<p>Identifying differential responses between sexes following traumatic brain injury (TBI) can elucidate the mechanisms behind disease pathology. Peripheral and central inflammation in the pathophysiology of TBI can increase sleep in male rodents, but this remains untested in females. We hypothesized that diffuse TBI would increase inflammation and sleep in males more so than in females. Diffuse TBI was induced in C57BL/6J mice and serial blood samples were collected (baseline, 1, 5, 7 days post‐injury [DPI]) to quantify peripheral immune cell populations and sleep regulatory cytokines. Brains and spleens were harvested at 7DPI to quantify central and peripheral immune cells, respectively. Mixed‐effects regression models were used for data analysis. Female TBI mice had 77%–124% higher IL‐6 levels than male TBI mice at 1 and 5DPI, whereas IL‐1β and TNF‐α levels were similar between sexes at all timepoints. Despite baseline sex differences in blood‐measured Ly6Chigh monocytes (females had 40% more than males), TBI reduced monocytes by 67% in TBI mice at 1DPI. Male TBI mice had 31%–33% more blood‐measured and 31% more spleen‐measured Ly6G+ neutrophils than female TBI mice at 1 and 5DPI, and 7DPI, respectively. Compared with sham, TBI increased sleep in both sexes during the first light and dark cycles. Male TBI mice slept 11%–17% more than female TBI mice, depending on the cycle. Thus, sex and TBI interactions may alter the peripheral inflammation profile and sleep patterns, which might explain discrepancies in disease progression based on sex.</p>
Data from: Rapid cold hardening protects against sublethal freezing injury in an Antarctic insect
Rapid cold hardening (RCH) is a type of beneficial phenotypic plasticity that occurs on extremely short time scales (minutes to hours) to enhance insects' ability to cope with cold snaps and diurnal temperature fluctuations. RCH has a well-established role in extending lower lethal limits, but its ability to prevent sublethal cold injury has received less attention. The Antarctic midge, Belgica antarctica is Antarctica's only endemic insect and has a well-studied RCH response that extends freeze tolerance in laboratory conditions. However, the discriminating temperatures used in previous studies of RCH are far below those ever experienced in the field. Here, we tested the hypothesis that RCH protects against nonlethal freezing injury. Larvae of B. antarctica were exposed to either control (2&[deg]C), direct freezing (-9&[deg]C for 24 h), or RCH (-5&[deg]C for 2 h followed by -9&[deg]C for 24 h). All larvae survived both freezing treatments, but RCH larvae recovered more quickly from freezing stress and had significantly higher metabolic rates during recovery. RCH larvae also sustained less damage to fat body and midgut tissue and had lower expression of two heat shock protein transcripts (hsp60 and hsp90), which is consistent with RCH protecting against protein denaturation. The protection afforded by RCH resulted in energy savings; directly frozen larvae experienced a significant depletion in glycogen energy stores that was not observed in RCH larvae. Together, these results provide strong evidence that RCH protects against a variety of sublethal freezing injuries and allows insects to rapidly fine-tune their performance in thermally variable environments.
Data from: Enzyme polymorphism, oxygen and injury: a lipidomic analysis of flight-induced oxidative damage in a SDH-polymorphic insect
When active tissues receive insufficient oxygen to meet metabolic demand, succinate accumulates and has two fundamental effects: it causes ischemia-reperfusion injury while also activating the hypoxia-inducible factor pathway (HIF). The Glanville fritillary butterfly (Melitaea cinxia) possesses a balanced polymorphism in Sdhd, shown previously to affect HIF pathway activation and tracheal morphology and used here to experimentally test the hypothesis that variation in succinate dehydrogenase affects oxidative injury. We stimulated butterflies to fly continuously in a respirometer (3 min duration), which typically caused episodes of exhaustion and recovery, suggesting a potential for cellular injury from hypoxia and reoxygenation in flight muscles. Indeed, flight muscle from butterflies flown on consecutive days had lipidomic profiles similar to rested paraquat-injected butterflies, but distinct from rested untreated butterflies. Many butterflies showed a decline in flight metabolic rate (FMR) on Day 2, and there was a strong inverse relationship between the ratio of Day 2 to Day 1 FMR and the abundance of sodiated adducts of phosphatidylcholines and coenzyme Q (CoQ). This result is consistent with elevation of sodiated lipids caused by disrupted intracellular ion homeostasis in mammalian tissues after hypoxia-reperfusion. Butterflies carrying the Sdhd M allele had higher abundance of lipid markers of cellular damage, but the association was reversed in field-collected butterflies, where focal individuals typically flew for seconds at a time rather than continuously. These results indicate that Glanville fritillary flight muscles can be injured by episodes of high exertion, but injury severity appears to be determined by an interaction between SDH genotype and behavior (prolonged vs. intermittent flight).
