Resonance energy transfer sensitizes and monitors in situ switching of LOV2-based optogenetic actuators
<p>Engineered light-dependent switches provide uniquely powerful opportunities to investigate and control cell regulatory mechanisms. Existing tools offer high spatiotemporal resolution, reversibility and repeatability. Yet cellular optogenetics applications remain limited in practice. Validation difficulties constrain most usage to easily verified protein translocation rather than inhibition of diffusible targets. Blue light levels commonly needed for actuation can be cytotoxic, precluding long-term experiments. We describe a simple approach overcoming these obstacles. Resonance energy transfer with an adjacent chromophore modulates actuation sensitivity over a 7-fold range. This simultaneously offers on-line monitoring of light-dependent switching and precise quantification of activation-relaxation properties in intact living cells. Applying this approach to different LOV2-based switches reveals relaxation times up to 11-fold faster than anticipated. <em>In situ</em>–measured parameter values guide the design of target-inhibiting actuation trains with minimal blue-light exposure, and context-based optimisation can increase sensitivity and experimental throughput a further 10-fold without loss of temporal precision.</p>
Exploiting Phase Transitions in Polymer Bilayer Actuators
<p>In many biological systems, the process of motion involves mechanically morphing materials, which change their mechanical properties and/or shape in response to a control signal. In the past two decades, significant progress has been made toward mimicking the underlying designs and resulting functions in artificial materials and systems. Polymer‐based bilayer bending actuators, in which the dissimilar thermal expansion of two materials is exploited, represent widely investigated actuation elements, but one limitation of this design approach is that the thermal expansion coefficient of polymers is generally small. Herein, it is shown that this problem is mitigated using a thermoplastic polyurethane elastomer with a crystallizable soft segment. The domains formed by the latter reversibly melt and crystallize in a convenient temperature range (30–60 °C), whereas the hydrogen‐bonded urethane hard segments serve as physical crosslinks that provide mechanical integrity and elastic behavior beyond this temperature. Melting the polyester results in a large nonlinear thermal expansion and thus actuation, in a narrow temperature range. The electrically controlled bilayer actuators are created by combining this material with cellulose acetate and integrating resistive heating electrodes. Optionally these devices are equipped with a thermally controlled supramolecular polymer adhesive “gripper.”</p>

Magnetically actuated gearbox for the wireless control of millimeter-scale robots
<p>The limited force or torque outputs of miniature magnetic actuators constrain the locomotion performances and functionalities of magnetic millimeter-scale robots. Here, we present a magnetically actuated gearbox with a maximum size of 3 millimeters for driving wireless millirobots. The gearbox is assembled using microgears that have reference diameters down to 270 micrometers and are made of aluminum-filled epoxy resins through casting. With a magnetic disk attached to the input shaft, the gearbox can be driven by a rotating external magnetic field, which is not more than 6.8 millitesla, to produce torque of up to 0.182 millinewton meters at 40 hertz. The corresponding torque and power densities are 12.15 micronewton meters per cubic millimeter and 8.93 microwatt per cubic millimeter, respectively. The transmission efficiency of the gearbox in the air is between 25.1 and 29.2% at actuation frequencies ranging from 1 to 40 hertz, and it lowers when the gearbox is actuated in viscous liquids. This miniature gearbox can be accessed wirelessly and integrated with various functional modules to repeatedly generate large actuation forces, strains, and speeds; store energy in elastic components; and lock up mechanical linkages. These characteristics enable us to achieve a peristaltic robot that can crawl on a flat substrate or inside a tube, a jumping robot with a tunable jumping height, a clamping robot that can sample solid objects by grasping, a needle-puncture robot that can take samples from the inside of the target, and a syringe robot that can collect or release liquids.</p>
