Development of a multivariable risk model integrating urinary peptide metabolites and Extracellular Vesicle RNA data to detect significant prostate cancer
<p>The aim of this study was to investigate whether the robust integration of expression data from urinary extracellular vesicle RNA (EV-RNA) with urine proteomic metabolites can accurately predict PCa biopsy outcome. Urine samples were analyzed by mass spectrometry and NanoString gene-expression analysis. As a result, four classifiers were generated: ‘MassSpec’ (CE-MS proteomics), ‘EV-RNA’, ‘SoC’ (standard of care) and ‘ExoSpec’. The best prediction for Gs³3+4 at initial biopsy (AUC=0.83, 95% CI:0.77-0.88) was achieved by applying ‘ExoSpec’ classifier and he outperformed other predictive classifiers. In addition, the results showed that the performance of ‘ExoSpec’ could reduce unnecessary biopsies by 30%.</p>
Supplemental data for: Development and validation of a polygenic risk score for stroke in the Chinese population
<div class="WordSection1"> <div class="WordSection1"> <strong>Objective</strong>: To construct a polygenic risk score (PRS) for stroke and evaluate its utility in risk stratification and primary prevention for stroke.</div> <div class="WordSection1"> </div> <div class="WordSection1"> <strong>Methods</strong>: Using meta-analytic approach and large genome-wide association results for stroke and stroke-related traits in East Asians, we generated a combined PRS (metaPRS) by incorporating 534 genetic variants in a training set of 2,872 patients with stroke and 2,494 controls. We then validated its association with incident stroke using Cox regression models in large Chinese population-based prospective cohorts comprising 41,006 individuals.</div> <div class="WordSection1"> </div> <div class="WordSection1"> <strong>Results</strong>: During a total of 367,750 person-years (mean follow-up 9.0 years), 1,227 participants developed stroke before age of 80 years. Individuals with high polygenic risk had an about 2-fold higher risk of incident stroke compared with those with low polygenic risk (HR: 1.99, 95% CI: 1.66-2.38), with the lifetime risk of stroke being 25.2% (95% CI: 22.5%-27.7%) and 13.6% (95% CI: 11.6%-15.5%), respectively. Individuals with both high polygenic risk and family history displayed the lifetime risk as high as 41.1% (95% CI: 31.4%-49.5%). Moreover, individuals with high polygenic risk achieved greater benefits in terms of absolute risk reductions from adherence to ideal fasting blood glucose and total cholesterol than those with low polygenic risk. Maintaining favorable cardiovascular health (CVH) profile could substantially mitigate the increased risk conferred by high polygenic risk to the level of the low polygenic risk (from 34.6 % to 13.2%).</div> <div class="WordSection1"> </div> <div class="WordSection1"> <strong>Conclusions</strong>: Our metaPRS has great potential for risk stratification of stroke and identification of individuals who may benefit more from maintaining ideal CVH. </div> <div class="WordSection1"> </div> <div class="WordSection1"> <strong>Classification of Evidence</strong>: This study provides Class I evidence that a meta-polygenic risk score is predictive of stroke risk.</div> <p> </p> </div>

Development under predation risk increases serotonin-signaling, variability of turning behavior and survival in adult fruit flies Drosophila melanogaster
<p>The development of high-throughput behavioral assays, where numerous individual animals can be analyzed in various experimental conditions, has facilitated the study of animal personality. Previous research showed that isogenic Drosophila melanogaster flies exhibit striking individual non-heritable locomotor handedness. The variability of this trait, i.e., the predictability of left-right turn biases, varies across genotypes and under the influence of neural activity in specific circuits. This suggests that the brain can dynamically regulate the extent of animal personality. It has been recently shown that predators can induce changes in prey phenotypes via lethal or nonlethal effects affecting the serotonergic signaling system. In this study, we tested whether fruit flies grown with predators exhibit higher variability/lower predictability in their turning behavior and higher survival than those grown with no predators in their environment. We confirmed these predictions and found that both effects were blocked when flies were fed an inhibitor (αMW) of serotonin synthesis. The results of this study demonstrate a negative association between the unpredictability of turning behavior of fruit flies and the hunting success of their predators. We also show that the neurotransmitter serotonin controls predator-induced changes in the turning variability of fruit flies, regulating the dynamic control of behavioral predictability.</p>
Data underlying the publication Development and implementation of a risk assessment tool for broiler farm biosecurity and a health intervention plan in the Netherlands, Greece, and Cyprus
<p>Introduction</p> <p>Preventing pathogens from entering and spreading on farms is the first step in reducing health problems.<br> For this study a BiosEcurity Assessment Tool was developed to identify strengths and weaknesses in biosecurity on broiler farms,<br> which was used as a starting point to formulate tailor-made health plans to improve broiler health and reduce antimicrobial use.</p> <p>Measurements and data collection</p> <p>Farms were divided into 3 separate areas according to associated biosecurity risk; high disease risk external areas (red zone), medium risk service areas (orange zone),<br> and the clean and highly secure access-restricted green zone. In the Netherlands, Cyprus, and Greece, 13, 15, and 7 broiler houses were monitored for 4 production cycles (2 preintervention and 2 postintervention cycles).<br> At the start of the study the BiosEcurity Assessment Tool assessment was performed and a health plan was made in consultation with the veterinarian.<br> After the second cycle a start was made with the implementation of the health plan.</p>
