Soil biota diversity and plant diversity both contributed to ecosystem stability in grasslands

<p><span>Understanding the effects of diversity on ecosystem stability in the context of global change has </span><span>become</span><span> an important goal of recent ecological research. </span><span>How</span><span>ever,</span><span> the </span><span>effects of </span><span>diversity at multiple scales and trophic levels </span><span>on</span><span> ecosystem stability across environmental gradients </span><span>remain</span><span> unclear. Here, we conducted a field survey of Ξ±-, Ξ²-, and Ξ³-diversity of plants and soil biota (bacteria, fungi, and nematodes) and estimated </span><span>the </span><span>temporal ecosystem stability of </span><span>NDVI</span> <span>in</span><span> 132 plots on the Mongolian Plateau. After climate and soil environmental variables were controlled for, both the Ξ±- and Ξ²-diversity of plants and soil biota (mainly via nematodes) together with precipitation explained most variation in ecosystem stability. These findings evidence that the diversity of both soil biota and plants </span><span>contribute</span><span>s</span><span> to ecosystem stability</span><span>. Model</span><span> predictions of</span> <span>the</span><span> future effects of global changes on terrestrial ecosystem stability will require field observations of diversity of both </span><span>plants</span><span> and soil biota.</span></p>

Foraging preferences and interspecific competition generate multimodal complexity-stability relationships in an adaptive food-web framework

<p><span>Ecological theory predicts that complex ecological networks are unstable and are unlikely to persist, despite many empirical studies of such complexity in nature. To resolve real complexity-stability relationships, coupling population dynamics and trait dynamics is considered to be an important way to understand the long-term stability of ecological community assemblages. However, modelling eco-evolutionary dynamics in ecologically realistic networks is still a challenge. Here, we establish an adaptive food web model to evaluate the complexity-stability debate in a mutualist-exploiters-specialist forager-generalist forager system. Our theoretical model predicts that the connectance-stability relationship may show positive monotonic (/), negative monotonic (\), peaked (∩) and double-peaked (oscillatory) patterns. Moreover, the double-peaked pattern is only obtained when both the adaptation intensity and interspecific competition are high, which may explain no complexity-stability relationships revealed in empirical data. Finally, we deduce that foraging adaptation alters positive and/or negative feedback loops to affect the stability of real food webs.</span></p>

An Evaluation of the Hydrolytic Stability of Selected Experimental Dental Matrices and Composites

<p>Materials with potential use as dental restoration should be evaluated in an aggressive environment. Such accelerated aging is widely used in other industries and allows the assessment of service life. In the presented study, three neat resins (UDMA/Bis-GMA/TEGDMA 70/10/20 wt.%, UDMA/Bis-GMA/TEGDMA 40/40/20 wt.% and UDMA/Bis-EMA/TEGDMA 40/40/20 wt.%) and three composites based on these matrices were tested before and after aging protocols (I-7500 cycles, 5 &deg;C and 55 &deg;C, water and 7 days, 60 &deg;C, 0.1&nbsp;M NaOH; II-5 days, 55 &deg;C, water and 7 days, 60 &deg;C, 0.1 M NaOH). Flexural strength (FS), diametral tensile strength (DTS) and hardness (HV) were determined. Applied aging protocols resulted in a decrease in the value of the FS, DTS and HV. Larger changes were noticed for the neat resins. Materials in which the content of bis-GMA was lower or substituted by bis-EMA showed better resistance to degradation. The choice of mixtures with monomers characterized by lower sorption values may favorably affect hydrolytic stability. It was shown that for composites there was a drastic decrease in hardness, which suggests a more superficial effect of the used protocols. However, degradation of the surface layer can result in a growing problem over time given that the mastication processes are an inherent element in the oral environment.</p> <p>&nbsp;</p>

Stability of the next-to-tribimaximal mixings under radiative corrections with the variation of SUSY breaking scale

<pre>We analyse the stability of the next-to-tribimaximal mixings ($NTBM$) under radiative corrections with the variation of SUSY breaking scale ($m_s$) for both normal and inverted hierarchical cases at the fixed value of seesaw scale $M_R = 10^{15}$ GeV and $\tan \beta = 30$. All the neutrino oscillation parameters receive varying radiative corrections irrespective of the $m_s$ values at the electroweak scale, which are all within $3\sigma$ range of the latest global fit data. NH case is found to be more stable than IH case for all four cases.</pre>

FTD-tau S320F mutation stabilizes local structure and allosterically promotes amyloid motif-dependent aggregation

