Data from: Cyanobacteria maintain constant protein concentration despite genome copy-number variation
The cyanobacterium Synechococcus elongatus PCC 7942 has multiple copies of its single chromosome, and the copy number varies in individual cells, providing an ideal system to study the effect of genome copy-number variation on cell size and gene expression. Using single-cell fluorescence imaging, we found that protein concentration remained constant across individual cells regardless of genome copy number. Cell volume and the total protein amount from a single gene were both positively, linearly correlated with genome copy number, suggesting that changes in cell volume play an important role in buffering genome copy-number variance. This study provides a quantitative examination of gene expression regulation in cells with variable genome copies and sheds light on the compensation mechanisms for variance in genome copy number.
Data from: In situ glacial survival maintains high genetic diversity of Mussaenda kwangtungensis on continental islands in subtropical China
<p>Generally, island populations are predicted to have less genetic variation than their mainland relatives. However, there has been exceptions, indicating that the relationships were impacted by several factors, e.g historical processes. In the present study, we chose a group of subtropical islands located in South China as the study system, which are quite younger and much closer to the mainland than most of the previous studied island systems, to test the hypothesis that<i> in situ</i> glacial survival contributes to high levels of genetic diversity in island populations. We conducted a comparison of genetic variation between 12 island and 11 nearby mainland populations of <i>Mussaenda kwangtungensis</i> using eleven nuclear microsatellite and three chloroplast markers, evaluated effects of the island area and distance to mainland on genetic diversity of island populations, and simulated the potential distribution over the past by ecological niche modelling, together with the genetic data to detect the role of islands during the glacial periods. The island populations displayed comparable levels of genetic diversity and differentiation with mainland populations, overall high levels of unique polymorphisms, and the greatest values of specific within-population genetic diversity. No significant correlation was detected between genetic diversity of island populations and distance to mainland, as well as area of islands, except that allelic richness was significantly positively correlated with the area of islands. Nuclear microsatellites revealed two main clusters, largely corresponding to islands and inland populations, which divergence dated to a time of island formation by ABC analysis. Ecological Niche Modelling predicted a highly climatic suitability on islands during the Last Glacial Maximum (LGM). Our results suggest that the islands have acted as refugia during the LGM and highlight the role of <i>in situ</i> glacial survival in maintaining high levels of genetic diversity of <i>M. kwangtungensis</i> in continental islands of subtropical China.</p>
Mimicry genes reduce pre-adult survival rate in Papilio polytes: A possible new mechanism for maintaining female-limited polymorphism in Batesian mimicry
<p>Batesian mimicry, in which harmless organisms resemble unpalatable or harmful species, is a well-studied adaptation for predation avoidance. The females of some Batesian mimic species comprise mimetic and non-mimetic individuals. Mimetic females of such polymorphic species clearly have a selective advantage due to decreased predation pressure, but the selective forces that maintain non-mimetic females in a population remain unclear. In the swallowtail butterfly, <em>Papilio polytes</em>, female polymorphism is controlled by the <em>H</em> (non-mimetic) and <em>h</em> (mimetic) alleles at a single autosomal locus. Here, we examined if the dominant <em>H</em> allele has a deleterious effect on the pre-adult survival rate (egg-to-adult emergence rate). We repeated an assortative mating-like treatment—i.e. breeding of males and females whose mothers had the same phenotype (mimetic or non-mimetic)—for three consecutive generations, while avoiding inbreeding. Results showed that pre-adult survival rate decreased over generations only in lines derived from mothers with the mimetic phenotype (hereafter, mimetic-assorted lines). This lowered survival was due to an increased mortality at the final instar larval stage and the pupal stages. Interestingly, the pre-adult mortality in the mimetic-assorted lines seemed to be associated with a male-biased sex ratio at adult emergence. These results suggest that the dominant <em>H</em> allele displays a mildly deleterious effect that is expressed more strongly in females and homozygous individuals than in heterozygous individuals. We propose that this cost of mimicry in larval and pupal stages contributes to the maintenance of female-limited polymorphism in <em>P. polytes</em>.</p>
Significant differentially expressed genes (DEG) for midgut tissue from bees maintained for four hours at 35°C or 45°C
<p><span><span>Honey bee colonies in the United States have suffered from increased die-off in the last few years with a complex set of interacting stresses playing a key role. With changing climate, an increase in the frequency of severe weather events, such as heat waves, is anticipated. Understanding how these changes may contribute to stress in honey bees is crucial. Individual honey bees appear to have a high capacity to endure thermal stress. One reason for this high-level endurance is likely their robust HSR which contributes to thermotolerance at the cellular level. However, less is known about other mechanisms of thermotolerance, especially those operating at the tissue level. To elucidate other determinants of this resilience in this species, we used <span>thermal stress coupled with RNAseq and identified broad transcriptional remodeling of a number of key signaling pathways in the honey bee, including those pathways known to be involved in digestive tract regeneration in the fruit fly such as the Hippo and JAK/STAT pathways. We also observe cell death and shedding of epithelial cells, which likely leads to induction of this regenerative transcriptional program. </span>We found that thermal stress affects many of these pathways in other tissues, suggesting a shared program of damage response. This study provides important foundational characterization of the tissue damage response program in this key pollinating species. In addition, our data suggest that a robust regeneration program may also be a critical contributor to thermotolerance at the tissue level, a possibility which warrants further exploration in this and other species. </span></span></p>
Interplay of above- and belowground resource limitations: a competition-facilitation shift maintains species coexistence
<p>Forest ecosystems are commonly characterised by a hierarchy of resources. During a disturbance of a forest community, increased light availability in the understorey can support competitive interactions at the expense of facilitation. This may overwhelm the role of belowground resource heterogeneity in maintaining species coexistence and so result in biotic homogenisation of a site. We re-surveyed species composition while estimating interspecific interactions along a microtopographic (moisture) gradient of a temperate swamp forest after increasing the availability of light into the undergrowth. We measured temporal changes in species diversity, compositional dissimilarity measures and functional traits, while belowground resource heterogeneity remained unchanged over time. To explain observed changes in terms of prevailing interspecific interactions, we evaluated temporal changes in the competition-facilitation balance. The productivity of understorey vegetation increased strongly over the five years of the study. This was accompanied by an increase in species diversity and the maintenance of compositional dissimilarity. The relative importance of competitive and facilitative interactions along the microtopographic gradient did not change over time. Rather than an overall biotic homogenisation of a site, we observed a spread of resource acquisitive species within the resource-rich sites and of stress-tolerant species within the resource-limited sites. We have shown that the temporary increase in productivity did not mitigate the effects of belowground resource heterogeneity on plant species turnover, which is likely maintained by contrasting interspecific interactions prevalent in different parts of the resource gradient. This suggests contrasting community assembly processes maintain diversity and productivity even in a local community.</p>
