Brood parasites that care: alternative nesting tactics in a subsocial wasp

<div> <p>Hosts and brood parasites are a classic example of conflict. Parasites typically provide no offspring care after laying eggs, imposing costs on hosts. Female subsocial wasps, <em>Ammophila pubescens</em>, alternated between initiating their own nests and an 'intruder' tactic of replacing eggs in nests of unrelated conspecifics. Hosts could respond by substituting new eggs of their own, with up to eight reciprocal replacements. Remarkably, intruders usually provisioned offspring in host nests, often alongside hosts. We used field data to investigate why intruders provision and to understand the basis of interactions. We found that intruders could not increase their fitness payoffs by using the typical brood parasite tactic of not provisioning offspring. Intruders using the typical tactic would benefit when hosts provisioned in their stead, but their offspring would starve when hosts failed to provision. Although some hosts obtained positive payoffs when intruders mistakenly provisioned their offspring, on average utilizing a conspecific nest represents parasitism: hosts pay costs while intruders benefit. Both females used the same tactic of egg replacement, but intruders more often laid the final egg. Selection should favour better discrimination of offspring, which could lead to repeated cycles of costly egg replacement.</p> </div>

Alternative Covid-19 mitigation measures in school classrooms: Analysis using an agent-based model of SARS-CoV-2 transmission

<p>The SARS-CoV-2 epidemic continues to have major impacts on children's education, with schools required to implement infection control measures that have led to long periods of absence and classroom closures. We have developed an agent-based epidemiological model of SARS-CoV-2 transmission that allows us to quantify projected infection patterns within primary school classrooms, and related uncertainties; the basis of our approach is a contact model constructed using random networks, informed by structured expert judgment. The effectiveness of mitigation strategies is considered in terms of effectiveness at suppressing infection outbreaks and limiting pupil absence. Covid-19 infections in schools in the UK in Autumn 2020 are re-examined and the model used for forecasting infection levels in autumn 2021, as the more infectious Delta-variant was emerging and school transmission was thought likely to play a major role in an incipient new wave of the epidemic. Our results are in good agreement with available data and indicate that testing-based surveillance of infections in the classroom population with isolation of positive cases is a more effective mitigation measure than bubble quarantine both for reducing transmission in primary schools and for avoiding pupil absence, even accounting for the insensitivity of self-administered tests. Bubble quarantine entails large numbers of pupils being absent from school, with only a modest impact on classroom infection levels. However, maintaining a reduced contact rate within the classroom can have a major beneficial impact on managing Covid-19 in school settings.</p>

Alternative splicing and genetic variation of MHC-E: Implications for rhesus cytomegalovirus-based vaccines

<p>We used long-read sequencing to interrogate rhesus macaque (RM) MHC-E (Mamu-E) alternative splicing and genetic variations. Full-length Mamu-E RNA isoforms were recovered using the PacBio Iso-Seq method.&nbsp;Incomplete&nbsp;5&#39; ends of Mamu-E&nbsp;isoforms were confirmed using Sanger sequencing, where we identified three additional isoforms. Full-length&nbsp;human MHC-E (HLA-E) isoforms were also recovered using the PacBio Iso-Seq method. Isoform sequences and annotations are provided for both Mamu-E and HLA-E in addition to Mamu-E Sanger sequencing data. HLA-E annotations are reported using the hg38 reference, while Mamu-E annotations are shown using rhesus MHC Class I and II assemblies&nbsp;previously generated using Bacterial Artificial Cloning (BAC) technology (https://www.ncbi.nlm.nih.gov/nuccore/AC148696.1).</p> <p>Using PacBio Long Amplicon Analysis, we sequenced complete Mamu-E coding regions of 59 RMs and additionally captured 3&#39; UTR polymorphism using mRNA-seq haplotype phasing analysis. The complete genotyping data for these animals are provided as well as animal metadata. Genotyping data is shown using the Mamu-E canonical isoform (Mamu-E1 from Iso-Seq analysis) as reference.</p>
