Label-free three-photon imaging of intact human cerebral organoids: Tracking early events in brain development and deficits in Rett Syndrome

<p>Human cerebral organoids are unique in their development of progenitor-rich zones akin to ventricular zones from which neuronal progenitors differentiate and migrate radially. Analyses of cerebral organoids thus far have been performed in sectioned tissue or in superficial layers due to their high scattering properties. Here, we demonstrate label-free three-photon imaging of whole, uncleared intact organoids (~2 mm depth) to assess early events of early human brain development. Optimizing a custom-made three-photon microscope to image intact cerebral organoids generated from Rett Syndrome patients, we show defects in the ventricular zone volumetric structure of mutant organoids compared to isogenic control organoids. Long-term imaging of live organoids reveals that shorter migration distances and slower migration speeds of mutant radially migrating neurons are associated with more tortuous trajectories. Our label-free imaging system constitutes a particularly useful platform for tracking normal and abnormal development in individual organoids, as well as for screening therapeutic molecules via intact organoid imaging.</p>

DeepCellMap: a Deep Learning Approach Coupled to Spatial Statistics to unravel Microglial Spatial Organization in the Developing Human Brain

<p>DeepCellMap is a deep-learning-assisted tool that integrates multi-scale image processing with advanced spatial and clustering statistics. This pipeline is designed to map microglial organization during normal and pathological brain development but can be adapted to any cell type.} Using DeepCellMap, we can capture the morphological diversity of microglia,&nbsp; identify strong coupling between proliferative and phagocytic phenotypes, and show that distinct spatial clusters rarely overlap as human brain development progresses. Additionally, we uncover a novel association between microglia and blood vessels in fetal brains exposed to maternal SARS-CoV-2. These findings offer insights into whether various microglial phenotypes form networks in the developing brain to occupy space, and in conditions involving haemorrhages, whether microglia respond to, or influence changes in blood vessel integrity. DeepCellMap is available as open-source software and is a powerful tool for extracting spatial statistics and analyzing cellular organization in large tissue sections, accommodating various imaging modalities. </p>

Understanding brain waste: Unequal opportunities for skills development between highly skilled women and men, migrants and nonmigrants

<p>Brain waste studies seldom analyse the skills development of tertiary-educated</p> <p>workers according to gender and country of origin. Combining statistical analyses,</p> <p>participatory workshops and 77 qualitative interviews on employment trajectories</p> <p>with highly skilled workers in Switzerland, I offer three contributions. An</p> <p>intersectional perspective reveals unequal opportunities of skills development:</p> <p>Swiss-born men have the highest rates of employment, income and jobs commensurate</p> <p>with their education, followed by foreign-born men, then Swiss-born women</p> <p>and lastly foreign-born women. Second, a multiscalar perspective shows how actions</p> <p>by individuals, families, employers and the state favour or constrain skills advancement</p> <p>among women and men. Individual perceptions of gender roles and unequal</p> <p>childcare responsibilities within couples constrain women&#39;s skills development. Further</p> <p>constraints are set by the state and employers&#39; policies and practices regarding</p> <p>migration, childcare, taxation and hiring. Third, a spatial&ndash;temporal perspective unveils</p> <p>that gender inequalities arise at particular moments (migration and childbirth) and</p> <p>places (lacking childcare services and limited employment).</p>
