeiss, with a reduced resolution and edited adding the ring-count reference points and the reference scale.

Raw images of the manuscript entitle: "Detecting life by behavior, the overlooked sensitivity of behavioral assays"

Description: Using a magnetotactic bacterial species, Magnetospirillum magneticum, we conduct a lab sensitivity experiment comparing PCR with the hanging drop behavioral assay, using a dilution series.

Data:

1.-Gel image resulted from the Magnetospirillum magneticum PCR assays.

2.-Microphotographs of Magnetospirillum magneticum obtained using the hanging drop technique and serial dilution.

3.-Videos 1 to 4.Environmental samples were taken from Agmon Hula lake, (33° 10′ N 35° 60′ E). We used the HDT (see main MS) to morphologically identify magnetotactic bacterial species.
