Theory predicts that males have a limited amount of resources to invest in reproduction, suggesting a trade-off between traits that enhance mate acquisition and those enhancing fertilization success. Here we investigate the relationship between pre- and post-copulatory investment by comparing the mating behavior and reproductive morphology of four European and five North American populations of the dung fly Sepsis punctum (Diptera) that display a reversal of sexual size dimorphism (SSD). We show that the geographic reversal in SSD between the continents (male-biased in Europe, female-biased in North America) is accompanied by differential investment in pre- versus post-copulatory traits. We find higher re-mating rates in European populations, where larger males acquire more matings and consequently have evolved relatively larger testes and steeper hyper-allometry with body size. American populations, in sharp contrast, display much reduced, if any, effect of body size on those traits. Instead, North American males demonstrate an increased investment in mate acquisition prior to copulation, with more mounting attempts and a distinctive abdominal courtship display that is completely absent in Europe. When controlling for body size, relative female spermathecal size is similar on both continents, so we find no direct evidence for the co-evolution of male and female internal reproductive morphology. By comparing allopatric populations of the same species that apparently have evolved different mating systems and consequently SSD, we thus indirectly demonstrate differential investment in pre- vs. post-copulatory mechanisms increasing reproductive success.

Data from: Blood-brain barrier deterioration and hippocampal gene expression in polymicrobial sepsis: an evaluation of endothelial MyD88 and the vagus nerve

Systemic infection can initiate or exacerbate central nervous system (CNS) pathology, even in the absence of overt invasion of bacteria into the CNS. Recent epidemiological studies have demonstrated that human survivors of sepsis have an increased risk of long-term neurocognitive decline. There is thus a need for improved understanding of the physiological mechanisms whereby acute sepsis affects the CNS. In particular, MyD88-dependent activation of brain microvascular endothelial cells and a resulting loss of blood-brain barrier integrity have been proposed to play an important role in the effects of systemic inflammation on the CNS. Signaling through the vagus nerve has also been considered to be an important component of CNS responses to systemic infection. Here, we demonstrate that blood-brain barrier permeabilization and hippocampal transcriptional responses during polymicrobial sepsis occur even in the absence of MyD88-dependent signaling in cerebrovascular endothelial cells. We further demonstrate that these transcriptional responses can occur without vagus nerve input. These results suggest that redundant signals mediate CNS responses in sepsis. Either endothelial or vagus nerve activation may be individually sufficient to transmit systemic inflammation to the central nervous system. Transcriptional activation in the forebrain in sepsis may be mediated by MyD88-independent endothelial mechanisms or by non-vagal neuronal pathways.

Data from: New criteria for sepsis-induced coagulopathy (SIC) following the revised sepsis definition: a retrospective analysis of a nationwide survey

OBJECTIVE: Recent clinical studies have shown that anticoagulant therapy might be effective only in specific at-risk subgroups of patients with sepsis and coagulation dysfunction. The definition of sepsis was recently modified and old scoring systems may no longer be proper for the diagnosis of sepsis-associated coagulopathy. The aim of this study was to evaluate prognostic factors in patients diagnosed as sepsis and coagulopathy according to the new sepsis definition and assess their accuracy in comparison to existing models. DESIGN, SETTING AND SUBJECTS: The data set was obtained from a post-marketing survey examining recombinant human soluble thrombomodulin and offered by Japanese Society on Thrombosis and Hemostasis. A total of 1498 patients with sepsis and coagulopathy complications who were treated with recombinant thrombomodulin were analyzed in this study. MAIN OUTCOME MEASURES: The platelet count, prothrombin time (PT) ratio, fibrinogen and fibrin degradation products (FDP), systemic inflammatory response syndrome (SIRS) score, and sequential organ failure assessment (SOFA) score obtained just before the start of treatment were examined in relation to the 28-day mortality rate. RESULTS: The platelet count, PT ratio, and total SOFA were independent predictors of a fatal outcome in a logistic regression model. A sepsis-induced coagulation (SIC) score was defined using the 3 above-mentioned variables with a positivity threshold of 4 points or more. The SIC score predicted higher 28-day mortality rate compared to the Japanese Association for Acute Medicine (JAAM)-DIC score (38.4% vs. 34.7%). CONCLUSIONS: The SIC scoring is based on readily available parameters, is easy to calculate and owns a high predictive value for 28-day mortality. Future studies are warranted to evaluate whether the SIC score may guide the decision to initiate anticoagulant therapy.

Berberine protects against sepsis-related acute lung injury in rats via the upregulation of the PPAR - Υ signaling pathway, with amelioration at the cellular level - A functional, immune-histological and biochemical study

<p>Aim: To assess the therapeutic effects of berberine in experimentally induced lung sepsis.  Examining its effects on selected cytokines, gene and protein expression and the histopathological findings.</p> <p>Results: Berberine significantly reduced the wet/dry lung ratio, the BALF protein, cell, and neutrophils percentage &amp; the BALF cytokines levels. In addition, pretreatment with berberine decreases the myeloperoxidase and MDA levels and decreases gene expression of NFKB, MCP -1 and the ICAM - 1 by RT - PCR analysis. Thus, suggesting an antioxidant and anti-inflammatory mode of the action. Western blot analysis revealed increased PPAR – Υ expression in the berberine pretreated group compared to the CLP group. Histopathological examination revealed improved histopathological parameters, in the berberine pretreated group, compared to the CLP group.</p> <p>Conclusion: Berberine improves the outcome in sepsis via antioxidant and anti-inflammatory effects, modulation of cytokine levels, upregulation of PPAR - Υ protein expression, and histopathological amelioration.</p>
