The role of cell-envelope synthesis for envelope growth and cytoplasmic density in Bacillus subtilis
<p>All cells must increase their volumes in response to biomass growth to maintain intracellular mass density within physiologically permissive bounds. Here, we investigate the regulation of volume growth in the Gram-positive bacterium <em>Bacillus subtilis</em>. To increase volume, bacteria enzymatically expand their cell envelopes and insert new envelope material. First, we demonstrate that cell-volume growth is determined indirectly, by expanding their envelopes in proportion to mass growth, similarly to the Gram-negative <em>Escherichia coli</em>, despite their fundamentally different envelope structures. Next, we studied, which pathways might be responsible for robust surface-to-mass coupling: We found that both peptidoglycan synthesis and membrane synthesis are required for proper surface-to-mass coupling. However, surprisingly, neither pathway is solely rate-limiting, contrary to wide-spread belief, since envelope growth continues at a reduced rate upon complete inhibition of either process. To arrest cell-envelope growth completely, the simultaneous inhibition of both envelope-synthesis processes is required. Thus, we suggest that multiple envelope-synthesis pathways collectively confer an important aspect of volume regulation, the coordination between surface growth and biomass growth.</p>

A thematic synthesis on the adoption of regression testing techniques in Android projects
<p>In software testing, employing regression techniques is a viable strategy to deal with the complexity and the constant evolution of applications since its primary goal is to ensure that changes made between versions do not change the system’s behavior. Although the literature has dedicated efforts to developing new regression testing techniques suitable for the Android mobile platform, studies are limited concerning demonstrating which techniques software developers employ in practice. This study aims to report on a thematic synthesis of adopting regression testing techniques in Android projects. The research encompassed four stages: (i) conducting a structured literature review on regression testing techniques for the Android platform, (ii) carrying out an expert survey, (iii) conducting interviews with industry professionals, and (iv) building a thematic synthesis. The thematic synthesis presented a model from analyzing the results obtained in this multimethod study on regression testing techniques. With such a stud, we could present empirical evidence on how professionals perform regression testing in Android projects, identify the commonly used regression testing techniques, and leverage the requirements for automating Android applications through regression testing.</p>

An in vivo stable isotope labeling method to investigate individual matrix protein synthesis, ribosomal biogenesis, and chondrocyte proliferation in murine articular cartilage
<p>These experiments have used a stable-isotope method using <em>in vivo</em> deuterium oxide labeling and mass spectrometry to measure protein concentration, protein half-life, cell proliferation, and ribosomal biogenesis in a single sample of murine articular cartilage. We hypothesized that a 60-day labeling period would capture age-related declines in cartilage matrix protein content, protein synthesis rates, and chondrocyte proliferation. Knee cartilage was isolated from 25- and 90-week-old female C57BL/6J mice treated with deuterium oxide for 15, 30, 45 and 60 days. We measured protein abundance and half-lives using high resolution accurate mass spectrometry (HRAM) and d2ome data processing software. </p>
Synthesis and scaled-up of standard and active Belite-Alite-Ye'elimite clinker (BAY)
<p><strong>ABSTRACT:</strong> Clinkers with the coexistence of belite-alite-ye´elimite (BAY) are considered as eco-friendly cements due to the reduced CO<sub>2</sub> emissions released during their manufacture. All BAY clinkering parameters have been optimized: raw materials, mineralizers/activators, clinkering conditions and scaled-up methodology. Two types of clinkers were prepared and scaled-up to have ~2 kg: i) standard-BAY (28.4 wt% of b-belite, 41.5 wt% of alite and 17.4 wt% of ye'elimite) by adding CaF<sub>2</sub> and ZnO to the raw mixture; and ii) active-BAY (18.1 wt% of α'<sub>H</sub>-belite, 42.1 wt% of b-belite, 15.8(2) wt% of alite, and 7.7(2) wt% of ye'elimite) by using optimized amounts of CaF<sub>2</sub>, ZnO and B<sub>2</sub>O<sub>3</sub>. The microscopy[P1] study has revealed that primary particle size of belite is larger (~10 µm) in the active clinker than in the standard one (~5 µm). Moreover, ferrite and alite have incorporated most of the Zn and F in their structures in both clinkers, while belite (b and α’<sub>H</sub> forms) has preferentially incorporated sulphur.</p>

INHIBITION OF NITRIC OXIDE SYNTHESIS PROMOTES INCREASED MORTALITY DESPITE REDUCTION OF PARASITEMIA IN Plasmodium berghei-INFECTED MICE
<p><strong>Backgrounds:</strong> Nitric oxide (NO) is an important mediator molecule in inflammatory processes, but its role in the pathophysiology of malaria is still uncertain.</p> <p><strong>Methods:</strong> To investigate the NO synthesis inhibition on the oxidative changes induced by <em>Plasmodium berghei </em>infection in mice, malaria was induced in 150 animals, of which 75 animals were treated with NO inhibitor L-NAME; the remaining are sham controls. All animals underwent euthanasia after 1, 5, 10, 15 or 20 days after the infection for the collection of lungs, brain, and blood. Parasitemia was determined and the survival of the animals evaluated. Tissue samples were assayed for nitrites and nitrates (NN), thiobarbituric acid reactive substances (TBARS), and total Trolox equivalent antioxidant capacity (TEAC). A histopathological study was performed.</p> <p><strong>Results:</strong> Mortality rates in the L-NAME were always higher in relation to the controls. In brains, NN was lower in groups L-NAME. Parasitemia and its progression rate were greater in control groups. Data from: Forestry impacts on stream flows and temperatures: A quantitative synthesis of paired catchment studies across the Pacific salmon range
<p>Forestry is pervasive across temperate North America and may influence aquatic environmental conditions such as flows and temperatures, as well as important species such as Pacific salmon (<em>Oncorhynchus</em> spp.). While there have been many large-scale forestry experiments using paired catchment designs, these studies have yet to be quantitatively synthesized. Thus, it remains unclear whether forestry impacts are consistent, context-dependent, or unpredictable. This study aims to quantitatively synthesize forestry impacts on streamflow and temperature, through a systematic review and synthesis of paired catchment studies across the range of Pacific salmon. Specifically, we investigated whether generalizable relationships exist between forestry intensity (percent watershed harvested) and impacts to streamflow and temperature. We also examined whether watershed features (climate, hydrology, lithology) and harvest method mediated forestry impacts. We extracted information from 35 unique paired-catchments from California to Alaska. Forestry had strong impacts on peak and low flows and maximum summer water temperatures, but responses were quite variable. Across all catchments, forestry elevated peak flows ~20% (n = 31 catchments), reduced low flows ~25% (n = 13 catchments), and increased maximum summer temperatures ~15% (n = 35 catchments) on average. However, these impacts were variable and were not predictable based on forestry intensity, thus broader stressor-response relationships were not supported. Forestry impacts on peak flows and maximum summer temperatures varied spatially. Peak flow impacts increased with increased northward latitude and temperature impacts decreased with eastward longitude. However, the magnitude of impacts were unrelated to other watershed attributes, which included climate (precipitation and aridity), rain vs. snow hydrology, elevation, and bedrock lithology. Harvest method and riparian buffer presence also had no detected effects on forestry impacts across studies and statistical models explained a low proportion of variation overall. Collectively, our results indicate that forestry can have substantial impacts on key environmental conditions; however, the magnitude of impact was variable and could not be clearly linked to easily-measured watershed characteristics. This implies that forestry impacts are not broadly predictable. Probabilistic risk models based on distributions of potential impacts may therefore be more useful for watershed management in data-poor situations.</p>