<p>Amyloid deposition of the microtubule-associated protein tau is a unifying theme in a multitude of neurodegenerative diseases. Disease-associated missense mutations in tau are associated with frontotemporal dementia (FTD) and enhance tau aggregation propensity. However, the molecular mechanism of how mutations in tau promote tau assembly into amyloids remains obscure. There is a need to understand how tau folds into pathogenic conformations to cause disease. Here we describe the structural mechanism for how an FTD-tau S320F mutation drives spontaneous aggregation. We use recombinant protein and synthetic peptide systems, computational modeling, cross-linking mass spectrometry, and cell models to investigate the mechanism of spontaneous aggregation of the S320F FTD-tau mutant. We discover that the S320F mutation drives the stabilization of a local hydrophobic cluster which allosterically exposes the <sup>306</sup>VQIVYK<sup>311</sup> amyloid motif. We identify a suppressor mutation that reverses the S320F aggregation phenotype through the reduction of S320F-based hydrophobic clustering <em>in vitro</em> and in cells. Finally, we use structure-based computational design to engineer rapidly aggregating tau sequences by optimizing nonpolar clusters in proximity to the S320 site revealing a new principle governing the regulation of tau aggregation. We uncover a mechanism for regulating aggregation that balances transient nonpolar contacts within local protective structures or in longer-range interactions that sequester amyloid motifs. The introduction of a pathogenic mutation redistributes these transient interactions to drive spontaneous aggregation.&nbsp; We anticipate that more profound knowledge of this process will permit control of tau aggregation into discrete structural polymorphs to aid the design of reagents that can detect disease-specific tau conformations.</p>

Stability of oral and faecal microbiome at room temperature: impact on diversity

<p>When collecting oral and faecal samples for large epidemiological microbiome studies, optimal storage conditions such as immediate freezing, are not always feasible. It is fundamental to study the impact of temporary room temperature (RT) storage and shipping on the microbiome diversity obtained in different types of samples. We performed a pilot study aimed to validate the sampling protocol based on the viability of the 16S rRNA gene sequencing in microbiome samples.</p> <p>Faecal and oral samples from five participants were collected and preserved in different conditions: i) 70% ethanol; ii) in a FIT tube for stool samples; and iii) in a chlorhexidine solution for oral wash samples. Four aliquots were prepared per sample, which were stored at RT, and frozen at days 0, 5, 10 and 15, respectively.</p>

PDB files of "Characterization of ligand-induced thermal stability of the human organic cation transporter 2 (OCT2)"

<p>The predicted alphafold&nbsp;structure of OCT2 (Uniprot O15244) was embedded in a DPPC model membrane, solvated with TIP3P water and subjected to 250 ns molecular dynamics simulation using the Desmond module of Schroedingers Drug Discovery Suite. The last 150 ns were used for clustering and the cluster with most of the members (cluster0) was selected as a model for OCT2. Based on that (still predicted, but optimised) model we performed docking with various substrates for which we also provide the obtained docking pose.&nbsp;See Publication in Frontiers in Pharmacology (DOI:&nbsp;10.3389/fphar.2023.1154213)&nbsp;for details and results.</p>

Community reorganization stabilizes freshwater ecosystems in intensively managed agricultural fields

<ol> <li>Sustainable intensification may depend on associating precision farming with the harnessing of ecological principles in crop fields and with integrating farms and non-farmed land in productive landscapes. Small wetlands could play an important role in both pursuits for having high per-unit-area rates of element cycling and species richness while deeply penetrating crop fields. However, their potential for ecosystem service provisioning is unlikely to be met if land management intensification promotes ecosystem destabilization in biomass production.Β </li> <li>We tested the consequences of land use intensification on various dimensions of freshwater ecosystem stability by means of a large-scale field experiment converting extensive pastures to intensive pastures and sugarcane plantations in Southeastern Brazil. Nested within experimental plots were 4,000-L mesocosms simulating ponds and puddles commonly found in productive landscapes. Mesocosms were monitored for basic physico-chemical parameters, nutrients, pesticides, phytoplankton standing crop, and the spontaneously colonizing biodiversity.</li> <li>3. Despite severe environmental change, the stability of sugarcane communities was no different from that of extensive and intensive pastures. This occurred because the local extinction of a sensitive top dragonfly predator following the application of vinasse and insecticide was compensated by colonization of a suite of more tolerant invertebrate mesopredators such as beetles and bugs. Community stability tended to increase with biomass asynchrony and species richness, evidencing a portfolio effect of biodiversity. Unfortunately, the species richness necessary to stabilize biomass production is unlikely to be available in many sugarcane fields and several other row crops.</li> <li> <em>Synthesis and applications.</em> Ponds and puddles could be effective centers of irradiation of ecosystem service provisioning in agricultural fields in terms of pest control; nutrient accumulation, cycling, and export back to fields; and habitat and stepping stones for freshwater biodiversity. However, the impoverished biodiversity that results from a combination of harsh local conditions and spatial isolation renders pond communities inherently unstable. Given the unlikely, immediate reduction in agrochemical use in much of the intensively managed crop area, a combination of large, protected source wetlands at the margin of fields and small constructed or naturally forming ponds and puddles in plantations could contribute to sustainable intensification.</li> </ol>

