GDPR and the principle of purpose limitation in connecting Data Spaces demonstrators

<p>The presentation focuses on legal challenges arising from the GDPR when connecting Data Spaces using the Demonstrators. Special attention is given to the principle of purpose limitation of the GDPR. The Demonstrators serve a crucial role in connecting the data spaces of the NFDI and GAIA-X. The data in the two data spaces is gathered and used for usually completely different purposes. Thus, when connecting the spaces,&nbsp; the data may be processed for completely different purposes. The GDPR &ndash; when applicable &ndash; limits legitimate purposes of such further processing. Even if a purpose is legitimate, the GDPR imposes different obligations on controllers and processors. The presentation will dive into the applicability of the GDPR to the demonstrators, the roles of controller and processor regarding the demonstrators and will discuss the principle of purpose limitation and the obligations connected to it.</p>

Target cell-specific synaptic dynamics of excitatory to inhibitory neuron connections in supragranular layers of human neocortex

<p>Rodent studies have demonstrated that synaptic dynamics from excitatory to inhibitory neuron types are often dependent on the target cell type. However, these target cell-specific properties have not been well investigated in human cortex, where there are major technical challenges in reliably obtaining healthy tissue, conducting multiple patch-clamp recordings on inhibitory cell types, and identifying those cell types. Here, we take advantage of newly developed methods for human neurosurgical tissue analysis with multiple patch-clamp recordings, <em>post-hoc</em> fluorescent <em>in situ</em> hybridization (FISH), machine learning-based cell type classification, and prospective GABAergic AAV-based labeling to investigate synaptic properties between pyramidal neurons and PVALB- vs. SST-positive interneurons. We find that there are robust molecular differences in synapse-associated genes between these neuron types, and that individual presynaptic pyramidal neurons evoke postsynaptic responses with heterogeneous synaptic dynamics in different postsynaptic cell types. Using molecular identification with FISH and classifiers based on transcriptomically identified PVALB neurons analyzed by Patch-seq, we find that PVALB neurons typically show depressing synaptic characteristics, whereas other interneuron types including SST-positive neurons show facilitating characteristics. Together, these data support the existence of target cell-specific synaptic properties in human cortex that are similar to rodents, thereby indicating evolutionary conservation of local circuit connectivity motifs from excitatory to inhibitory neurons and their synaptic dynamics.</p>

Woodland expansion in the presence of deer: 30 years of evidence from the Cairngorms Connect landscape restoration partnership

<ol> <li>Restoring native woodlands to areas where they have been lost is a key element for tackling the nature and climate crises. Natural regeneration offers the potential to achieve this cheaply and at scale, but browsing ungulates like deer can inhibit this or alter the pattern of regeneration. This issue is particularly pronounced in the Scottish Highlands, a heavily deforested region with high deer numbers.</li> <li>We describe the work of the 600 km<sup>2</sup> landscape restoration partnership, Cairngorms Connect, in speeding up natural woodland expansion. We use 30 years of regeneration monitoring to show a consistent, large-scale expansion of native woodland, largely through natural regeneration alongside deer culling, without the use of fences. This was achieved across the partnership, despite differing management histories and land-managing organisations (comprising two statutory agencies, one private landowner and one non-governmental organisation).</li> <li>During peak periods of woodland expansion, the area of new woodland (i.e. exceeding 100 trees per hectare) increased by 1.2%, 1.7%, 2.7% and 6.0% annually in the four landholdings' regeneration zones, equating to a total of approximately 164 ha annually of new woodland.</li> <li>Natural regeneration is however patchy and hard to predict. Higher levels of management intervention may be needed to increase species that are rarer, more palatable or further from seed sources; we recommend long-term field trials to inform this, such as those underway in Cairngorms Connect. Further research should develop techniques for remote-sensing of woodland expansion, verified against field data, combined with development of process-based models to enable us to predict the outcomes of different management scenarios.</li> <li> <em>Synthesis and applications:</em> We show that collaborative deer management across multiple adjoining landholdings can achieve rapid landscape-scale native woodland expansion with minimal need for planting or fencing. Our results show the power of monitoring regeneration directly, to inform deer management for an area. We demonstrate that by uniting over a shared vision, organisations with differing management approaches and histories can build understanding alongside landscape-scale ecological restoration.</li> </ol>

Low-coverage whole genome sequencing for highly accurate population assignment: Mapping migratory connectivity in the American Redstart (Setophaga ruticilla)

