Data and code for: Failure to purge: Population and individual inbreeding effects on fitness across generations of wild Impatiens capensis

<p>Inbreeding exposes deleterious recessive alleles in homozygotes, lowering fitness and generating inbreeding depression (ID). Both purging (via selection) and fixation (via drift) should reduce segregating deleterious mutations and ID in more inbred populations. These theoretical predictions are not well-tested in wild populations, which is concerning given purging/fixation have opposite fitness outcomes. We examined how individual- and population-level inbreeding and genomic heterozygosity affected maternal and progeny fitness within and among 12 wild populations of <em>Impatiens capensis</em>. We quantified maternal fitness in home sites, maternal multilocus heterozygosity (using 12,560 SNPs), and lifetime fitness of selfed and predominantly outcrossed progeny in a common garden. These populations spanned a broad range of individual- (<span class="s1"><em>f</em></span><span class="s2"><sub>i</sub></span><em> </em>= -0.17–0.98) and population-level inbreeding (<span class="s1"><em>F</em></span><span class="s2"><sub>IS</sub></span> = 0.25–0.87). More inbred populations contained fewer polymorphic loci, less fecund mothers, and smaller progeny, suggesting higher fixed loads. However, despite appreciable ID (mean: 8.8 lethal equivalents per gamete), ID did not systematically decline in more inbred population. More heterozygous mothers were more fecund and produced fitter progeny in outcrossed populations, but this pattern unexpectedly reversed in highly inbred populations. These observations suggest that persistent overdominance or some other force acts to forestall purging and fixation in these populations.</p>

NCCD-PF - A pre-failure narrow concrete cracks dataset for engineering structures damage classification and semantic segmentation

<p>The&nbsp;NCCD-PF dataset was developed for the classification and semantic segmentation of narrow concrete cracks in engineering structures elements at the pre-failure state. It only includes cracks whose width is narrower than 0.3 mm, i.e. the limit value specified in EC 1992-1-1 for typical elements of engineering structures and environmental conditions.</p> <p>This dataset is dedicated to the early crack detection at a stage when the serviceability limit state has not yet been exceeded and the failure of a structural element has not occurred. By implementing the early crack detection approach, it is possible to protect cracks in order to stop or slow down their propagation and thus to extend the structure's lifespan.</p> <p>This dataset contains images of cracks appearing on various elements of engineering structures (bridges, viaducts, tunnels) made of reinforced concrete (including abutments, tunnel walls, concrete barriers, pillars). The images were captured on construction sites and during inspections of engineering structures, at different stages of the reinforced concrete structure's working conditions - from the construction stage (when the elements are loaded only by their own weight) to the structure's use stage (when the elements are loaded by most of the design loads). The images are also differentiated by the cause of the cracking (ex., thermal and shrinkage stresses in young concrete, excessive stresses). The images were acquired using fixed-focus cameras without prior conditioning in order to represent the real working conditions of a bridge engineer during structural inspections. The images are characterised by a high degree of complexity due to the quality of the concrete surface finish (e.g. presence of formwork marks, concrete trowel marks), which could potentially be recognised&nbsp;as cracks.</p> <p>This dataset is dedicated to researchers working in the fields of computer vision, machine learning and deep learning. In particular, it contains domain knowledge in structural health monitoring, so that it can support the work of engineers in detecting cracks of concrete elements in a pre-failure state.</p> <p>A detailed description of the dataset is presented in <a href="https://www.nature.com/articles/s41597-023-02839-z" target="_blank" rel="noopener">A pre-failure narrow concrete cracks dataset for engineering structures damage classification and segmentation</a> (DOI: 10.1038/s41597-023-02839-z).</p>

The Ubiquitin Ligase WWP1 Contributes to Shifts in Matrix Proteolytic Profiles and a Myocardial Aging Phenotype with Diastolic Heart Failure

<p><strong><em>Aims</em></strong>. Ubiquitylation is a key event that regulates protein turnover, and induction of the ubiquitin ligase E3 WWP1 has been associated with age. Left ventricular hypertrophy (LVH) commonly occurs as a function of age and can cause heart failure with a preserved ejection fraction (EF; HFpEF). We hypothesized that overexpression (O/E) of WWP1 in the heart would cause LVH as well as functional and structural changes consistent with the aging HFpEF phenotype.</p> <p><strong><em>Methods and Results.</em></strong> Global WWP1 O/E was achieved in mice (n=11) and echocardiography (40 MHz) performed to measure LV mass, EF, Doppler velocities (early-E, late/atrial-A), myocardial relaxation (E&rsquo;), and isovolumetric relaxation time (IVRT) at 4, 6, and 8 weeks. Age matched wild type animals (n=15) were included as referent controls. LV EF was identical (60+1% vs 60+1%, p&gt;0.90) with no difference in LV mass (67+3 vs 75+5, p&gt;0.25) at 4 weeks. However, at 8 weeks of age, LV mass increased by over two-fold, E/A fell (impaired passive filling), and E/E&rsquo; was lower and IVRT prolonged (impaired LV relaxation) - all p&lt;0.05. Collagen percent area increased by over two-fold and fibrillar collagen expression (rtPCR) by over 1.5 fold (p&lt;0.05) with WWP1 O/E. WWP1 with an anti-WWP1 antibody could be identified in isolated cardiac fibroblasts with WWP1 increased by over two-fold in O/E fibroblasts (p&lt;0.05).</p> <p><strong><em>Conclusion.</em></strong> Inducing WWP1 expression caused LVH, preserved systolic function, but impaired diastolic dysfunction, consistent with the HFpEF phenotype. Targeting the WWP1 pathway may be a novel therapeutic target for this intractable form of HF associated with aging.</p>
