Association of chronic inflammation and accelerated atherosclerosis among an indigenous black population with chronic kidney disease
<p>Introduction: Inflammation plays a major role in the development of atherosclerosis and cardiovascular morbidity and mortality in chronic kidney disease (CKD) patients. Toll-like receptor-4 (TLR4) is a major receptor for lipopolysaccharides (endotoxin) and other ligands involved in the pathogenesis of inflammation. We determined whether endotoxin levels and the presence of TLR4 polymorphisms are associated with markers of inflammation and atherosclerosis among South African CKD patients.</p> <p><br> Materials and methods: Endotoxin, lipopolysaccharide binding protein (LBP), serum CD14 (sCD14), interleukin-8 (IL-8), monocyte chemoattractant protein-1 (MCP-1) and carotid intima media thickness (CIMT) were measured in 160 participants (120 CKD patients and 40 controls). Associations between endotoxins and CIMT in the presence of sCD14, IL-8 and MCP-1, were assessed using odds ratios. Participants were screened for the presence of Asp299Gly and Thr399Ile TLR4 polymorphisms, and CIMT and inflammatory markers were compared between subjects with and without TLR4 polymorphisms.</p> <p>Results: Endotoxin levels correlated with sCD14 (r=0.441, p<0.001) and MCP-1 (r=0.388, p<0.001) levels while increased CIMT was associated with MCP-1 (r=0.448, p<0.001), sCD14 levels (r=0.476, p<0.001), LBP (r=0.340, p<0.001), and IL-8 (r=0.395, p<0.001). Atherosclerosis was associated with endotoxin levels (odds ratio: 4.95; 95% confidence interval: 2.52-9.73; p<0.001), and was predicted by higher serum levels of inflammatory markers. Analysis of patients with TLR4 polymorphisms showed reduced serum levels of inflammatory markers and CIMT values compared with the patients carrying the wild type TLR4 alleles.</p> <p>Conclusion: The study demonstrated associations between circulating endotoxaemia, systemic inflammation and accelerated atherosclerosis among South African CKD patients, and showed that the atherogenic predictive power of endotoxaemia was significantly increased by the presence of elevated levels of inflammatory markers. Additional findings, which must be confirmed, suggest that TLR4 polymorphisms are associated with low levels of inflammatory markers and CIMT values.</p>

Data from: Accelerated epigenetic age and cognitive decline among urban-dwelling adults
Objectives. Epigenetic modifications are closely linked with aging, but their relationship with cognition remains equivocal. Given known sex differences in epigenetic aging, we explored sex-specific associations of three DNA methylation-based (DNAm) measures of epigenetic age acceleration (EAA) with baseline and longitudinal change in cognitive performance among middle-aged urban adults. Methods. We used exploratory data from a sub-group of participants in the Healthy Aging in Neighborhoods of Diversity across the Life Span (HANDLS) study with complete DNA samples and whose baseline ages were >50.0y (2004-2009) to estimate three DNAm EAA measures: (A) universal epigenetic age acceleration (AgeAccel); (B) intrinsic epigenetic age acceleration (IEAA) ; and (C) extrinsic epigenetic age acceleration (EEAA). Cognitive performance was measured at baseline visit (2004-2009) and first follow-up (2009-2013) with 11 test scores covering global mental status and specific domains such as learning/memory, attention, visuo-spatial, psychomotor speed, language/verbal and executive function executive. A series of mixed-effects regression models were conducted adjusting for covariates and multiple testing (N=147-156, ~51% men, k=1.7-1.9 observations/participant, mean follow-up time~4.7y). Results. EEAA, a measure of both biological age and immunosenescence, was consistently associated with greater cognitive decline among men on tests of visual memory/visuo-constructive ability (Benton Visual Retention Test: γ11=0.0512±0.0176, p=0.004) and attention/processing speed (Trail making test, part A: γ11=0.219±0.080, p=0.007). AgeAccel and IEAA were not associated with cognitive change in this sample. Conclusions. EEAA capturing immune system cell aging was associated with faster decline among men in domains of attention and visual memory. Larger longitudinal studies are needed to replicate our findings.
Replication data for High-impact details of play and movements with higher trunk acceleration in female basketball game
<p>This study aimed to identify the high-impact details of play and movements with higher acceleration and their frequency during a female basketball match. Trunk acceleration was measured during a simulated basketball game with eight female players. We categorized the various actions as details of play and movements generated at >6 and 8G resultant accelerations using a video recording and an accelerometer attached to the players’ trunk. The frequency and ratio of the details of play and movements about all detected movements were calculated. A total of 1062 and 223 play actions were detected for the resultant acceleration thresholds of >6 and 8G, respectively. For these acceleration thresholds, in terms of details of play, positioning on the half-court was the most frequently observed (29.6% and 23.8%, respectively). In the terms of movements, deceleration was the most frequently detected movement (21.5% and 23.3%, respectively), followed by landing (7.6 and 15.7%, respectively). The results also showed that characteristics of movements or playing style and position of play might have an effect on acceleration patterns during a basketball game. Monitoring the frequency and intensity of these movements and details of play can help to quantify the biomechanical load experienced by basketball players.</p>
