Data for: The role of ecdysis in repair of an attachment system: a case study using geckos
<p>Skin provides functions such as protection and prevention of water loss. In some taxa, the outer surface of skin has been modified to form structures that enable attachment to various surfaces. Constant interaction with surfaces is likely to cause damage to these attachment systems and reduce function. It seems logical that when skin is shed via ecdysis, its effectiveness will increase, through repair of damage or other rejuvenating mechanisms. We address two questions using three diplodactylid geckos as model species. (1) Does repeated mechanical damage affect clinging ability in geckos to the point that they cannot support their own body weight? (2) Does use without induced damage reduce effectiveness of the attachment system, and if so, does ecdysis restore clinging ability? We found that repeated damage reduced clinging ability in all three species, although at different rates. Additionally, use reduced clinging ability over time when no apparent damage was incurred. Clinging ability increased after ecdysis in all three species, both when damage was specially induced, and when it was not. After normal use without induced damage, the increase in clinging ability after ecdysis was statistically significant in two of three species. Our findings show that use decreases clinging ability, and mechanical damage also affects geckos' capacity to exert shear forces consistently. Thus, ecdysis improves clinging ability both in scenarios where damage is induced and more generally. In addition to the physiological functions provided by skin, our study highlights an important function of ecdysis in a speciose vertebrate group.</p>
Data from: Triggered Golgi membrane enrichment promotes PtdIns(4,5)P2 generation for plasma membrane repair
<p><span>The maintenance of plasma membrane integrity and a capacity for efficiently repairing damaged membranes are essential for cell survival. Large-scale wounding depletes various membrane components at the wound sites, including phosphatidylinositols, yet little is known about how phosphatidylinositols are generated after depletion. Here, working with our <em>in</em> <em>vivo C. elegans</em> epidermal cell wounding model, we discovered phosphatidylinositol 4-phosphate (PtdIns4<em>P</em>) accumulation and local phosphatidylinositol 4,5-bisphosphate [PtdIns(4,5)<em>P<sub>2</sub></em>] generation at the wound site. We found that PtdIns(4,5)<em>P<sub>2</sub></em> generation depends on the delivery of PtdIns4<em>P</em>, PI4K, and PI4P 5-kinase PPK-1. In addition, we show that wounding triggers enrichment of the Golgi membrane to the wound site, and that is required for membrane repair. Moreover, genetic and pharmacological inhibitor experiments support that the Golgi membrane provides the PtdIns4<em>P</em> for PtdIns(4,5)<em>P<sub>2</sub></em> generation at the wounds. Our findings demonstrate how the Golgi apparatus facilitates membrane repair in response to wounding and offers a valuable perspective on cellular survival mechanisms upon mechanical stress in a physiological context. </span></p>
Low-Cost Sustainable Engineered Geopolymer Composites (EGCs) for Repair and New Construction of Transportation Infrastructure
<p>This study evaluated the use of cost-effective alternative materials in the formulation of novel low-cost Engineered Geopolymer Composite (EGC) materials for repair and new construction of transportation infrastructure in Region 6. Previous studies and reports have shown that EGCs have excellent mechanical properties, which makes them an eco-friendly and more sustainable alternative to Engineered Cementitious Composites (ECCs). However, the mass adoption of these emerging composites is expected to be hindered by their cost, which is mainly driven by the use of PVA reinforcing fibers, silica fume (SF), and manufactured microsilica sand (MS). To address this key shortcoming, novel low-cost EGC materials for repair and new construction of transportation infrastructure in Region 6 were developed by: (1) metakaolin (MK) and fly ash (FA) as replacements of commonly used SF for GP binders; (2) low-cost PP fiber, PVA fiber, and hybrid systems of PP and PVA fiber; and (3) natural sand instead of commonly used manufactured MS. The MK-FA GP binders were tested for compressive strength and characterized with scanning electron microscopy (SEM) and energy-dispersive X-ray spectroscopy (EDS), while the MK-FA GP composites were tested for workability, compressive strength, and uniaxial tensile strength (UTT). Based on the experimental findings, it was concluded that 20-40 wt% of FA replacement was the optimal range for Na-based MK-FA GP with the strength values of ~12-14 MPa, while 50 wt% of FA replacement had the highest strength for K-based MK-FA GP of 17.2 MPa at 90-day. For composites, the mortar with the highest compressive strength reached a value of 10.6 MPa with 45% of river sand, and with 4 wt% SF and 1.5% PVA+0.25% PP fibers, it reached 21 MPa as a fiber-reinforced composite at 28-day. The composite with 10 wt% SF and 1.75% PE had the highest UTT of 3.97 MPa and a strain capacity of 1.78%. All the slant shear test specimens meet the 28-day shear bond strength requirement according to ASTM C882.</p>

The weaken-fill-repair model for cell budding: Linking cell wall biosynthesis with mechanics
<p>The interplay between cellular mechanics and other biochemical processes leading to the cell cycle is poorly understood. Here, we propose a quantitative model of the mechanism of cell budding, particularly for Saccharomyces cerevisiae, as a ''weaken-fill-repair'' process. This model links Newtonian mechanics over the cell wall with biochemical processes that change the mechanical properties of the wall. Our model shows that: (1) The oscillatory behavior in the size of the mother compartment during cell division, as observed experimentally, is an inevitable consequence of the budding mechanism; (2) The asymmetry of cell division is necessary for the daughter cell to maintain mechanical stiffness; (3) Although different aspects of a cell are strictly constrained and intricately interconnected, the budding process is governed by a single reduced parameter that, when at an appropriate value, can automatically achieve homeostasis. Our quantitative model of the evolution of cell walls and their impact on cell division could provide valuable insights into cell morphology from a system perspective. This knowledge could be instrumental in developing effective antimicrobial treatments against microorganisms with cell walls and even in building synthetic cells.</p>