Using Rice Husk Ash (RHA) as Stabilizing Agent for Problematic Subgrade Soils and Embankments

<p>Arkansas produces the most of the rice in the United States. About 20% of poddy is rice husk (RH), which is burnt under controlled conditions to produce rice rusk ash (RHA). The RHA is considered an environmental hazard and a significant challenge for rice millers. However, RHA is rich in pozzolanic material, which is mainly silica. In this study, RHA is used to stabilize poor soils. Another commonly used stabilizer, hydrated lime (HL), has also been evaluated for comparison purposes. Thus, this study aimed to determine the optimum percentages of RHA, HL, or a combination of these two agents by evaluating selective physical, mechanical, and chemical properties of four subgrade soils (two from Arkansas and two from Oklahoma). Various amounts (by mass of soil) of RHA (3, 6, and 9%), HL (1, 3, and 5%), and RHA+HL have been utilized to know their optimum dosages. Routine tests including the Atterberg limits, Modified proctor, pH, California Bearing Ratio (CBR), unconfined compressive strength (UCS), free swell, and one-direction shrinkage were conducted for untreated and treated soils. Further, scanning electron microscopy (SEM) with X-ray diffractometer tests were performed to evaluate the changes in microstructure due to stabilization. Treated soils show a significant improvement in UCS and CBR data. Additionally, the free swell test also indicates a reduced swelling of treated soils. The optimum dosages of RHA and HL were found to be 6% and 3%, respectively. When these two agents were used together, a blend of 4%RHA and 1%HL was found to be the most effective in improving engineering properties&nbsp; The findings of this research can significantly benefit the construction industry in Arkansas by reducing costs associated with traditional soil stabilization methods and finding a sustainable and environmentally way of using RHA.</p>

Data from: HIV-1 capsid stability enables inositol phosphate-independent infection of target cells and promotes integration into genes

<p>The mature HIV-1 capsid is stabilized by host and viral determinants. The capsid protein CA binds to the cellular metabolites inositol hexakisphosphate (IP6) and its precursor inositol (1, 3, 4, 5, 6) pentakisphosphate (IP5) to stabilize the mature capsid. In target cells, capsid destabilization by the antiviral compounds lenacapavir and PF74 reveals an HIV-1 infectivity defect due to IP5/IP6 (IP5/6) depletion.Β  To test whether intrinsic HIV-1 capsid stability and/ or host factor binding determines HIV-1 insensitivity to IP5/6 depletion, a panel of CA mutants was assayed for infection of IP5/6-depleted T cells and wildtype cells.Β  Four CA mutants with unstable capsids exhibited dependence on host IP5/6 for infection and reverse transcription (RTN).Β  Adaptation of one such mutant, Q219A, by spread in culture resulted in Vpu truncation and a capsid three-fold interface mutation, T200I.Β  T200I increased intrinsic capsid stability as determined by <em>in vitro</em> uncoating of purified cores and partially reversed the IP5/6-dependence in target cells for each of the four CA mutants. T200I further rescued the changes to lenacapavir sensitivity associated with the parental mutation. The premature dissolution of the capsid caused by the IP5/6-dependent mutations imparted a unique defect in integration targeting that was rescued by T200I.Β  Collectively, these results demonstrate that T200I restored other capsid functions after RTN for the panel of mutants. Thus, the hyperstable T200I mutation stabilized the instability defects imparted by the parental IP5/6-dependent CA mutation. The contribution of Vpu truncation to mutant adaptation was linked to BST-2 antagonization, suggesting that cell-to-cell transfer promoted replication of the mutants.Β  We conclude that interactions at the three-fold interface are adaptable, key mediators of capsid stability in target cells and are able to antagonize even severe capsid instability to promote infection.</p>