Clinical characteristics of patients hospitalized for ocular chemical injuries in Shanghai from 2012 to 2017
Objective: To summarize the clinical characteristics of patients with ocular chemical injuries and evaluate their potential relationship with the visual outcome by analyzing the medical records of these patients from January 1, 2012, to December 31, 2017. Design: A retrospective case series study. Setting: Jinshan Hospital of Fudan University in Shanghai. Methods: Patient data included age, gender, occupational classification, location of ocular chemical injury, initial and final best-corrected distance visual acuity (BCDVA), intraocular pressure (IOP), nature and chemical phase, distribution and severity of chemical injury, management methods, and complications. All variables were evaluated for their potential relationship with visual outcome. Results: A total of 160 patients were hospitalized with ocular chemical injuries. The majority of the patients were factory workers and arrived at the consultation room less than 24 hours after injury. The most common ocular injury setting, classification of severity, causative chemical, chemical phase, and complications were workplace, grade Ⅱ, unknown and mixed substance, liquid, and elevated IOP, respectively. Initial BCDVA was significantly worse in the solid group than in the liquid group (P = 0.009). The risk factors for poor final BCDVA were identified as older age, poor initial BCDVA, and irrigation 24 hours after injury (P < 0.001, P < 0.001, and P = 0.011, respectively). Conclusion: We elaborate the clinical characteristics and outcomes of patients with ocular chemical injuries in Jinshan District, Shanghai. A comprehensive education program should be established and the use of protective eyewear should be promoted to prevent occupation-related ocular chemical injuries.
Replication data for Anterior Cruciate Ligament Injury: identifying information sources and risk factor awareness among the general population
<p>Creating awareness about a disorder is important for prevention from the perspective of public health. However, for sports injuries, like the anterior cruciate ligament (ACL) injury, there is no study which has investigated the awareness of risk factors for the injury and prevention methods among the general population, to the best of our knowledge. The sources of information among the population are also unclear. The purpose of present study was to identify these aspects of public awareness about the ACL injury.</p> <p>A questionnaire was randomly distributed among the general population registered with a web based questionnaire supplier, to recruit 900 participants who were aware about the ACL injury. The questionnaire consisted of two parts: Question 1 asked them about their sources of information regarding the ACL injury; Question 2 asked them about the risk factors for ACL injury. Multivariate logistic regression was used to determine the information sources that provide a good understanding of the risk factors.</p> <p>The leading source of information for ACL injury was television (57.0%). However, the results of logistic regression analysis revealed that television was not an effective medium to create awareness about the risk factors, among the general population. Instead “Lecture by a coach”, “Classroom session on Health”, and “Newspaper” were significantly effective in creating a good awareness of the risk factors (p < 0.001).</p>

The Swedish History Museum holds a large collection of bones of various kinds, including human remains. We living humans are both appalled by and attracted to visible evidence of death. The scanned remains presented on Sketchfab from our museum are important museum artefacts, chosen because they represent a specific historical event. They also represent one of humanity's worst acts: violence towards other humans. It is therefore our wish that you, as a viewer, use the 3D-models with respect. The image shows a human tibia with a substantial cut caused by an edged weapon. The tibia is one of several thousand artifacts found in a mass grave outside Visby on the island Gotland. All remains and objects found on the site stem from a battle between Gotlandic peasants and Danish mercenaries on 27 July 1361. Many of the skeletons display severe injuries to their feet and legs. Dexmedetomidine preconditioning reduces ischemia-reperfusion injury in equine model of large colon volvulus
<p><strong>Background:</strong> Large colon volvulus is a common cause of colic in horses with high morbidity and mortality when not promptly resolved surgically. More treatment options are needed to improve the outcome of these cases by ameliorating the damage caused by ischemia and reperfusion injury to the large colon.</p> <p><strong>Objective:</strong> To determine the effect of preconditioning with dexmedetomidine prior to induction of ischemia-reperfusion (IR) injury in a large colon volvulus model in the horse.</p> <p><strong>Study Design:</strong> Randomized prospective experimental blinded study.</p> <p><strong>Methods: </strong>Horses received either a dexmedetomidine (DEX) or saline (CON) constant rate infusion (CRI) immediately following induction of anesthesia. Venous, arterial, and transmural occlusion of a section of the large colon was then performed for three hours, after which the ligatures and clamps were removed to allow for reperfusion for further three hours. Full thickness biopsies of the large colon were taken at baseline, 1 and 3 hours of ischemia, and at 1 and 3 hours of reperfusion for histological analysis.</p> <p><strong>Results:</strong> The severity of crypt epithelial loss (DEX= 2.1 (0.8-2.8), CON= 3.1(2.5-4), p = 0.03) and mucosal hemorrhage was decreased (DEX= 2.1 (1.3-3), CON= 3.5(2.5-4), p = 0.03) in group DEX compared to group CON. Crypt length remained longer (DEX= 369.5±91.7 µm, CON= 238.5±72.6 µm, p = 0.02) and interstitium to crypt (I:C) ratio remained lower (DEX= 1.4 (1-1.7), CON= 2.6 (1.8-5.9), p = 0.03) in group DEX compared to group CON during reperfusion.</p> <p><strong>Main Limitations:</strong> Clinical applicability of pharmacologic preconditioning is limited.</p>
