Multidimensional control of therapeutic human cell function with synthetic gene circuits
<p>Synthetic gene circuits that precisely control human cell function could expand the capabilities of gene and cell-based therapies. However, platforms for developing circuits in primary human cells that drive robust functional changes in vivo and have compositions suitable for clinical use are lacking. Here, we develop synthetic transcriptional regulators that are compact and based largely on human-derived proteins (synZiFTRs). As a proof of principle, we engineer gene switches and circuits that allow precise, user-defined control over therapeutically-relevant genes in primary T cells using orthogonal, FDA-approved small molecule inducers. Our circuits can instruct T cells to sequentially activate multiple cellular programs, such as proliferation and anti-tumor activity, to drive synergistic therapeutic responses. This platform should accelerate development and clinical translation of synthetic gene circuits in diverse human cell types and contexts.</p>
Data for: Multidimensional stoichiometric mismatch explains differences in detritivore biomass across three forest types
<p><span>The ecological stoichiometry theory provides a framework to understand organism fitness and population dynamics based on the stoichiometric mismatch between organisms and their resources. Recent studies have revealed that different soil animals occupy distinct multidimensional stoichiometric niches (MSNs), which likely determine their specific stoichiometric mismatches and population responses facing resource changes. The goals of the present study are to examine how long-term forest plantations affect the multidimensional elemental contents of litter and detritivores and the population size of detritivores that occupy distinct MSNs.</span></p> <p><span>We evaluated the contents of ten elements of two detritivore taxa (lumbricid earthworms and julid millipedes) and their litter resources, quantified their MSNs and the multidimensional stoichiometric mismatches, and examined how such mismatch patterns influence the density and total biomass of detritivores across three forest types spanning from natural forests (oak forest) to plantations (pine and larch forests).</span></p> <p><span>Sixty-year pine plantations changed the multidimensional elemental contents of litter but did not influence the elemental contents of the two detritivore taxa. Earthworms and millipedes exhibited distinct patterns of MSNs and stoichiometric mismatches, but they both experienced severer stoichiometric mismatches in pine plantations than in oak forests and larch plantations. Such stoichiometric mismatches led to lower density and biomass of both earthworms and millipedes in pine plantations. In other words, under conditions of low litter quality and severe stoichiometric mismatches in pine plantations, detritivores maintained their body elemental contents but decreased their population biomass.</span></p> <p><span>Our study illustrates the success of using the multidimensional stoichiometric framework to understand the impact of forest plantations on animal population dynamics, which may serve as a useful tool in addressing ecosystem responses to global environmental changes.</span></p>
Resilience indicators used in the multidimensional characterization of the resilience of urban areas prone to flash floods in the region of Castilla y León (Spain)
<p>Database containing resilience indicators used in the characterization of all dimensions of resilience (social, economic, ecosystemic, physical, institutional and cultural) in those municipalities susceptible to flash floods in the region of Castilla y León (Spain). The database includes a total of 191 resilience indicators, of which 48 correspond to social resilience, 32 to economic resilience, 34 to ecosystem resilience, 44 to physical resilience, 27 to institutional resilience and 6 to cultural resilience.</p> <p>The Excel (.xlsx) file is organized by dimensions, where the prefix "SOC_" corresponds to the social dimension, "ECON_" to the economic dimension, "ECOS_" to the ecosystemic dimension, "PHY_" to the physical dimension, "INS_" to the institutional dimension and "CUL_" to the cultural dimension. Each dimension of resilience occupies two tabs: the first tab (suffixes "_data") contains the description of the indicators included (i.e., indicator code, unit of measurement, year of information, source of information, direct link to the information and bibliographic references that support the consideration of the different indicators); while the second tab (suffixes "_variables") contains the values of the different indicators (identified by their codes, which appear in the first tab) for each unit of analysis. </p>
Comparing Finnish universities' publication profiles using multidimensional field-normalized indicators - dataset

Precipitation, rather than temperature drives coordination of multidimensional root traits with ectomycorrhizal fungi in alpine coniferous forests
<ol> <li><span>The interactions between roots and mycorrhizal fungi are critical for our understanding of the multidimensional root economics space. Our knowledge on their relationships comes mainly from arbuscular mycorrhizal (AM) plants, and less is known about how roots are coordinated with ectomycorrhizal (ECM) fungal communities, especially in ECM-dominated alpine forests that are highly sensitive to climate change worldwide.</span></li> <li> <span>Here, we investigated the coordination between roots and ECM fungi and their drivers by measuring </span><span>multiple</span><span> root traits, ECM fungal </span><span>composition and environmental factors of 47 coniferous populations across the alpine coniferous forests </span><span>on the Tibetan Plateau.</span> </li> <li> <span>Our results reveal two independent fine-root trait dimensions, i.e., root foraging dimension and root uptake dimension, which are represented by</span> <span>root diameter-specific root length, root tissue density-root N concentration. Importantly, the hyphal exploration-type-based ECM foraging correlated significantly with both root foraging and root uptake dimension. Further, in the low-temperature plateau, it is precipitation-induced changes in soil moisture, soil nutrients and pH that drive the proportion of </span><span>longer-</span><span>distance hyphal exploration types to increase with </span><span>higher root </span><span>foraging </span><span>by higher </span><span>specific root length, and </span><span>to </span><span>decrease with </span><span>higher </span><span>uptake </span><span>by higher </span><span>root N concentration.</span> </li> <li> <span><em>Synthesis</em>.</span><span> The coordination of multidimensional root traits with ECM fungi differs greatly from the well-recognized pattern in AM plants that mycorrhizal fungi connect predominantly with root foraging and that roots and mycorrhizal fungi are temperature sensitive. These findings provide a new insight for our holistic understanding of how roots and mycorrhizal fungi vary collaboratively and hence driving plant community assembly and responses to the changing climate.</span> </li> </ol>
