Data Archival for Economic Cost Modeling of Chinook Habitat Restoration in the Stillaguamish River Basin
<p>We used geospatial data to model economic cost estimates of habitat restoration in the Stillaguamish River Basin in the Puget Sound. We utilized data pertaining to the streams/rivers, floodplain habitat, subbasins, elevation, distance to roads, demographics, and land use within the Stillaguamish River Basin to do so. Analysis included using the different attributes of the Stillaguamish River Basin to create low and high cost estimates for floodplain, engineered log jam, and riparian planting habitat restoration. We specifically looked at the slope and size of streams, area of habitat that needed to be restored, slopes of the riparian area, distance to nearest road, and canopy angles as our model inputs. We followed cost estimate guidance provided by the Puget Sound Shared Strategy to identify our cost ranges and updated them to todays prices using the producer price index. An additional land use analysis was performed to quantify the total area and cost of potential agricultural land in the basin. Lastly, we investigated the demographics of the region to identify areas of POC and low income in relation to proposed restoration actions.</p>

Biophysical models unravel associations between glucocorticoids and thermoregulatory costs across avian species
<ol> <li>Glucocorticoids (GCs) are metabolic hormones that regulate physiological and behavioural responses to environmental change and mediate homeostasis maintenance in vertebrates. Despite the assumption that GCs covary with energy metabolism, we yet lack a mechanistic understanding of how environmental factors such as temperature modulate GC variation through their effect on organismal energy balance. In particular, the mechanisms linking temperature-dependent metabolic rate and GCs at broad spatial scales and across species remain poorly investigated.</li> <li>Here we used biophysical models to calculate thermoregulatory costs (i.e., the amount of heat required to keep body temperature in homeostasis) of free-living birds as a function of environmental conditions, body size, shape, and insulating layer of feathers. We then investigated the link between ambient temperature, cost of thermoregulation and baseline plasma GC concentrations in a comparative study including GC data from 94 bird species from HormoneBase.</li> <li>We found a significant, positive association between thermoregulatory costs and baseline GC concentrations. Interestingly, models including thermoregulatory costs better explained GC variation when compared to those including ambient temperature as a predictor variable. This result suggests that body size and shape fundamentally modulate energy requirements for thermoregulation and thereby GC concentrations in the wild.</li> <li>By providing a mechanistic description of the link between ambient temperature and thermoregulatory metabolism, biophysical models provide a tool to predict the impact of environmental conditions on energy metabolism. Our work demonstrates that differences in thermoregulation modulate variation in GC concentrations across a broad climatic gradient.</li> </ol>

Recent advances in high-throughput sequencing library preparation and subgenomic enrichment methods have opened new avenues for population genetics and phylogenetics of non-model organisms. To multiplex large numbers of indexed samples while sequencing predominantly orthologous, targeted regions of the genome, we propose modifications to an existing, in-solution capture that utilizes PCR products as target probes to enrich library pools for the genomic subset of interest. The sequence capture using PCR-generated probes (SCPP) protocol requires no specialized equipment, is highly flexible, and significantly reduces experimental costs for projects where a modest scale of genetic data is optimal (25-100 genomic loci). Our alterations enable application of this method across a wider phylogenetic range of taxa and result in higher capture efficiencies and coverage at each locus. Efficient and consistent capture over multiple SCPP experiments and at various phylogenetic distances is demonstrated, extending the utility of this method to both phylogeographic and phylogenomic studies.
