Data from: Corticosteroid-binding globulin deficiency independently predicts mortality in septic shock

<p>Context Hydrocortisone administration in septic shock remains controversial. Corticosteroid-binding globulin (CBG) transports cortisol to inflammatory sites and is depleted in septic shock. Objective To determine if severely deficient serum CBG &lt;200 nmol/L (Ref range 269-641 nmol/L) independently predicts septic shock mortality. Methods A prospective observational study in patients with septic shock. Patients were categorised into two groups: mean plasma CBG concentrations &lt;200 nmol/L and ≥200 nmol/L (day 1/2), with additional categorisation by nadir CBG. Primary outcome was ICU mortality. Secondary outcomes were 28- and 90-day mortality, norepinephrine requirements, renal-replacement therapy, and clinician-instituted hydrocortisone. Results 135 patients were included. Mortality rates in ICU were higher in the CBG &lt;200 nmol/L vs the CBG ≥200 nmol/L group: 32.4% vs 13.9%; Odds ratio (OR), 2.97, (95% confidence intervals (CI) 1.19,7.41); P=0.02: 28-days; OR 2.25 (95% CI 0.99,5.11): 90-days; OR 2.21 (95% CI 0.99,4.91). Multivariate analysis revealed 4 factors independently associated with ICU mortality: CBG &lt;200 nmol/L (OR 3.23, 95% CI 1.06,9.88), Acute Physiology and Chronic Health Evaluation II &gt;25 (OR 3.58, 95% CI 1.20,10.68); SOFA liver score (OR 1.98, 95% CI 1.04,3.72); and renal-replacement therapy (OR 6.59, 95% CI 2.17,20.01). Nadir CBG levels were associated with higher SOFA cardiovascular scores, norepinephrine total dose (ug) P&lt;0.01 and duration (days) P&lt;0.01. Plasma cortisol concentrations and hydrocortisone administration did not relate to ICU mortality. Conclusion Septic shock patients with CBG &lt;200 nmol/L had higher norepinephrine requirements and 3.2-fold higher ICU mortality. CBG concentration was the only directly reversible independent mortality risk factor.</p>

Data from: Shear-sensitive adhesion enables size-independent adhesive performance in stick insects

<p>The ability to climb with adhesive pads conveys significant advantages, and is hence widespread in the animal kingdom. The physics of adhesion predict that attachment is more challenging for large animals, whereas detachment is harder for small animals, due to the difference in surface-to-volume ratios. Here, we use stick insects to show that this problem is solved at both ends of the scale by linking adhesion to the applied shear force. Adhesive forces of individual insect pads, measured with perpendicular pull-offs, increased approximately in proportion to a linear pad dimension across instars. In sharp contrast, whole-body force measurements suggested area-scaling of adhesion. This discrepancy is explained by the presence of shear forces during whole-body measurements, as confirmed in experiments with pads sheared prior to detachment. When we applied shear forces proportional to either pad area or body weight, pad adhesion also scaled approximately with area or mass, respectively, providing a mechanism that can compensate for the size-related loss of adhesive performance predicted by isometry. We demonstrate that the adhesion-enhancing effect of shear forces is linked to pad sliding, which increased the maximum adhesive force per area sustainable by the pads. As shear forces in natural conditions are expected to scale with mass, sliding is more frequent and extensive in large animals, thus ensuring that large animals can attach safely, while small animals can still detach their pads effortlessly. Our results therefore help to explain how nature's climbers maintain a dynamic attachment performance across seven orders of magnitude in body weight.</p>

Data for: Modeling climate-driven range shifts in populations of two bird species limited by habitat independent of climate

<p>Ranges of species around the world are expected to contract in response to climate change. Species distribution models (SDMs) are a powerful tool for predicting changes in habitat availability, but the variables selected to create SDMs influence their performance. In addition to climate, habitat characteristics and species traits can play a role in predicted species distribution. In this paper, we consider how variable selection influences the accuracy of SDMs when applied to isolated subpopulations of two widely distributed bird species: the great gray owl (<em>Strix</em> <em>nebulosa</em>) and the willow flycatcher (<em>Empidonax</em> <em>traillii</em>). In the Sierra Nevada of California, these species are restricted largely to discrete patches of meadow habitat within a forest matrix, providing the potential to identify specific locations to target conservation efforts. We contrast predictions made by SDMs that consider climatic variables alone with those that incorporate both climate and geophysical variables. Adding geophysical variables resulted in differing model predictions. For willow flycatchers, adding geophysical variables improved predictive performance. In the case of great gray owls, models with and without geophysical variables had nearly identical performance under historical conditions but differed starkly in their predictions. The full model (climatic and geophysical variables) predicted habitat availability to decrease moderately, whereas the climate-only model predicted nearly complete loss of favorable habitat by 2099. The climate-only model is consistent with expectations based on previous SDMs of birds across North America, but previous studies also assume homogeneity in species traits and range-wide habitat requirements. The full model appears more consistent with recent trends in great gray owl numbers in the Sierra Nevada specifically, where the population has remained relatively stable over recent decades. Given contradictions in our model predictions, care should be taken when trying to apply similar SDM models to other systems.</p>

Data from: An efficient independence sampler for updating branches in Bayesian Markov chain Monte Carlo sampling of phylogenetic trees

Sampling tree space is the most challenging aspect of Bayesian phylogenetic inference. The sheer number of alternative topologies is problematic by itself. In addition, the complex dependency between branch lengths and topology increases the difficulty of moving efficiently among topologies. Current tree proposals are fast but sample new trees using primitive transformations or re-mappings of old branch lengths. This reduces acceptance rates and presumably slows down convergence and mixing. Here, we explore branch proposals that do not rely on old branch lengths but instead are based on approximations of the conditional posterior. Using a diverse set of empirical data sets, we show that most conditional branch posteriors can be accurately approximated via a Γ distribution. We empirically determine the relationship between the logarithmic conditional posterior density, its derivatives, and the characteristics of the branch posterior. We use these relationships to derive an independence sampler for proposing branches with an acceptance ratio of ∼90% on most data sets. This proposal samples branches between 2× and 3× more efficiently than traditional proposals with respect to the effective sample size per unit of runtime. We also compare the performance of standard topology proposals with hybrid proposals that use the new independence sampler to update those branches that are most affected by the topological change. Our results show that hybrid proposals can sometimes noticeably decrease the number of generations necessary for topological convergence. Inconsistent performance gains indicate that branch updates are not the limiting factor in improving topological convergence for the currently employed set of proposals. However, our independence sampler might be essential for the construction of novel tree proposals that apply more radical topology changes.