Physico-chemical characterization of sterile citrated stabilized Au nanoparticles by XPS / HAXPES / SEM

<p>Here a dataset of XPS, HAXPES and SEM measurements for the physico-chemical characterization of nanoparticles is presented. The measurements are part of the H2020 project &ldquo;NanoSolveIT&rdquo;.</p> <p>Sterile citrated stabilized Au nanoparticles with a diameter of approx. 30 nm were synthesized under aseptic conditions at Mintek, South Africa (EMR-Identifier&nbsp;in the project: ERM00000582).</p> <p>Prior to the measurements, the samples were prepared from the solution as drop cast on Si wafers. &nbsp;First SEM measurements were performed, followed by XPS and HAXPES measurements on the same samples. Here at first survey spectra were recorded followed by high resolution spectra. For XPS / HAXPES several Si-wafers were mounted together on one platen. The platen was left in the intro chamber of the instrument for several hours before the measurement started.</p> <p>Equipment:</p> <p>SEM images were acquired with a <em>Supra 40 </em>(Zeiss) SEM.</p> <p>For X-ray spectroscopy experiments, a combined XPS / HAXPES spectrometer (<em>Quantes </em>from ULVAC-PHI) was used, where XPS is measured at 1486.6 eV (monochromatic Al K&alpha; source) and HAXPES at 5414.9 eV (monochromatic Cr K&alpha; source). Here it is possible to perform the measurements at the exact same position.</p> <p>Data:</p> <p>For SEM, the data are given in .tif format. For XPS / HAXPES the raw data are given as .spe (PHI format) and .npl (VAMAS format) files. Data from: Effects of temperature and obsidian content on the friction and stability of simulated basalt gouges: Implications for shallow moonquakes

<p><span>Basalt is a major component of crust on both the Earth and Moon. Mineral composition and temperature influence frictional instability and thus the potential for seismicity on basaltic faults. We performed velocity-stepping shear experiments on basalt gouges at a confining pressure of 100 MPa, temperatures in the range 100-400Β°C and with varied obsidian contents of 0-100 wt.% under wet/dry conditions to investigate the frictional strength and stability of basaltic faults. We observe a transition from velocity-neutral to velocity-weakening behaviors with increasing obsidian content. The frictional stability response of the mixed obsidian/basalt gouges is characterized by a transition from velocity-strengthening to velocity-weakening at 200Β°C and another transition to velocity-strengthening at &gt;300Β°C. Conversely, frictional strengths of the obsidian-bearing gouges are insensitive to temperature and wet/dry conditions. These results suggest that obsidian content dominates the potential seismic response of basaltic faults with the effect of temperature controlling the range of seismogenic depths. These observations contribute to a better understanding of the nucleation mechanism of shallow moonquakes and also seismicity on terrestrial faults in basalt.</span></p>

Africa's ecosystems exhibit a tradeoff between resistance and stability following disturbances

<p class="MsoNormal">Environmental disturbances may prevent ecosystems from consistently performing their critical ecological functions. Two important properties of ecosystems are their resistance and stability, which respectively reflect their capacities to withstand and recover from disturbance events (e.g., droughts, wildfires, pests, etc.). Theory suggests that resistant and stable ecosystems possess opposing characteristics, but this has seldom been established across diverse ecosystem attributes or broad spatial scales. Here, we compare the resistance and stability of &gt;1,000 protected area ecosystems in Africa to disturbance-induced losses in primary productivity from 2000-2019. We quantitatively evaluated each ecosystem such that following disturbances, an ecosystem is more resistant if it experiences lower-magnitude losses in productivity, and more stable if it returns more rapidly to pre-disturbance productivity levels. To compare the characteristics of resistant versus stable ecosystems, we optimized random forest models that use ecosystem attributes (representing their climatic and environmental conditions, plant and faunal biodiversity, and exposure to human impacts) to predict their resistance and, separately, stability values. We visualized each attribute's relationship with resistance and stability after accounting for all other attributes in the model framework. Ecosystems that are more resistant to disturbances are less stable, and vice versa. The ecosystem attributes with the most predictive power in our models all exhibit contrasting relationships with resistance versus stability. Notably, highly resistant ecosystems are generally more arid and exhibit high habitat heterogeneity and mammalian biodiversity, while highly stable ecosystems are the opposite. We discuss the underlying mechanisms through which these attributes engender resistance or, conversely, stability. Our findings suggest that resistance and stability are fundamentally opposing phenomena. A balance between the two must be struck if ecosystems are to maintain their identity, structure, and function in the face of environmental change.</p>
