Resting-state fMRI data for locating causal hubs of memory consolidation in spontaneous brain network

The mouse fMRI data for the paper "Locating causal hubs of memory consolidation in spontaneous brain network in male mice" published in Nature Communications (DOI: 10.1038/s41467-023-41024-z). This includes longitudinal resting-state fMRI data in mice after behavioural training for 1-Day or 5-Day Active Place Avoidance (APA) task, acquired at post-training day 1 and day 8. Due to the large datasets, each group has been packed into several 2GB zip files. They need to be downloaded into the same folder and unpacked together (e.g. 1-Day APA Post training day 1 has five zip files starting with "1DAPA_PostDay1"). The structural and EPI templates and the ROI labels in the AMBMC atlas space are provided in the AMBMC_label.zip.

Data from: The inner junction complex of the cilia is an interaction hub that involves tubulin post-translational modifications

Microtubules are cytoskeletal structures involved in stability, transport and organization in the cell. The building blocks, the α- and β-tubulin heterodimers, form protofilaments that associate laterally into the hollow microtubule. Microtubule also exists as highly stable doublet microtubules in the cilia where stability is needed for ciliary beating and function. The doublet microtubule maintains its stability through interactions at its inner and outer junctions where its A- and B-tubules meet. Here, using cryo-electron microscopy, bioinformatics and mass spectrometry of the doublets of Chlamydomonas reinhardtii and Tetrahymena thermophila, we identified two new inner junction proteins, FAP276 and FAP106, and an inner junction-associated protein, FAP126, thus presenting the complete answer to the inner junction identity and localization. Our structural study of the doublets shows that the inner junction serves as an interaction hub that involves tubulin post-translational modifications. These interactions contribute to the stability of the doublet and hence, normal ciliary motility.

A synaptomic analysis reveals dopamine hub synapses in the mouse striatum : Source Data

Source data for our study on dopamine hub synapses

Dopamine transmission is involved in reward processing and motor control, and its impairment plays a central role in numerous neurological disorders. Despite its strong pathophysiological relevance, the molecular and structural organization of the dopaminergic synapse remains to be established. Here, we used targeted labelling and fluorescence activated sorting to purify striatal dopaminergic synaptosomes. We provide the proteome of dopaminergic synapses with 57 proteins specifically enriched. Beyond canonical markers of dopamine neurotransmission such as dopamine biosynthetic enzymes and cognate receptors, we validated 6 proteins not previously described as enriched (Cpne7, Apba1/Mint1, Cadps2, Cadm2/SynCAM 2, Stx4, Mgll). Moreover, our data reveal the adhesion of dopaminergic synapses to glutamatergic, GABAergic or cholinergic synapses in structures we named “dopamine hub synapses”. At glutamatergic synapses, pre- and postsynaptic markers are significantly increased upon association with dopamine synapses. Dopamine hub synapses may thus support local dopaminergic signalling, complementing volume transmission thought to be the major mechanism by which monoamines modulate network activity.

Climate Hazards Data Integration and Visualization for the Climate Adaptations Solutions Accelerator through School-Community Hubs

Community engagement in planning is essential for effective and just climate adaptation. However, historically underserved communities are often difficult to reach through traditional means of soliciting public input. The Climate Adaptation Solutions Accelerator (CASA) through School-Community Hubs project identifies public schools as promising sites for building both community engagement and community capacity for climate adaptation. To serve in this role, schools need information about the intersecting threats climate change poses to the communities they serve. The Climate Hazard Dashboard for California Schools is a platform that maps the current and future risks associated with five climate hazards, including wildfire, extreme heat days, wildfire extreme precipitation, flooding, and sea level rise, for the nearly 10,000 public schools serving Kindergarten through Grade 12 students in California. Each hazard is mapped and visualized at the school level, providing an accessible way for administrators, teachers, students, and neighborhoods to explore data reflecting the climate hazards they face, at a scale relevant to their communities. The dashboard also provides an aggregate summary hazard metric.

COMPUTING HUBS IN THE HIPPOCAMPUS AND CORTEX

Neural computation, which relies on the active storage and sharing of information, occurs within large neuron networks in the highly dynamic context of varying brain states. Whether such functions are performed by specific subsets of neurons and whether they occur in specific dynamical regimes remains poorly understood. Using high density recordings in the hippocampus, medial entorhinal and medial prefrontal cortex of the rat, we identify computing substates, or discrete epochs, in which specific computing hub neurons perform well defined storage and sharing operations in a brain state-dependent manner. We retrieve a multiplicity of distinct computing substates within each global brain state, such as REM and nonREM sleep. Half of recorded neurons act as computing hubs in at least one substate, suggesting that functional roles are not firmly hardwired but dynamically reassigned at the second timescale. We identify sequences of substates whose temporal organization is dynamic and stands between order and disorder. We propose that global brain states constrain the language of neuronal computations by regulating the syntactic complexity of these substate sequences.
