All data support the published articel "Loop-optimization of Trichoderma reesei endoglucanases for balancing the activity–stability trade-off through cross-strategy between machine learning and the B-factor analysis"

<p><em>Trichoderma reesei</em> endoglucanases (EGs) have limited industrial applications due to its low thermostability and activity. Here, we aimed to improve the thermostability of EGs from<em> T.reesei</em> without reducing its activity counteracting the activity-stability trade-off. A cross-strategy combination of machine learning and B-factor analysis was used to predict beneficial amino acid substitution in EG loop optimization. Experimental validation showed single-site mutated EG concomitantly improved enzymatic activity and thermal properties by 17.21%&ndash;18.06% and 49.85%&ndash;62.90%, respectively, compared with wild-type EGs. Furthermore, the mechanism explained mutant variants had lower RMSD values and a more stable overall structure than the wild type. According to this study, EGs loop optimization is crucial for balancing the activity-stability trade-off, which may provide new insights into how loop region function interacts with enzymatic characteristics. Moreover, the cross-strategy between machine learning and B-factor analysis improved superior enzyme activity-stability performance, which integrated structure-dependent and sequence-dependent information.</p>

Trade-off between dispersal traits in a heterocarpic plant across its invasion route

<p>Dispersal ability can vary across plant species or populations, such as among core compared to leading populations of invasive plants. However, in heterocarpic plants, which produce propagules with varying dispersal abilities, dispersal potential can also increase via investment in the proportion of dispersing morphs (termed dispersal rate). Nevertheless, very little is known about the interplay between investment in dispersing structures vs. dispersal rate or how each is affected by varying environmental pressures.</p> <p><strong><em>Methods</em></strong></p> <p>This study examined the interplay between investment in dispersing structures and dispersal rate across the invasion route of the heterocarpic plant <em>Heterotheca subaxillaris</em>. <em>H. subaxillaris</em>'s capitula were collected from eight populations along its invasion route in the Eastern Mediterranean coastal plain. The dispersal ability of dispersing pappus-bearing achenes was measured as the ratio between their pappus width and biomass. The dispersal rate was calculated as the ratio between the number of dispersing achenes and total achenes per capitulum. </p> <p><em><strong>Key Results </strong></em></p> <p>The dispersal ability of the dispersing achenes and dispersal rate were found to be negatively correlated across <em>H. subaxillaris</em>' populations, with a greater investment in pappus length in populations at the leading edge of the invasion compared to a greater number of dispersing achenes in core populations.</p> <p><em><strong>Conclusions</strong></em> </p> <p>Our results suggest a trade-off might exist between dispersal ability and dispersal rate, which could change along the invasion route of heterocarpic plants such as <em>H. subaxillaris</em> and contribute to their invasive success. This study highlights the importance of examining both dispersal traits when studying the dispersal potential of heterocarpic species. </p>

Data from: Trade-offs between baseline thermal tolerance and thermal tolerance plasticity are much less common than it appears

<p>Thermal tolerance plasticity is a core mechanism by which organisms can mitigate the effects of climate change. As a result, there is a need to understand how variation in tolerance plasticity arises. The Baseline Tolerance/Plasticity Trade-off Hypothesis (hereafter referred to as the Trade-off Hypothesis, TOH) has recently emerged as a potentially powerful explanation. The TOH posits that organisms with high baseline thermal tolerance have reduced thermal tolerance plasticity relative to those with low baseline tolerance. Many studies have found support for the TOH. However, this support must be regarded cautiously because the most common means of testing the TOH can yield spurious 'trade-offs' due to regression to the mean. I acquired data for 25 previously-published analyses that supported the TOH at the intraspecific level and reanalyzed them after applying a method that adjusts plasticity estimates for regression to the mean. Only six of the 25 analyses remained statistically significant after adjustment, and effect size and variance explained decreased in all cases. The few data sets in which support for the TOH was maintained after adjustment point to areas of future study, but are too few to make generalizations at this point. In sum, regression to the mean has led to a substantial overestimation of support for the TOH and must be accounted for in future tests of the hypothesis. </p>

Biodiversity and pollination benefits trade off against profit in an intensive farming system

<p>Agricultural expansion and intensification have boosted global food production but have come at the cost of environmental degradation and biodiversity loss. Biodiversity-friendly farming that boosts ecosystem services, such as pollination and natural pest control, is widely being advocated to maintain and improve agricultural productivity while safeguarding biodiversity. A vast body of evidence showing the agronomic benefits of enhanced ecosystem service delivery represents important incentives to adopt practices enhancing biodiversity. However, the costs of biodiversity-friendly management are rarely taken into account and may represent a major barrier impeding uptake by farmers. Whether and how biodiversity conservation, ecosystem service delivery, and farm profit can go hand in hand is unknown. Here we quantify the ecological, agronomic and net economic benefits of biodiversity-friendly farming in an intensive grassland-sunflower system in southwest France. We found that reducing land-use intensity on agricultural grasslands drastically enhances flower availability and wild bee diversity, including rare species. Biodiversity-friendly management of grasslands furthermore resulted in an up to 17% higher revenue on neighboring sunflower fields through positive effects on pollination service delivery. However, the opportunity costs of reduced grassland forage yields consistently exceeded the economic benefits of enhanced sunflower pollination. Our results highlight that profitability is often a key constraint hampering adoption of biodiversity-based farming and uptake critically depends on society's willingness to pay for associated delivery of public goods such as biodiversity.</p>

Ismay Trading Post, Utah

Ismay Trading Post is on the map at the confluence of McElmo and Yellow Jacket canyons. It is the last stop on the road to the Navajo reservation from the east side in Colorado. For many decades it offered one-stop shopping for people living beyond the end of the road. Customers could pick up mail, general store goods, seasonal fresh produce and flat-tire repairs all in one place. Eleanor Heffernen Ismay, Robert's mother, and his father, John Ismay, built the adobe trading post in 1921 with the help of Eleanor's father, Jim Heffernen, who also built the Oljeto Trading Post, which is still in operation today. In a 1976 oral-history recording of Eleanor Ismay, archived in the Heritage West database of historical resources, she says, "McElmo is a good place for all kinds of fruit.
