Data from: Combined analysis of extant Rhynchonellida (Brachiopoda) using morphological and molecular data

Independent molecular and morphological phylogenetic analyses have often produced discordant results for certain groups which, for fossil-rich groups, raises the possibility that morphological data might mislead in those groups for which we depend upon morphology the most. Rhynchonellide brachiopods, with more than 500 extinct genera but only 19 extant genera represented today, provide an opportunity to explore the factors that produce contentious phylogenetic signal across datasets, as previous phylogenetic hypotheses generated from molecular sequence data bear little agreement with those constructed using morphological characters. Using a revised matrix of 66 morphological characters, and published ribosomal DNA sequences, we performed a series of combined phylogenetic analyses to identify conflicting phylogenetic signals. We completed a series of parsimony-based and Bayesian analyses, varying the data used, the taxa included, and the models used in the Bayesian analyses. We also performed simulation-based sensitivity analyses to assess whether the small size of the morphological data partition relative to the molecular data influenced the results of the combined analyses. In order to compare and contrast a large number of phylogenetic analyses and their resulting summary trees, we developed a measure for the incongruence between two topologies, and simultaneously ignore any differences in phylogenetic resolution. Phylogenetic hypotheses generated using only morphological characters differed amongst each other, and with previous analyses, while molecular-only and combined Bayesian analyses produced extremely similar topologies. Characters historically associated with traditional classification in the Rhynchonellida have very low consistency indices on the topology preferred by the combined Bayesian analyses. Overall, this casts doubt on the use of morphological systematics to resolve relationships among the crown rhynchonellide brachiopods. However, expanding our dataset to a larger number of extinct taxa with intermediate morphologies is necessary to exclude the possibility that the morphology of extant taxa is not dominated by convergence along long branches.

VCF datasets and analysis scripts for: The combination of genomic offset and niche modelling provides insights into climate change-driven vulnerability

<p>Global warming is increasingly exacerbating biodiversity loss. Populations locally adapted to spatially heterogeneous environments may respond differentially to climate change, but this intraspecific variation has only recently been considered when modelling vulnerability under climate change. Here, we incorporate intraspecific variation in genomic offset and ecological niche modelling to estimate climate change-driven vulnerability in two bird species in the Sino-Himalayan Mountains. We found that the cold-tolerant populations show higher genomic offset but risk less challenge for niche suitability decline under future climate than the warm-tolerant populations. Based on a genome-niche index estimated by combining genomic offset and niche suitability change, we identified the populations with the least genome-niche interruption as potential donors for evolutionary rescue, i.e., the populations tolerant to climate change. We evaluated potential rescue routes via a landscape genetic analysis. Overall, we demonstrate that the integration of genomic offset, niche suitability modelling, and landscape connectivity can improve climate change-driven vulnerability assessments and facilitate effective conservation management.</p>

The application of a mode of double preconcentration and double stripping steps in combination with a flow system to decrease the detection limit and increase the selectivity of determinations in stripping analysis of As(III)

<p><span>Due to the fact that arsenic </span><span>has very harmful and toxic effects on living organisms </span><span>monitoring of its content in various environmental samples </span><span>at trace level concentrations</span><span> </span><span>is desirable and highly recommended. Such advantages as low cost and portable instrumentation, achieving low detection limits and most of all the possibility of the direct determination of As(III) with no separation step make stripping voltammetry suitable for the determination of arsenic.</span><span>&nbsp;</span><span>&nbsp;<br></span></p> <p><span>These data concer the application of double deposition and double stripping steps mode in combination with a flow system for anodic stripping voltammetric determination of As(III) ions. An advantage of the proposed way of the voltammetric measurements is a decrease of the detection limit as compared to the values obtained using a traditional three-electrode system thanks to an initial preconcentration of the analyte on the first working electrode as well as the significant increase in a selectivity of As(III) determination thanks to the possibility of solution exchange after the first deposition step.</span></p> <p><span><span>The proposed way of the voltammetric measurements leads to the following analytical advantages: (i) a decrease of the detection limit as compared to the values obtained using traditional three-electrode system by an initial preconcentration of the analyte using the large-surface area electrode; (ii) an increase of the selectivity of voltammetric determinations thanks to the application of a flow system and exchange the solution after the first deposition step; (iii) simplification of performing the measurements in comparison to the original version of double accumulation and stripping steps mode due to the lack of a need to change the position of the first working electrode; (iv) the possibility of analysis of samples with a complex matrix thanks to the use of a flow system leading to minimization of matrix effects in the second deposition step. The developed procedure of As(III) determination offers a wide linear dynamic range in the range from 1 &times; 10<sup>-9</sup> to 5 &times; 10<sup>-8</sup> mol L<sup>-1</sup> at a deposition time of 120 s at both working electrodes. The detection limit for As(III) determination was 4.8 &times; 10<sup>-10</sup> mol L<sup>-1</sup>. The undoubted advantage of the described procedure is its high selectivity. The tolerable limit of excess of Cu(II), which is the main interferent in the course of anodic stripping voltammetric As(III) determination, was significantly increased thanks to the solution exchange after the first deposition step. The correctness of the proposed procedure was confirmed by analysis of a real water sample and lake water certified reference material by obtaining satisfactory recovery values. The additional advantage of the proposed procedure is a miniaturization of the measurement system and the use of environmentally friendly electrodes.</span></span></p>
