Data from: Sensitivity analysis of conservation targets in systematic conservation planning
Systematic conservation planning has rapidly advanced in the past decade and has been increasingly incorporated in multiple studies and conservation projects. One of its requirements is a quantitative definition of conservation targets. While the Convention on Biological Diversity aims to expand the world's protected area network to 17% of the land surface, in many cases such uniform policy-driven targets may not be appropriate for achieving persistence of various species. Targets are often set arbitrarily, often because information required for the persistence of each species is unavailable or unknown in the focal region. Conservation planners therefore need to establish complementary novel approaches to address the gaps in setting targets. Here, we develop and present a novel method that aims to help guide the selection of conservation targets, providing support for decision makers, planners, and managers. This is achieved by examining the overall flexibility of the conservation network resulting from conservation prioritization, and aiming for greater flexibility. To test this approach we applied the decision support tool Marxan to determine marine conservation priority areas in the eastern Mediterranean Sea as a case study. We assessed the flexibility of the conservation network by comparing 80 different scenarios in which conservation targets were gradually increased and assessed by a range of calculated metrics (e.g., the percentage of the total area selected, the overall connectivity). We discovered that when conservation targets were set too low (i.e., below 10% of the distribution range of each species), very few areas were identified as irreplaceable and the conservation network was not well defined. Interestingly, when conservation targets were set too high (over 50% of the species' range), too many conservation priority areas were selected as irreplaceable, an outcome which is realistically infeasible to implement. As a general guideline, we found that flexibility in a conservation network is adequate when ~10−20% of the study area is considered irreplaceable (selection frequency values over 90%). This approach offers a useful sensitivity analysis when applying target-based systematic conservation planning tools, ensuring that the resulting protected area conservation network offers more choices for managers and decision makers.
Data from: Genome-wide sensitivity analysis of the microsymbiont Sinorhizobium meliloti to symbiotically important, defensin-like host peptides
The model legume species Medicago truncatula expresses more than 700 nodule-specific cysteine-rich (NCR) signaling peptides that mediate the differentiation of Sinorhizobium meliloti bacteria into nitrogen-fixing bacteroids. NCR peptides are essential for a successful symbiosis in legume plants of the inverted-repeat-lacking clade (IRLC) and show similarity to mammalian defensins. In addition to signaling functions, many NCR peptides exhibit antimicrobial activity in vitro and in vivo. Bacterial resistance to these antimicrobial activities is likely to be important for symbiosis. However, the mechanisms used by S. meliloti to resist antimicrobial activity of plant peptides are poorly understood. To address this, we applied a global genetic approach using transposon mutagenesis followed by high-throughput sequencing (Tn-seq) to identify S. meliloti genes and pathways that increase or decrease bacterial competitiveness during exposure to the well-studied cationic NCR247 peptide and also to the unrelated model antimicrobial peptide polymyxin B. We identified 78 genes and several diverse pathways whose interruption alters S. meliloti resistance to NCR247. These genes encode the following: (i) cell envelope polysaccharide biosynthesis and modification proteins, (ii) inner and outer membrane proteins, (iii) peptidoglycan (PG) effector proteins, and (iv) non-membrane-associated factors such as transcriptional regulators and ribosome-associated factors. We describe a previously uncharacterized yet highly conserved peptidase, which protects S. meliloti from NCR247 and increases competitiveness during symbiosis. Additionally, we highlight a considerable number of uncharacterized genes that provide the basis for future studies to investigate the molecular basis of symbiotic development as well as chronic pathogenic interactions.

Implementing Riverine Biogeochemical Inputs in ECCO-Darwin: A Sensitivity Analysis of Terrestrial Fluxes in a Data-Assimilative Global Ocean Biogeochemistry Model
<p>Resolving riverine biogeochemical inputs in ocean biogeochemistry models is pivotal for capturing the spatiotemporal variability of nutrients and carbon in coastal regions and in the global ocean. ECCO-Darwin is a pioneering data-assimilative global-ocean biogeochemistry model, which, to date, has focused on the pelagic zone. In this work, we use an optimized version of ECCO-Darwin to perform a sensitivity analysis to quantify the response of the open ocean and coastal margins to lateral inputs of carbon and nutrients. We generate riverine inputs by combining point-source freshwater discharge from JRA55-do with the Global NEWS 2 watershed model, accounting for lateral inputs from 5171 watersheds worldwide. While adding carbon and nutrients along with freshwater improves biogeochemical skill in river plume regions and coastal waters, the open-ocean response may be overestimated due to an excess of carbon and nutrients advected offshore. This highlights the need for a more nuanced representation of land-to-ocean and nearshore processes for quantifying how global ocean primary production and carbon cycling respond to land-to-ocean inputs.</p>

<p>Each of the folders contains SESMG results for a sensitivity analysis of the study "Potential-Risk and No-Regret Options for Urban Energy System Design - A Sensitivity Analysis". More information can be found in this publication. Each folder contains two subfolders. The "cost-minimum" subfolder contains the results for financially optimized systems, and the "emission-minimum" subfolder contains the results for GHG emission-optimized systems. Within these subfolders, the results for different gradations of the respective sensitivity parameters are stored in separate sub-subfolders. The 01_reference_total_ghg_emissions folder has a slightly different structure. Since the results are not separated into financially and emissions-optimized scenarios, the results of different gradations are stored directly in the main folder of this sensitivity analysis.</p>
