<p>Pre-processed data.&nbsp;</p> <p>Data were re-referenced off-line to the average of left and right mastoid electrodes, bandpass filtered from 5 to 100 Hz (4th order Butterworth filter) and then segmented to include 200 ms before and 2000 ms after stimulus onset. Epoched data were normalized based on a prestimulus period of 200 ms, and then evaluated according to a sample-by-sample procedure to remove noisy sensors that were replaced using spherical splines. Raw and processed data for publication "Quantitative microscopy reveals dynamics and fate of clustered IRE1alpha"

<p>This dataset contains all raw and processed data for our PNAS paper titled &quot;Quantitative microscopy reveals dynamics and fate of clustered IRE1alpha&quot;. Using the associated analysis code (10.5281/zenodo.3544482), all figures in the paper can be reproduced from the data in this upload.</p> <p>Abstract of the paper is included below:</p> <p>&quot;The endoplasmic reticulum (ER) membrane-resident stress sensor IRE1 governs the most evolutionarily conserved branch of the unfolded protein response. Upon sensing an accumulation of unfolded proteins in the ER lumen, IRE1 activates its cytoplasmic kinase and ribonuclease (RNase) domains to transduce the signal. IRE1 activity correlates with its assembly into large clusters, yet the biophysical characteristics of IRE1 clusters remain poorly characterized. We combined super-resolution microscopy, single-particle tracking, fluorescence recovery and photoconversion to examine IRE1 clustering quantitatively in living human and mouse cells. Our results revealed that (1) by contrast to qualitative impressions gleaned from microscopic images, IRE1 clusters comprise only a small fraction (~5%) of the total IRE1 in the cell. (2) IRE1 clusters have complex topologies that display features of higher-order organization. (3) IRE1 clusters contain a diffusionally constrained core, indicating that they are not phase-separated liquid condensates. &nbsp;(4) IRE1 molecules in clusters remain diffusionally accessible to the free pool of IRE1 molecules in the general ER network. (5) When IRE1 clusters disappear at later timepoints of ER stress as IRE1 signaling attenuates, their constituent molecules are released back into the ER network and not degraded. (6) IRE1 cluster assembly and disassembly are mechanistically distinct. (7) IRE1 clusters&rsquo; mobility is nearly independent of cluster size. Taken together, these insights define the clusters as dynamic assemblies with unique properties. The analysis tools developed for this study will be widely applicable to investigations of clustering behaviors in other signaling proteins.&quot;</p>

<p>The datasets contains the ouput simulation data of the Exo-FMS GCM and of the post-processing with gCMCRT for Effects of the internal temperature on vertical mixing and on cloud structures in Ultra Hot Jupiters</p> <p>Context. The vertical mixing in hot Jupiter atmospheres plays a critical role in the formation and spacial distribution of cloud particles<br>in their atmospheres. This affects the observed spectra of a planet through cloud opacity, which can be influenced by the degree of<br>cold trapping of refractory species in the deep atmosphere.<br>Aims. We aim to isolate the effects of the internal temperature on the mixing efficiency in the atmospheres of Ultra Hot Jupiters (UHJ)<br>and the spacial distribution of cloud particles across the globe.<br>Methods. We couple a simplified tracer based cloud model, picket fence radiative-transfer scheme and mixing length theory to the<br>Exo-FMS general circulation model. We run the model for five different internal temperatures at typical UHJ atmosphere system<br>parameters.<br>Results. Our results show the convective eddy diffusion coefficient remains low throughout the vast majority of the atmosphere, with<br>mixing dominated by advective flows. However, some regions can show convective mixing in the upper atmosphere for colder interior<br>temperatures. The vertical extent of the clouds is reduced as the internal temperature is increased. Additionally, a global cloud layer<br>gets formed below the radiative-convective boundary (RCB) in the cooler cases.<br>Conclusions. Convection is generally strongly inhibited in UHJ atmospheres above the RCB due to their strong irradiation. Convective<br>mixing plays a minor role in keeping cloud particles aloft in ultra hot Jupiters with warm interiors. Our results suggest isolated upper<br>atmosphere regions above cold interiors may exhibit strong convective mixing, allowing aerosols to be better retained in these areas.</p>
