<p><span>1. Plants or seeds produced in botanic gardens or nurseries have become an important source of plant material for reintroductions or population reinforcements. However, recent research has shown that these living collections bear the risk of being genetically impoverished and adapted to the artificial habitat. In particular, many studies have reported a decline of seed dormancy during ex situ cultivation, which may compromise their suitability for reintroduction programs. However, the impact of those ex situ-derived changes on the germination and establishment of reintroduced plant populations is still unclear. </span></p> <p><span>2. We studied the germination behaviour, population establishment and plant fitness over three years of reintroduced plants of the short-lived perennial <em>Digitalis</em> <em>lutea</em>, comparing plants grown from (1) a 30-year botanic garden population, (2) seeds from a seed bank representing the initial starting point of the botanic garden culture, and (3) a re-sampled corresponding wild population. </span></p> <p><span>3. Under laboratory conditions, wild-collected seeds required cold stratification to germinate, whereas seeds from the garden population germinated without stratification. This pattern was strongly reduced in an outdoor pot experiment, where only a few garden seeds germinated before winter, and all seeds remained dormant when seeded in the natural area of origin. In a transplant experiment, reintroduced plants from the wild population outperformed both, the garden and the seed bank plants, in their fitness in the first 3 years after reintroduction suggesting adaptation to current climatic conditions. </span></p> <p><span>4. Synthesis and Applications: Our study demonstrates that trait changes that occurred during ex situ cultivation can negatively impact the establishment of reintroduced plants. We conclude that wild plant material collected from contemporary populations is best suited for reintroduction and should be preferred over ex situ-cultivated and seed bank stored material, especially when the cultivation spanned multiple generations. However, our study also shows that germination requirements change in complex ways, and the loss of dormancy observed under laboratory conditions may not always be directly transferable to natural conditions. When established standards are respected, ex situ propagated material may thus still be a valuable resource, especially when wild material is not available in sufficient quantities. </span></p>

<p>Captive breeding programs for endangered species can increase population numbers for eventual reintroduction to the wild. Captive populations are typically small and isolated, which results in inbreeding and reduction of genetic variability, and may lead to an increased risk of extinction. The Omkoi Wildlife Breeding Center maintains the only Thai captive Chinese goral (<i>Naemorhedus griseus</i>) population, and has plans to reintroduce individuals into natural isolated populations. Genetic variability was assessed within the captive population using microsatellite data. Although no bottleneck was observed, genetic variability was low (allelic richness = 7.091 ± 0.756, <i>H</i><sub>e</sub> = 0.455 ± 0.219; <i>H</i><sub>e</sub> < <i>H</i><sub>o</sub>) and 11 microsatellite loci were informative that likely reflect inbreeding. Estimates of small effective population size and limited numbers of founders, combined with wild-born individuals within subpopulations, tend to cause reduction of genetic variability over time in captive programs. This leads to low reproductive fitness and limited ability to adapt to environmental change, thereby increasing the risk of extinction. Management of captive populations as evolutionarily significant units with diverse genetic backgrounds offers an effective strategy for population recovery. Relocation of individuals among subpopulations, or introduction of newly captured wild individuals into the captive program will help to ensure the future security of Chinese goral. Implications for future conservation actions for the species are discussed herein.</p>
Data from: Structural diversity of naturally regenerating Chinese yew (Taxus wallichiana var. mairei) populations in an ex situ conservation
The Chinese yew (Taxus wallichiana var. mairei) is ranked among the first class of important wild endangered plants in China. However, due to its overexploitation, it now occurs scattered in the forest undergrowth along the Yangtze River Valley. To improve this tree's conservation management, we used structural indices to investigate the structural diversity of naturally regenerating yew populations that have established via ex situ conservation. The results show that most yews had larger non-yew tree neighbors; these were 30–70% larger than their reference trees. Collectively, the average distances between the yews and the three nearest-neighboring trees were short (<3 m). This result suggests that the yews likely face strong interspecific competition from neighbors. In these two forest stands, most of the pole-sized yews are found beneath a single tall neighboring tree (height ≥10 m), and their growth was enhanced under a single neighboring tree but not under two, three or zero neighboring trees. Finally, we recommend simple silvicultural treatments to reduce interspecific competition; specifically, the cutting or pruning of branches of large neighboring trees in tandem with the thinning of canopy trees growing next to the mother yews.
When more is not merrier: using wild population dynamics to understand the effect of density on ex situ seahorse mating behaviors
Seahorses are one of the most iconic examples of a monogamous species in the animal kingdom. Documenting monogamous behaviors of seahorses have proven to be quite complicated to study in the wild because of their low population densities and cryptic habits. Another challenge involves interpreting these behaviors in captivity because recreating realistic densities of wild populations in the laboratory can be difficult due to their patchy distributions. This study investigates the relationship between stocking density and mating and competitive behavior from the context of the field biology of the dwarf seahorse, Hippocampus zosterae (Jordan & Gilbert). Animals were housed in 38 liter tanks at a range of densities and sex ratios (from 2-8 animals per tank), and their reproductive and other social behaviors were monitored from tank introduction through copulation. At low tank densities and even sex ratios but comparatively high field densities, activity level in trials was low. A higher level of males in tanks across all densities increased competition, activity levels, and aggression leading to partial egg transfers and failed pregnancies, resulting in lower reproductive success. Across seahorse species, mean and maximum wild densities were consistently lower than those used in captive breeding, with adult sex ratios that were significantly female biased. However, significant variation exists in wild seahorse densities across species, with higher densities detected in focal/mark recapture studies and on artificial habitat structures than reported with belt transect sampling techniques. Interchange of knowledge gained in both captive and wild contexts will allow us to better understand the biology of this genus, and improve reproduction in captivity. Interpreting captive reproductive behaviors of seahorses within various densities reported from natural populations will help us predict the impact of conservation efforts and increase the likelihood of long-term persistence of populations for this threatened genus.
Data from: Distinguishing the victim from the threat: SNP‐based methods reveal the extent of introgressive hybridization between wildcats and domestic cats in Scotland and inform future in situ and ex situ management options for species restoration
The degree of introgressive hybridisation between the Scottish wildcat and domestic cat has long been suspected to be advanced. Here we use a 35-SNP-marker test, designed to assess hybridisation between wildcat and domestic cat populations in Scotland, to assess a database of 265 wild-living and captive cat samples, and test the assumptions of the test using 3097 SNP markers generated independently in a subset of the data using ddRAD. We discovered that despite increased genetic resolution provided by these methods, wild-living cats in Scotland show a complete genetic continuum or hybrid swarm structure when judged against reference data. The historical population of wildcats, although hybridised, clearly groups at one end of this continuum, as does the captive population of wildcats. The interpretation of pelage scores against nuclear genetic data continues to be problematic. This is probably because of a breakdown in linkage disequilibrium between wildcat pelage genes as the two populations have become increasingly mixed, meaning that pelage score or SNP score alone are poor diagnostic predictors of hybrid status. Until better tools become available, both should be used jointly, where possible, when making management decisions about individual cats. We recommend that the conservation community in Scotland must now define clearly what measures are to be used to diagnose a wildcat in the wild in Scotland, if future conservation action is to be effective.