<p>Understanding the geographic linkages among populations across the annual cycle is an essential component for understanding the ecology and evolution of migratory species and for facilitating their effective conservation. While genetic markers have been widely applied to describe migratory connections, the rapid development of new sequencing methods, such as low-coverage whole genome sequencing (lcWGS), provides new opportunities for improved estimates of migratory connectivity. Here, we use lcWGS to identify fine-scale population structure in a widespread songbird, the American Redstart (<em>Setophaga</em> <em>ruticilla</em>), and accurately assign individuals to genetically distinct breeding populations. Assignment of individuals from the nonbreeding range reveals population-specific patterns of varying migratory connectivity. By combining migratory connectivity results with demographic analysis of population abundance and trends, we consider full annual cycle conservation strategies for preserving numbers of individuals and genetic diversity. Notably, we highlight the importance of the Northern Temperate-Greater Antilles migratory population as containing the largest proportion of individuals in the species. Finally, we highlight valuable considerations for other population assignment studies aimed at using lcWGS. Our results have broad implications for improving our understanding of the ecology and evolution of migratory species through conservation genomics approaches.</p>

Online Workshop for Supporting Inclusive and Sustainable Research Infrastructure for Systematics by Connecting Scientists and their Specimens

<p>The project, &quot;Supporting inclusive and sustainable research infrastructure for systematics (SISRIS) by connecting scientists and their specimens&quot; is supported by a&nbsp;<a href="https://www.nsf.gov/awardsearch/showAward?AWD_ID=2247631&amp;HistoricalAwards=false">conference grant from the US National Science Foundation</a>&nbsp;to Dr. Andrea Weeks (George Mason University; Ted R. Bradley Herbarium), and Drs. Shawn Zeringue-Krosnick and Twanelle Majors (Tennessee Tech University; Hollister Herbarium). It supports two US-based workshops and symposia at the 2023 annual meetings of the&nbsp;<a href="https://www.sebiologists.org/asb-2023.html">Association of Southeastern Biologists</a>&nbsp;and&nbsp;<a href="https://2023.botanyconference.org/">Botany 2023</a>. The purpose of the work is to share new practices that improve attribution of natural history specimens and highlight research advances in systematics that can be made as a consequence of improved people-data within biodiversity informatics databases. This four hour workshop&nbsp;was held Sunday August 6, 2023 in an online format to accommodate participants who could not attend the in-person event at Botany 2023&nbsp;on July 23, 2023 in Boise, Idaho. Exploring the connection between autophagy and heat-stress tolerance in Drosophila melanogaster

<p>Mechanisms aimed at recovering from heat-induced damages are closely associated with the ability of ectotherms to survive exposure to stressful temperatures. Autophagy, a ubiquitous stress-responsive catabolic process, has recently gained renewed attention as one of these mechanisms. By increasing the turnover of cellular structures as well as the clearance of long-lived protein and protein aggregates, the induction of autophagy has been linked to increased tolerance to a range of abiotic stressors in diverse ectothermic organisms. However, whether a link between autophagy and heat-tolerance exists in insect models remains unclear despite broad ecophysiological implications thereof. Here, we explored the putative association between autophagy and heat-tolerance using <em>Drosophila</em> <em>melanogaster</em> as a model. We hypothesized that (i) heat-stress would cause an increase of autophagy in flies' tissues, and (ii) rapamycin exposure would trigger a detectable autophagic response in adults and increase their heat-tolerance. In line with our hypothesis, we report that flies exposed to heat-stress present signs of protein aggregation and appear to trigger an autophagy-related homoeostatic response as a result. We further show that rapamycin feeding causes the systemic effect associated with target of rapamycin (TOR) inhibition, induces autophagy locally in the fly gut, and increases the heat-stress tolerance of individuals. These results argue in favour of a substantial contribution of autophagy to the heat-stress tolerance mechanisms of insects.</p>

Data for: Ecosystem connectivity and configuration can mediate instability at a distance in metaecosystems

<ol> <li><span>Ecosystems are connected by flows of nutrients and organisms. Changes to connectivity and nutrient enrichment may destabilise ecosystem dynamics far from the nutrient source.</span></li> <li><span>We used gradostats to examine the effects of trophic connectivity (movement of consumers and producers) versus nutrient-only connectivity on the dynamics of <em>Daphnia</em> <em>pulex</em> (consumers) and algae (resources) in two metaecosystem configurations (linear vs. dendritic). </span></li> <li><span>We found that <em>Daphnia</em> peak population size and instability (coefficient of variation; CV) increased as distance from the nutrient input increased, but these effects were lower in metaecosystems connected by all trophic levels compared to nutrient-only connected systems and/or in dendritic compared to linear systems. </span></li> <li><span>We examined the effects of trophic connectivity (i.e. both trophic levels move rather than one or the other) using a generic model to qualitatively assess whether the expectations align with the ecosystem dynamics we observed. </span></li> <li><span>Analysis of our model shows that increased <em>Daphnia</em> population sizes and fluctuations in consumer-resource dynamics are expected with nutrient connectivity, with this pattern being more pronounced in linear rather than dendritic systems. </span></li> <li><span>These results confirm that connectivity may propagate and even amplify instability over a metaecosystem to communities distant from the source disturbance, and suggest a direction for future experiments, that recreate conditions closer to those found in natural systems.</span></li> </ol>
